Interaction of the exr and lon Genes in Escherichia coli 总被引:6,自引:5,他引:1
Strains of Escherichia coli carrying the gene lon typically produced excess capsular polysaccharide, and were sensitive to ultraviolet light (UV) irradiation, thymine starvation, and nalidixic acid, forming long filaments after these treatments. Sensitivity was reduced by a number of posttreatments. In the presence of a second UV sensitivity gene, exr, some of these properties were suppressed: long filaments were not formed, the effect of lon on UV and nalidixic acid sensitivity was greatly reduced, and irradiation posttreatments gave an enhancement of survival characteristic of exr rather than lon strains. Production of capsular polysaccharide was not affected by the exr gene. 相似文献

The ultraviolet sensitivity gene of Escherichia coli B was introduced into a K-12 recipient by transduction with phage P1. The uvs gene of E. coli B is cotransducible with the proC locus of K-12, is closely linked to tsx, is not linked to lacZ, and only rarely to purE. The transductants are mucoid, filamentous on irradiation, and show plating-medium response. The order of markers is lacZ proC tsx uvs purE. 相似文献

Summary Following UV irradiation of AB1157 31 mucoid ultraviolet light UV sensitive mutants were isolated. These were all induced to form filaments by UV irradiation, i.e. they had all the phenotypic properties of Lon mutants. These lon mutants fell into two phenotypic classes based on their sensitivity to UV. The gene determining UV sensitivity and mucoidy in all mutants of both Class A and Class B was cotransducible with proC. Intra-class crosses by Pl transduction yielded no UV resistant recombinants. Inter-class crosses yielded UV resistant nonmucoid recombinants, the frequency depending on the direction of the cross. The data imply two adjacent blocks in the lon region of E. coli and the order of markers in this region is probably proC tsx lon Class A lon purE Class B.This work was carried out under Public Health Service Grant CA 05687-08 from the National Cancer Institute.Recipient of a Public Health Service Career Development Award. 相似文献

Mutants of Escherichia coli K12 were constructed which carry the exrA mutation addition to the various recombination deficient mutations recA recB and recC. The double mutant containing the exrA recA genotype is found to be slightly more sensitive to UV irradiation at very low doses of UV but essentially is very similar to the exrA + recA at the high UV doses. The recombination deficiency, λ induction and DNA degradation of the exrA recA shows a slight increase in the defectiveness of each of these functions. The double mutants of exrA recB and exrA recC show an increase in UV sensitivity and recombination deficiency and λ induction. The DNA degradation following UV-irradiation of these mutants is more characteristic of the recB and recC mutant alone.These results give further support to the theory that exrA and lex are probably mutants within the same cistron and also suggest that exrA, lex and recA are involved in a common DNA repair pathway and that the gene products of all three functions are required to regulate recB+ and recC+ endonuclease induced DNA degradation. 相似文献
