Controlling accumulations of unwanted biofilms requires an understanding of the mechanisms that organisms use to interact with submerged substrata. While the substratum properties influencing biofilm formation are well studied, those that may lead to cellular or biofilm detachment are not. Surface-grafted stimuli-responsive polymers, such as poly (N-isopropylacrylamide) (PNIPAAm) release attached cells upon induction of environmentally-triggered phase changes. Altering the physicochemical characteristics of such polymeric systems for systematically studying release, however, can alter the phase transition. The physico-chemical changes of thin films of PNIPAAm grafted from initiator-modified self-assembled monolayers (SAMs) of ω-substituted alkanethiolates on gold can be altered by changing the composition of the underlying SAM, without affecting the overlying polymer. This work demonstrates that the ability to tune such changes in substratum physico-chemistry allows systematic study of attachment and release of bacteria over a large range of water contact angles. Such surfaces show great promise for studying a variety of interactions at the biointerface. Understanding of the source of this tunability will require further studies into the heterogeneity of such films and further investigation of interactions beyond those of water wettability.  相似文献

Adult Rhipicephalus appendiculatus Muguga, having high or low intensities of Theileria parva Muguga infection in their salivary glands, were exposed to 20 °C and 85% relative humidity in the laboratory or quasi-natural conditions. Survival of the ticks and T. parva infections in their salivary glands was then monitored over a two year period. Ticks, having an average infection level of 2 infected acini per female, survived for up to 70 or 106 weeks after moulting under the laboratory or quasi-natural conditions respectively. Those having an infection level of 26 infected acini per female, survived for a similar duration except that those under quasi-natural conditions survived for a slightly shorter duration (102 weeks). Similarly, T. parva parasites survived for much longer periods under quasi-natural conditions than under the laboratory conditions. They survived for up to 38 or 78 weeks post salivary gland infection under the laboratory or quasi-natural conditions respectively in both categories of infection levels. There was apparently a density dependent relationship in T. parva survival, with a dramatic fall in infection occurring in ticks with high levels of infection between weeks 10 and 18 or weeks 38 and 46 post salivary gland infection in those exposed to laboratory or quasi-natural conditions before levelling off. We have studied the temporal variation in viral abundances and community assemblage in the eutrophic Lake Loosdrecht through epifluorescence microscopy and pulsed field gel electrophoresis (PFGE). The virioplankton community was a dynamic component of the aquatic community, with abundances ranging between 5.5 x 10(7) and 1.3 x 10(8) virus-like particles ml(-1) and viral genome sizes ranging between 30 and 200 kb. Both viral abundances and community composition followed a distinct seasonal cycle, with high viral abundances observed during spring and summer. Due to the selective and parasitic nature of viral infection, it was expected that viral and host community dynamics would covary both in abundances and community composition. The temporal dynamics of the bacterial and cyanobacterial communities, as potential viral hosts, were studied in addition to a range of environmental parameters to relate these to viral community dynamics. Cyanobacterial and bacterial communities were studied applying epifluorescence microscopy, flow cytometry, and denaturing gradient gel electrophoresis (DGGE). Both bacterial and cyanobacterial communities followed a clear seasonal cycle. Contrary to expectations, viral abundances were neither correlated to abundances of the most dominant plankton groups in Lake Loosdrecht, the bacteria and the filamentous cyanobacteria, nor could we detect a correlation between the assemblage of viral and bacterial or cyanobacterial communities during the overall period. Only during short periods of strong fluctuations in microbial communities could we detect viral community assemblages to covary with cyanobacterial and bacterial communities. Methods with a higher specificity and resolution are probably needed to detect the more subtle virus-host interactions. Viral abundances did however relate to cyanobacterial community assemblage and showed a significant positive correlation to Chl-a as well as prochlorophytes, suggesting that a significant proportion of the viruses in Lake Loosdrecht may be phytoplankton and more specific cyanobacterial viruses. Temporal changes in bacterial abundances were significantly related to viral community assemblage, and vice versa, suggesting an interaction between viral and bacterial communities in Lake Loosdrecht.  相似文献

