Functional and Biochemical Characterization of Alvinella pompejana Cys-Loop Receptor Homologues

Cys-loop receptors are membrane spanning ligand-gated ion channels involved in fast excitatory and inhibitory neurotransmission. Three-dimensional structures of these ion channels, determined by X-ray crystallography or electron microscopy, have revealed valuable information regarding the molecular mechanisms underlying ligand recognition, channel gating and ion conductance. To extend and validate the current insights, we here present promising candidates for further structural studies. We report the biochemical and functional characterization of Cys-loop receptor homologues identified in the proteome of Alvinella pompejana, an extremophilic, polychaete annelid found in hydrothermal vents at the bottom of the Pacific Ocean. Seven homologues were selected, named Alpo1-7. Five of them, Alpo2-6, were unidentified prior to this study. Two-electrode voltage clamp experiments revealed that wild type Alpo5 and Alpo6, both sharing remarkably high sequence identity with human glycine receptor α subunits, are anion-selective channels that can be activated by glycine, GABA and taurine. Furthermore, upon expression in insect cells fluorescence size-exclusion chromatography experiments indicated that four homologues, Alpo1, Alpo4, Alpo6 and Alpo7, can be extracted out of the membrane by a wide variety of detergents while maintaining their oligomeric state. Finally, large-scale purification efforts of Alpo1, Alpo4 and Alpo6 resulted in milligram amounts of biochemically stable and monodisperse protein. Overall, our results establish the evolutionary conservation of glycine receptors in annelids and pave the way for future structural studies.     

Quantitative label-free screening for antibodies using scattering biophotonic microarray imaging

A biophotonic array based on gold nanoparticles functionalized with antigen proteins has been used to determine the concentrations of the respective antibodies in solution. Four proteins—fibrinogen, bovine serum albumin, transferrin, and C-reactive protein—were used to construct a test array with the assay repeated a number of times. The antibody-antigen association and dissociation rate constants were determined for the antibody assays from a series of calibration experiments. The label-free determination of the unknown antibody concentrations was performed using two related kinetic analyses. From these results, the current array assay sensitivity is 250 ng ml^-1 with an accuracy of 15% using an 8-min kinetic measurement and a 16-spot averaged assay.     

Changes in the parasitic fish fauna of the Ka?rak-Kum reservoir over a long period

During last few years a tendency for the reduction in the number of parasites of all investigated fishes has been observed. This is connected with the general tendency for the impoverishment of the reservoir flora and fauna, that may be the result of the fluctuations of its water level. Finally this process leads to the reduction in the number of many species of parasites and even to their disappearance.     

A Formal Representation of the WHO and UNICEF Estimates of National Immunization Coverage: A Computational Logic Approach

Production of official statistics frequently requires expert judgement to evaluate and reconcile data of unknown and varying quality from multiple and potentially conflicting sources. Moreover, exceptional events may be difficult to incorporate in modelled estimates. Computational logic provides a methodology and tools for incorporating analyst''s judgement, integrating multiple data sources and modelling methods, ensuring transparency and replicability, and making documentation computationally accessible. Representations using computational logic can be implemented in a variety of computer-based languages for automated production. Computational logic complements standard mathematical and statistical techniques and extends the flexibility of mathematical and statistical modelling. A basic overview of computational logic is presented and its application to official statistics is illustrated with the WHO & UNICEF estimates of national immunization coverage.     

Label-free antibody-antigen binding detection by optical sensor array based on surface-synthesized gold nanoparticles

Gold nanoparticles grown in situ from printed seed particles on a glass substrate have been fabricated into a biosensor array. The light-scattering properties of the resulting surfaces show sensitivity to changes in the local refractive index. Each array spot is functionalized with fibrinogen or bovine serum albumin and scattered radiation is used to monitor the refractive index change on label-free binding of the antibodies to their antigens from whole blood antiserum. Data were collected real-time and the association rate constants for the specific antibody-antigen binding were derived from a kinetic analysis. The minimum antibody concentration detection sensitivity is of 100 nM.     

Detection and characteristics of foci of arbovirus infection in the Kazakh region

Complex studies carried out on the territory of Kazakhstan have allowed one to detect arboviruses, heretofore unknown in the republic, to determine their role in human infectious pathology and to obtain additional data on the ecology of viruses causing tick-borne encephalitis and Crimean hemorrhagic fever under the conditions of different geographical zones in the republic. On the basis of the results thus obtained the map indicating the spread of arbovirus infections and their causative agents on the territory of the Kazakh SSR has been drawn. This map may serve as a practical aid in organizing antiepidemic measures.     

Temperature Gradient Measurements by Using Thermoelectric Effect in CNTs-Silicone Adhesive Composite

This work presents the fabrication and investigation of thermoelectric cells based on composite of carbon nanotubes (CNT) and silicone adhesive. The composite contains CNT and silicon adhesive 1∶1 by weight. The current-voltage characteristics and dependences of voltage, current and Seebeck coefficient on the temperature gradient of cell were studied. It was observed that with increase in temperature gradient the open circuit voltage, short circuit current and the Seebeck coefficient of the cells increase. Approximately 7 times increase in temperature gradient increases the open circuit voltage and short circuit current up to 40 and 5 times, respectively. The simulation of experimental results is also carried out; the simulated results are well matched with experimental results.     

Changes in nitric oxide in heart of intact and sympathectomized rats of different age

Nitric oxide production in heart tissues of rats of different age in the norm and after pharmacological sympathectomy was studied by electron spin resonance spin-trapping. Rats at the age of 14, 21, 70, and 100 days were used in the experiment. The concentration of nitric oxide produced in rat heart tissues proved to considerably decrease during ontogeny. Pharmacological sympathectomy notably decreased nitric oxide production in the heart in 14-and 21-day-old rats: the nitric oxide concentration in the spin trap as well as the level of R and T conformers of hemoglobin nitrosyl complexes decreased. In 70-day-old rats, pharmacological sympathectomy had no notable effect on the level of nitric oxide-containing paramagnetic complexes.
