Abstract The chloroplast ultrastructure, especially the thylakoid organization, the polypeptide composition of the thylakoid membranes and photosynthetic O2 evolution rate, chlorophyll (Chl) content and Chi a/b ratio were studied in leaves of nine plants growing in contrasting biotopes in the wild in South Finland. All the measurements were made at the beginning of the period of main growth on leaves approaching full expansion, when the CO2-saturated O2 evolution rate (measured at 20°C and 1500 μmol photons m?2s?1) was at a maximum, ranging from 19.2 to 6.9 μmol O2 cm?2 h?1. Among the species, the Chi a/b ratio varied between 3.75 and 2.71. In the mesophyll chloroplasts, the ratio of the total length of appressed to non-appressed thylakoid membranes varied between 1.07 and 1.79, the number of partitions per granum varied between 2.8 and 12.0 and the grana area between 21 and 42% of the chloroplast area. There was a significant relationship between the rate of O2 evolution of the leaf discs and the thylakoid organization in the mesophyll chloroplasts. The higher the O2 evolution rate, the lower was the ratio of the total length of appressed to non-appressed thylakoid membranes and also the lower the grana area. Although the relationship of the photosynthetic rate with the Chi content and the Chi a/b ratio of the leaves was not as clear, a significant negative correlation existed between the Chi a/b ratio and the ratio of appressed to non-appressed thylakoid membranes, indicating lateral heterogeneity in the distribution of different Chl- protein complexes.  相似文献

After one month of cultivation in the dark in inorganic medium the chloroplasts of protonemata of Ceratodon purpureus have larger grana than chloroplasts from light-grown cultures. Incubation of dark-grown material with ALA increases the chlorophyll content and chlorophyll a/b ratio. On polyacrylamide-gel electrophoresis, a préferential labelling of chlorophyll-protein complex I is obtained after treatment with (3H) ALA in darkness. In contrast, in light, much higher activity is found in chlorophyll-protein complex II. The free pigment zone is highly labelled in both environments.  相似文献

Screening of 55 different cyanobacterial strains revealed that an extract from Nostoc XPORK14A drastically modifies the amplitude and kinetics of chlorophyll a fluorescence induction of Synechocystis PCC 6803 cells. After 2 d exposure to the Nostoc XPORK14A extract, Synechocystis PCC 6803 cells displayed reduced net photosynthetic activity and significantly modified electron transport properties of photosystem II under both light and dark conditions. However, the maximum oxidizable amount of P700 was not strongly affected. The extract also induced strong oxidative stress in Synechocystis PCC 6803 cells in both light and darkness. We identified the secondary metabolite of Nostoc XPORK14A causing these pronounced effects on Synechocystis cells. Mass spectrometry and nuclear magnetic resonance analyses revealed that this compound, designated as M22, has a non‐peptide structure. We propose that M22 possesses a dual‐action mechanism: firstly, by photogeneration of reactive oxygen species in the presence of light, which in turn affects the photosynthetic machinery of Synechocystis PCC 6803; and secondly, by altering the in vivo redox status of cells, possibly through inhibition of protein kinases.  相似文献

The total amount of fatty acids in the mono- (MGDG) and diglycosyl diglyceride (DGDG) and more polar lipid fractions of frozen Ceratodon purpureus shoots was 4.6, 3.4 and 4.0 mg/g dry weight, respectively. The respective values for the tops of frozen Pleurozium schreberi were 2.6, 3.3 and 3.8 mg/g dry weight. The molar ratios MGDG/DGDG and MGDG + DGDG/chlorophyll were 1.3 and 3.7, respectively, for C. purpureus and 0.8 and 3.5 for P. schreberi. In C. purpureus the main fatty acids in the MGDG fraction were C 18:3ω3 (44% of the total fatty acids) and C 16:3ω3 (26%); in the DGDG fraction C 18:3ω3 (70%); and in the more polar lipid fraction C 18: 3ω3 (26%) and C 16:0 (25%). The proportion of C 20 polyunsaturated fatty acids was 15, 12 and 19% of the total fatty acids found in the MGDG, DGDG and more polar lipid fractions, respectively. In P. schreberi the proportion of C 20 polyunsaturated fatty acids was high in all polar lipid fractions (47, 42 and 25% in MGDG, DGDG and more polar lipid fractions, respectively). In addition, MGDG and DGDG fractions contained abundantly C 18:3ω3 (32 and 45%, respectively), and the more polar lipid fraction both C 18: 3ω3 (24%) and C 16:0 (27%).  相似文献

