Comparative Study of Microsporidian Spores by Flow Cytometric Analysis   总被引:2,自引:0,他引:2  
ABSTRACT. Spore suspensions of microsporidian parasites of fish (Microsporidium ovoideum, Glugea stephani, Glugea atherinae and Spraguea lophii ) have been analyzed by flow cytometry. Spore nuclei were dyed either by propidium iodide or bis-benzimide (Hoechst 33342). By observation of forward light scatter and fluorescence the four species could be distinguished and the mono- and diplokaryotic populations of S. lophii identified. Staining of DNA by bis-benzimide was better and easier than propidium iodide. Forward light scatter and fluorescence values were characteristic of each species and remained unchanged throughout the year, so flow cytometry can be used for distinction of spores of some microsporidian parasites once their flow cytometric parameters are known. However, special care has to be taken in tool calibration and material preparation for analysis because of the high precision of the technique.  相似文献

The presence of an endemic cyclopoid fauna in anchihaline caves is confirmed in this paper after the discovery of two new genera of cyclopinids in caves of the Canary and Balearic Islands. Oromiina fortunata sp. nov. is described from a flooded lava tube in Lanzarote; it is characterized by the extraordinary length attained by the antennae. Ginesia longi-caudata sp. nov. is described from a flooded karstic cave in Mallorca. The body is ornamented dorsally with conspicuous integumental outgrowths, and along the lateral margins of both the cephalosome and the second pedigerous somite of male, by a linear array of pores. The new taxa are most closely related to forms found in the shallow water hyperbenthos, although one trait of Ginesia suggest an affinity with deep water taxa since, for example, the linear arrays of pores were previously known only in Cyclopicina within the Cyclopinidae. A key to the 37 genera of the family Cyclopinidae is presented.  相似文献

Ferns are thought to have lower photosynthetic rates than angiosperms and they lack fine stomatal regulation. However, no study has directly compared photosynthesis in plants of both groups grown under optimal conditions in a common environment. We present a common garden comparison of seven angiosperms and seven ferns paired by habitat preference, with the aims of (1) confirming that ferns do have lower photosynthesis capacity than angiosperms and quantifying these differences; (2) determining the importance of diffusional versus biochemical limitations; and (3) analysing the potential implication of leaf anatomical traits in setting the photosynthesis capacity in both groups. On average, the photosynthetic rate of ferns was about half that of angiosperms, and they exhibited lower stomatal and mesophyll conductance to CO2 (gm), maximum velocity of carboxylation and electron transport rate. A quantitative limitation analysis revealed that stomatal and mesophyll conductances were co‐responsible for the lower photosynthesis of ferns as compared with angiosperms. However, gm alone was the most constraining factor for photosynthesis in ferns. Consistently, leaf anatomy showed important differences between angiosperms and ferns, especially in cell wall thickness and the surface of chloroplasts exposed to intercellular air spaces.  相似文献

A new species of the thermophylic Tethyan relict prawn Typhlatya is described from two anchialine caves near Perpignan (southern France). The new species is closely related to a congener known only from a freshwater cave at Castellón (eastern Spain), about 400 km to the south-west, differing apparently only in the size and shape of the rostrum and the armature of the dactylus of the fifth pereiopod. Based on palaeogeographical evidence and assuming a sister-group relationship between both species, we suggest that their common ancestor could not be older than early Pliocene in age, and that it was already a stygobiont taxon adapted to live in shallow-water marine crevicular habitats. This ancestor would have vanished from the western Mediterranean after the cooling associated with the onset of northern Hemisphere glaciation, about 3 Mya, as documented for other Mediterranean marine taxa. Indeed, the genus is completely stygobiont and does not occur in fluvial environments. The Pyrenees represent a watershed boundary that eliminates the possibility of the derivation of one species from the other by active dispersal after establishment in continental waters.  © 2005 The Linnean Society of London, Zoological Journal of the Linnean Society , 2005, 144 , 387–414.  相似文献