NPY receptors are identified in calf frontal cortex and hippocampus membrane preparations by binding of N-[propionyl-3H] neuropeptide Y. Saturation and competition binding data with PYY, NPY-(18–36) and NPY itself fit with a single class of sites: for the radioligand KD = 1.4 ± 0.5 nM, Bmax = 434 ± 180 fmol/mg protein in frontal cortex, KD = 0.7 ± 0.2 nM, Bmax = 267 ± 50 fmol/mg protein in hippocampus. Competition curves of the Y1-subtype selective agonist [Leu31, Pro34]NPY are biphasic in both membrane preparations: high affinity sites (i.e. Y1-subtype) amount to 80% in frontal cortex and 23% in hippocampus. The remaining sites are of the Y2-subtype. Out of 23 Conus venom preparations, 17 inhibit the binding of [3H]NPY in both membrane preparations, but only two of them (from Conus aulicus and C. pennaceus) do so with high potency (ic50 < 5 μg protein/ml). Only one venom preparation (from C. mercator) had weak discriminatory properties (ic50Y2/ic50Y1 = 6). Venom from C. anemone increased the [3H]NPY binding 5-fold and with an ic50 of 15–18 μg protein/ml. This binding occurred to the venom itself and was unrelated to the NPY receptors since it was equally potent when displaced by [Leu31, Pro34]NPY, NPY-(18–36), PYY and NPY. [Bryozoans from Miocene reef environments in the South-Rifian corridor of Morocco. In late Miocene coral reefs from the Fes area (Northern Morocco), bryozoans are the best-represented group. An inventory of the bryozoan fauna (59 species) was taken on two reefs with different organization and palaeogeographical situation in a late Miocene seaway between the Atlantic and the Mediterranean. A study of the bryozoans and their zoarial forms allows an improved reconstruction of reefal environments and available habitats. Membraniporiforrn (encrusting) colonies largely predominate, whereas celleporiforms (nodular colonies) are more rare. Rigid erect (vinculariiform, adeoniform, and reteporiform) and rigid articulated zoarial types (cellariiform and catenicelliform) are fairly well represented. At depths of about 10 m and on the reef front encrusting bryozoans were well developed in cryptic habitats on living coral substrates. Erect species lived in cavities provided by the coral framework, and plants offered flexible substrates for the attachment of epiphytal forms. □Bryozoa, coral reefs, Morocco, Messininn, pulueoecology. Dans les récifs coralliens du Miocène supérieur de la région de Fes (Maroc septentrional), les bryozoaires constituent, parrni les organismes conservés, le groupe le mieux représenté. Un recensement de la fame de bryozoaires (59 espèces) a été effectué dans deux récifs d'organisation et de situation paléogéographique différentes, dans un secteur de communication entre Atlantique et Méditerranée au MiocéFne supérieur. L'étude des bryozoaires et de leurs formes zoariales permet de proposer une meilleure reconstitution de I'environnement récifal et des habitats disponibles. Les colonies membraniporiformes (encrobtantes) predominent largement, alors que les cellepori-formes (colonies noduleuses) sont beaucoup plus rares. Les types zoariaux érigés rigides (vincula-riiforme, adéoniforme et rétéporiforme) et érigés articulés (cellariiforme et catenicelliforme) sont assez bien représentés. A des profondeurs denviron 10 m et dans la partie antérieure des constructions récifales, les bryozoaires encroûtants étaient bien développés dans des habitats cryptiques sur substrats coralliens vivants. The relationship between the intramolecular dynamics and the spectra has been analyzed by means of the molecular dynamics technique. Time autocorrelation functions of bond lengths, bending angles and torsional angles have been evaluated in a crystalline trans-polyacetylene system. The Fourier transforms of such functions have been compared with the densities of states obtained both for carbon and hydrogen atoms. This comparison is aimed at investigating the microscopic origin of the peaks which appear in the densities of states. This approach can be used in the analysis of the spectroscopic data of any molecular or polymeric system.  相似文献

1. Invertebrate bioturbation can strongly affect water‐sediment exchanges in aquatic ecosystems. The objective of this study was to quantify the influence of invertebrates on the physical characteristics of an infiltration system clogged with fine sediment. 2. Two taxa (chironomids and tubificids) with different bioturbation activities were studied in experimental slow infiltration columns filled with sand and gravel and clogged with a 2 cm layer of fine sediment at the surface. We measured the effects of each taxon separately and combined on hydraulic head, water mobility and sediment reworking. 3. The results showed that invertebrates could reduce sediment clogging and this effect was linked to the functional mode of bioturbation of each group. Tubificid worms dug networks of galleries in the fine sediment, creating pathways for water flow, which reduced the clogging of sediment. In contrast, the U‐shaped tubes of chironomids were restricted to the superficial layer of fine sediments and did not modify the hydraulic conductivity of experimental columns. The combination of invertebrates did not show any interactive effects between tubificids and chironomids. The occurrence of 80 tubificids in the combination was enough to maintain the same hydraulic conductivity that 160 worms did in monospecific treatment. 4. The invertebrates like tubificid worms can have a great benefit on functioning of clogged interfaces by maintaining high hydraulic conductivity, which contributes to increased water‐sediment exchanges and stimulates biogeochemical and microbial processes occurring in river sediments.  相似文献