The effect of Mg2+ during the isolation of chlorophyll-protein complexes was studied in two moss species (Pleurozium schreberi and Ceratodon purpureus) and New Zealand spinach (Tetragonia expansa). When 2 mM MgCl2 was included in all the extraction and separation phases, the proportions of chlorophyll-protein complex I. were very small in all plants studied. The withdrawal of Mg2+ considerably increased the proportions of CP I. The most pronounced increase in the chlorophyll present as CP I was found when Mg2+ was withdrawn from the gel, and this also increased the mobility of the CP II complex and free pigment zone. Exclusion of Mg2+ from the running buffer had very little effect. Although Mg2+ had little effect on the relative amount of chlorophyll in CP II, the withdrawal of Mg2+ from all the extraction and separation phases caused formation of polymers of CP II. In the mosses, the formation of polymers of CP II seemed to be more obvious in the species with large grana. Absence of Mg2+ from all the extraction and separation phases sometimes also produced a polymer of CP I.  相似文献

Differential redox regulation of thylakoid phosphoproteins was studied in winter rye plants in vivo. The redox state of chloroplasts was modulated by growing plants under different light/temperature conditions and by transient shifts to different light/temperature regimes. Phosphorylation of PSII reaction centre proteins D1 and D2, the chlorophyll a binding protein CP43, the major chlorophyll a/b binding proteins Lhcb1 and Lhcb2 (LHCII) and the minor light‐harvesting antenna protein CP29 seem to belong to four distinct regulatory groups. Phosphorylation of D1 and D2 was directly dependent on the reduction state of the plastoquinone pool. CP43 protein phosphorylation generally followed the same pattern, but often remained phosphorylated even in darkness. Phosphorylation of CP29 occurred upon strong reduction of the plastoquinone pool, and was further enhanced by low temperatures. In vitro studies further demonstrated that CP29 phosphorylation is independent of the redox state of both the cytochrome b6/f complex and the thiol compounds. Complete phosphorylation of Lhcb1 and 2 proteins, on the contrary, required only modest reduction of the plastoquinone pool, and was subject to inhibition upon increase in the thiol redox state of the stroma. Furthermore, the reversible phosphorylation of Lhcb1 and 2 proteins appeared to be an extremely dynamic process, being rapidly modulated by short‐term fluctuations in chloroplast redox conditions.  相似文献

When the mosses Pleurozium schreberi (Brid.) Mitt, and Ceratodon purpureus (Hedw) Brid. were exposed for 4 week to either continuous or rhythmie light of 75 or 150 μE m?2 s?1, the photosynthetic rate, calculated on the basis of chlorophyll and of dry weight, was lower in the moss plants exposed to continuous light. RUBP-carboxylase activity was lower in these plants, too. On the other hand more starch was accumulated in the chloroplasts in continuous than in rhythmic light. Neither moss species showed any differences in the dark respiration rates. The chlorophyll content was lower in continuous light, especially at the higher intensity; but Chi a/Chl b ratio showed little change compared with that in rhythmic light. The reduction of chlorophyll did not affect the two chlorophyll protein complexes equally; the percentage of chlorophyll associated with CP I decreased, while that associated with CP II showed liltle change. The decrease in CP I chlorophyll appeared to be due to the partial destruction of stroma thylakoids: the size or the grana increased in continuous light. Many of the effects of continuous light in these mosses resembled those observed in higher plants in the early phases of senescence.  相似文献

Chloroplasts perform essential signalling functions in light acclimation and various stress responses in plants. Research on chloroplast signalling has provided fundamental information concerning the diversity of cellular responses to changing environmental conditions. Evidence has also accumulated indicating that different cell types possess specialized roles in regulation of leaf development and stress acclimation when challenged by environmental cues. Leaf veins are flanked by a layer of elongated chloroplast-containing bundle sheath cells, which due to their central position hold the potential to control the flux of information inside the leaves. Indeed, a specific role for bundle sheath cells in plant acclimation to various light regimes is currently emerging. Moreover, perception of light stress initiates systemic signals that spread through the vasculature to confer stress resistance in non-exposed parts of the plant. Such long-distance signalling functions are related to unique characteristics of reactive oxygen species and their detoxification in bundle sheath cells. Novel techniques for analysis of distinct tissue types, together with Arabidopsis thaliana mutants with vasculature-specific phenotypes, have proven instrumental in dissection of structural hierarchy among regulatory processes in leaves. This review emphasizes the current knowledge concerning the role of vascular bundle sheath cells in light-dependent acclimation processes of C3 plants.  相似文献

Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration.
