Fungal populations of the haulm base of wheat

Abstract In a 2-year field survey, the phylloplane mycoflora on the haulm base of wheat was studied by the dilution plate technique. No significant colonisation was registered before the end of May. Cladosporium spp., 'white' and 'red' yeasts were found to be the most abundant fungi.     

Modulation of murine hemopoiesis in vivo by a synthetic hemoregulatory pentapeptide (HP 5b)

Abstract. Degeneration of the archenteron in middle gastrulae occurred in the presence of α,α'-dipyridyl or Zn²⁺, inhibitors of prolyl hydroxylase. In the presence of these substances the archenteron degenerated and was eventually destroyed. Adding Fe²⁺ to the embryo culture containing α,α'-dipyridyl protected the archenteron from further degeneration, but the collapsed archenteron was not restored to the upright position. At the late gastrula stage, α,α'-dipyridyl did not cause the degeneration of the archenteron. Treatment of the embryos by α,α'-dipyridyl, starting at the swimming blastula stage, resulted in the production of many mesenchyme-like cells but archenteron was not produced in the embryos. Addition of Fe²⁺ to α,α'-dipyridyl culture, just before the beginning of gastrulation of normal embryos, resulted in the formation of normal archenteron. α,α'-Dipyridyl inhibited hydroxylation of proline residues of collagen in sea urchin embryos and Fe²⁺ prevented the inhibition by α,α'-dipyridyl. Respiration was not inhibited by α,α'-dipyridyl.     

Quantifying Biomass Changes of Single CD8+ T Cells during Antigen Specific Cytotoxicity

Existing approaches that quantify cytotoxic T cell responses rely on bulk or surrogate measurements which impede the direct identification of single activated T cells of interest. Single cell microscopy or flow cytometry methodologies typically rely on fluorescent labeling, which limits applicability to primary cells such as human derived T lymphocytes. Here, we introduce a quantitative method to track single T lymphocyte mediated cytotoxic events within a mixed population of cells using live cell interferometry (LCI), a label-free microscopy technique that maintains cell viability. LCI quantifies the mass distribution within individual cells by measuring the phase shift caused by the interaction of light with intracellular biomass. Using LCI, we imaged cytotoxic T cells killing cognate target cells. In addition to a characteristic target cell mass decrease of 20–60% over 1–4 h following attack by a T cell, there was a significant 4-fold increase in T cell mass accumulation rate at the start of the cytotoxic event and a 2–3 fold increase in T cell mass relative to the mass of unresponsive T cells. Direct, label-free measurement of CD8+ T and target cell mass changes provides a kinetic, quantitative assessment of T cell activation and a relatively rapid approach to identify specific, activated patient-derived T cells for applications in cancer immunotherapy.     

A simple,rapid, and sensitive method for measuring protein concentration in subcellular membrane fractions prepared by sucrose density ultracentrifugation

The Multiskan spectrophotometric system and Coomassie brilliant blue G-250 protein-dye binding method have been used together to measure NaOH-solubilized protein in subcellular membrane fractions prepared from isolated rat adipose cells. Forty-eight samples can be read in duplicate within 1 min. Sucrose in concentrations up to 0.7 m interfere only moderately with the assay. A rapid and convenient method is, therefore, now available for multiple protein determinations following sucellular fractionation on sucrose gradients.     

The oxidation-reduction potential of P-700 in chloroplast lamellae and subchloroplast particles

The oxidation-reduction potential of P-700 has been determined in chloroplast lamellae and in subchloroplast particles by measuring the magnitude of flash-induced absorption changes at 820 or at 703 nm (due to the oxidation of P-700) in the presence of known concentrations of potassium ferro- and ferricyanide. A midpoint potential of about +490 mV was determined in chloroplast lamellae and in particles prepared with digitonin (D-144) or Triton (TSF-1). A lower potential was determined with Photosystem I particles obtained after harsher treatments with Triton or a mixture of detergents. The potential is even lower in chlorophyll-protein complex I particles prepared with sodium dodecyl sulfate (about +430 mV). Very similar values were determined from oxidized minus reduced spectra measured with a double-beam spectrophotometer. Titrations were also made with D-144 and TSF-I particles, with 66% glyeerol in the buffer, at 21 °C and at 77 °K. P-700 was found to be half-oxidized at ferricyanide/ferrocyanide ratios of about 60 at 21 °C and of about 1 at 77 °K. This shows that the redox equilibrium is largely perturbed by the cooling process.     

Assembly of heterodimeric luciferase after de novo synthesis of subunits in rabbit reticulocyte lysate involves hsc70 and hsp40 at a post-translational stage.

Heterodimeric luciferase from Vibrio harveyi had been established as a unique model enzyme for direct measurements of the effects of molecular chaperones and folding catalysts on protein folding and subunit assembly after de novo synthesis of subunits in rabbit reticulocyte lysate. It was observed that luciferase assembly can be separated in time from synthesis of the two subunits and that under these post-translational conditions assembly was inhibited by either ATP depletion or inhibition of peptidylprolyl cis/trans isomerases, that is, by addition of cyclosporin A or FK506. Furthermore, it was observed that the inhibitory effect of FK506 on luciferase assembly can be suppressed by addition of purified cyclophilin, thereby providing the first direct evidence for the involvement of peptidylprolyl cis/trans isomerases in protein biogenesis in the eukaryotic cytosol. Here the ATP requirement in luciferase assembly has been characterized. Depletion of either Hsp90 or CCT from reticulocyte lysate did not interfere with luciferase assembly. However, addition of purified Hsc70 stimulated luciferase assembly. While addition of purified Hsp40 did not have any effect on luciferase assembly, the stimulatory effect of Hsc70 was further increased by Hsp40. Thus, after synthesis of the two subunits in reticulocyte lysate assembly of heterodimeric luciferase involves Hsc70 and its co-chaperone Hsp40. Therefore, Hsc70 aids protein biogenesis in the eukaryotic cytosol not only at the levels of nascent polypeptide chains and precursor proteins that have to be kept competent for transport into cell organelles, but also at the level of subunits that have to be kept competent for assembly.     

Mouse erythrocyte carriers osmotically loaded with methotrexate

The mouse red blood cell (RBC) and red blood cell ghost (RBCG) have been studied as carriers of methotrexate (MTX). When incubated with high concentrations of MTX, RBCs take up significant quantities of it. However, when active loading techniques, such as the slow dialysis and preswell methods, are applied to those cells, up to 15 times more MTX can be entrapped. We have studied factors critical to the incorporation, leakage, and morphology of RBCGs during their loading with MTX by the slow dialysis and preswell methods. Compounds added to the buffers to maintain the ATP content of the cells and osmolarity play functional roles in this process. The fate of the material entrapped within the ghosts after in vivo administration was shown to be capture by the reticuloendothelial system. The pharmacological efficacy of MTX-loaded RBCGs in treating mice bearing hepatoma ascites tumors was demonstrated by increases in average survival time of 28.5-42.8%.