Montucaris distincta gen. nov. sp. nov. , a new genus and species of hirsutiid, is described from the bathyal floor of the northern South Atlantic, off Brazil. It is easily distinguished from other members of the family by the reduced, one-segmented exopods of pereopods 5 and 6 and the array of stout triangular spines present on the anterior margin of the basis and proximal endopodal segments of the third pereopod. This discovery is notable as it includes the first record of males in the family, demonstrating that hirsutiids are gonochoristic. Developmental stages including mancas, preparatory females and juvenile males were recovered, in addition to numerous brooding females and two morphs of adult males. We infer that the new genus has a life cycle involving non-feeding terminal males with regressed mouthparts. These males display a bizarre cephalic configuration, with a clearly defined transverse dorsal suture on the dorsal cephalothoracic shield just behind the articulation of mandibles, a feature present also in syncarida and in some Thermosbaenacea and that we interpret as secondary. The peracaridan affinities of the Hirsutiidae are firmly established here after settling definitively the oostegitic nature of the setose plates present on the posteromedial margin of the pereopodal coxae of brooding females. The separate ordinal rank for hirsutiids – as the Bochusacea – with respect to the Mictacea, within which they had been previously classified, is favoured here. The genome size of 51 populations of 20 species of the North American endemic sagebrushes (subgenus Tridentatae ), related species, and some hybrid taxa were assessed by flow cytometry, and were analysed in a phylogenetic framework. Results were similar for most Tridentatae species, with the exception of three taxonomically conflictive species:  Artemisia bigelovii Gray,  Artemisia pygmaea Gray, and  Artemisia rigida Gray. Genome size homogeneity (together with the high morphological, chemical, and karyological affinities, as well as low DNA sequence divergence) could support a recent diversification process in this geographically restricted group, thought to be built upon a reticulate evolutionary framework. The Tridentatae and the other North American endemic Artemisia show a significantly higher genome size compared with the other subgenera. Our comparative analyses including genome size results, together with different kinds of ecological and morphological traits, suggest an evolutionary change in lifestyle strategy linked to genome expansion, in which junk or selfish DNA accumulation might be involved. Conversely, weed or invasive behaviour in Artemisia is coupled with lower genome sizes. Data for both homoploid and polyploid hybrids were also assessed. Climate warming and associated sea ice reductions in Antarctica have modified habitat conditions for some species. These include the congeneric Adélie, chinstrap and gentoo penguins, which now demonstrate remarkable population responses to regional warming. However, inconsistencies in the direction of population changes between species at different study sites complicate the understanding of causal processes. Here, we show that at the South Orkney Islands where the three species breed sympatrically, the less ice‐adapted gentoo penguins increased significantly in numbers over the last 26 years, whereas chinstrap and Adélie penguins both declined. These trends occurred in parallel with regional long‐term warming and significant reduction in sea ice extent. Periodical warm events, with teleconnections to the tropical Pacific, caused cycles in sea ice leading to reduced prey biomass, and simultaneous interannual population decreases in the three penguin species. With the loss of sea ice, Adélie penguins were less buffered against the environment, their numbers fluctuated greatly and their population response was strong and linear. Chinstrap penguins, considered to be better adapted to ice‐free conditions, were affected by discrete events of locally increased ice cover, but showed less variable, nonlinear responses to sea ice loss. Gentoo penguins were temporarily affected by negative anomalies in regional sea ice, but persistent sea ice reductions were likely to increase their available niche, which is likely to be substantially segregated from that of their more abundant congeners. Thus, the regional consequences of global climate perturbations on the sea ice phenology affect the marine ecosystem, with repercussions for penguin food supply and competition for resources. Ultimately, variability in penguin populations with warming reflects the local balance between penguin adaptation to ice conditions and trophic‐mediated changes cascading from global climate forcing.  相似文献

The Gorrondatxe section, a prospective Lutetian Global Stratotype Section and Point (GSSP), has recently been used as the master reference section to reassess the correlation between Eocene magnetostratigraphic and calcareous planktonic biostratigraphic scales. However, the exact calibration of some events remained ill defined, as they were thought to be missing in Gorrondatxe due to a fault. The most important missing events were the first occurrence of the planktonic foraminifera Turborotalia frontosa and the C22n/C21r chron boundary. Either might be a reliable correlation criterion for the Ypresian/Lutetian boundary, as both approach the age of the original Lutetian Stratotype. New studies allowed the identification of the former event 9 m above the Gorrondatxe fault, within magnetic polarity Chron C21r and calcareous nannofossil Zone CP12a. Distinctive test features that characterize the most primitive morphotype of T. frontosa are described. Despite the high turbidite content, recurrent pelagic limestone–marl couplets and bundles occur, whose formation was driven by precession and eccentricity astronomical cycles. The first occurrence of T. frontosa was found 27 couplets and 5.5 bundles (60 m) below the first occurrence of the calcareous nannofossil Blackites inflatus , which is dated at 48 Ma. Hence, the age of the first occurrence of T. frontosa is estimated at 48.55 Ma, confirming that it is the most suitable planktonic foraminiferal correlation criterion for the Ypresian/Lutetian boundary. These results show that the stratigraphic interval missing due to the Gorrondatxe fault cannot be greater than a few metres and reinforce the value of this section as a prospective Lutetian GSSP.  相似文献

Thirteen Lower–Middle Eocene (Ypresian–Lutetian) successions, including the Gorrondatxe section in the western Pyrenees, show biomagnetostratigraphic correlation schemes that do not agree with the current standard framework. The main discrepancy concerns the position of the boundary between planktonic foraminiferal Zones P9 (=E7, approximately) and P10 (=E8, approximately), which was thought to occur within calcareous nannofossil Subzone CP12a and at the boundary between magnetic polarity Chrons C22n and C21r. However, in the differing correlation scheme the boundary between Zones P9 (=E7) and P10 (=E8) occurs close to the base of Subzone CP13a and to the boundary between Chrons C21n and C20r. An attempt at a new Ypresian–Lutetian boundary biomagnetochronology is made based on data from the Gorrondatxe section, which shows that the boundary between Zones P9 (=E7) and P10 (=E8) is 3.1 Myr younger than hitherto considered. Therefore, the duration of the Early Eocene, most commonly defined according to this planktonic foraminiferal zonal boundary, has generally been underestimated over the last four decades.  相似文献
