A two-step purification procedure for 5-aminolevulinic acid dehydratase (EC 4.2.1.24) from human red blood cells has been developed. It involves one ion exchange and one gel filtration step. The purification is about 1000-fold, and the yield is more than 85%. With the purified enzyme a direct spectrophotometric assay of product formation without subsequent reaction with Ehrlich's reagent is described.  相似文献

Separate continuous cultures of Pseudomonas putida R5-3, grown on toluene, and Pseudomonas alcaligenes C-O, grown on benzoate, were concentrated and continuously amalgamated on a ceramic bead column, which was subjected to a continuous stream of chlorobenzene vapors. A recombinant strain, P. putida CB1-9, was isolated in less than 1 month. P. alcaligenes C-0 grew on benzoate and 3-chlorobenzoate but not on toluene, P. putida R5-3 grew on benzoate and toluene but not on 3-chlorobenzoate, and neither strain grew on chlorobenzene or 1,4-dichlorobenzene; however, the recombinant P. putida CB1-9 grew on all of these substrates. Chlorobenzene-utilizing strains were not found in continuous cultures run at the lowest growth rate (0.05/h) or in the absence of the donor strain, P. alcaligenes C-0. Chloride was released in stoichiometric amounts when P. putida CB1-9 was grown on either chlorobenzene or 1,4-dichlorobenzene. The recombinant strain was related to P. putida R5-3, phenotypically and genetically. Restriction enzyme digests of the single 57-kilobase (kb) plasmid in R5-3 and of the single 33-kb plasmid in CB1-9 were similar, but also indicated rearrangement of plasmid DNA. Coincidental or causal to the loss of the 24-kb fragment was the observation that the recombinant--unlike its parent, R5-3--did not grow on xylenes or methylbenzoates. Although both ortho-pyrocatechase (OP) and meta-pyrocatechase (MP) were found in CB1-9 and R5-3, MP activity was 20- to 50-fold higher in R5-3 cells grown on 4-methylbenzoate than in the same cells grown on benzene.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Corynebacterium glutamicum is an industrially important bacterium for the manufacture of amino acids. We constructed genomic libraries of this Gram+ bacterium and screened for clones carrying isoleucine biosynthesis genes (ilv) by complementation of Escherichia coli mutants. Clones complementing ilvA, ilvB, and ilvC were isolated. As based on the functional analysis of the corresponding plasmids in C. glutamicum, the DNA fragments isolated encode threonine dehydratase, acetohydroxy acid synthase, and isomeroreductase, catalyzing three subsequent reactions in Ile synthesis. Subcloning and transposon mutagenesis revealed that ilvB and ilvC reside on a 7-kb chromosomal fragment and that these genes are transcribed in the same direction. A shuttle vector was constructed to allow exonuclease treatment and assay subsets of plasmids for gene expression in the original C. glutamicum background. These constructs and their enzyme activity determinations revealed that despite close linkage ilvC is expressed independently from ilvB. Using Southern blots, a 15-kb fragment of chromosomal DNA carrying the ilvBC cluster was characterized. This fragment does not contain ilvA, demonstrating the entirely different organization of the isoleucine biosynthesis genes in C. glutamicum from that in enterobacteria.  相似文献

Summary Phytoplankton production for three size classes (<20 m, 20–100 m, >100 m), total primary production and qualitative composition of phytoplankton populations were recorded from 18 stations in the south-eastern Weddell Sea in February/March 1983. Total primary production ranged between 80 and 1670 mg C m-2 d-1 with an average of 670 mg C m-2 d-1, nearly 70% of which was contributed by the <20 m size fraction (usually pennate and/or centric diatoms). Production of phytoplankton was in the higher range of values reported by other authors for the same region. Variations in primary production could not be attributed to composition of populations, ambient light levels or concentrations of macronutrients (N, P, Si). Phytoplankton populations had a higher diversity in the deeper parts of the Weddell Sea and coincided with different oceanographic situations. Three zones (along the shelf-ice edge from Atka Bay to Halley Bay, west of Halley Bay and off the Filchner/Rønne Ice Shelf) with different communities could be clearly distinguished.  相似文献

Zusammenfassung Die Bezeichnung Klimakammer wird in steigendem Masse für die Unterdruckkammern gebraucht, was einer starken Einengung des Begriffs gleichkommt. Denn unter einer Klimakammer versteht man allgemein einen Raum, in welchem zu bestimmten Zwecken klimatische Bedingungen hergestellt werden, die in einem oder mehreren Klimafaktoren vom natürlichen Lokalklima abweichen. Aus der Fülle der Klimafaktoren wurden wegen der therapeutischen Bedeutung die Variationsmöglichkeiten des Luftkolloids und des Luftdrucks ausführlicher entsprechend dem derzeitigen Stande der Kenntnisse besprochen.Die Ausschaltung des Luftkolloids führt zu den allergenfreien Kammern Storm v. Leeuwens. Die zusätzliche Beimengung medikamentöser Schwebstoffe zur Raumluft ergibt Rauminhalatorien, von denen als Prototypen die Säurekammern, Ultraschallnebelkammern, Solenebelkammern und Dispneumotherapiekammern beschrieben wurden. Durch Erhöhung des Luftdrucks entstehen Ueberdruck- und durch Herabsetzung Unterdruckkammern. Die physiologischen Wirkungen der Luftdruckänderung und die bewährten Möglichkeiten im therapeutischen Einsatz werden auf Grund umfassender praktischer Erfahrungen dargestellt.An extensive English summary of this article will appear in November 1960 in the book published by S.W.  Values for the exchange-coupling constant J and the double-exchange parameter B have been estimated for dimeric and hexameric mixed-valence iron clusters. For sulfur-bridged species the range of J values is 300–450 cm–1, and B values vary between 320 and 400 cm–1. For an OH-bridged diiron cluster B is as large as 1300 cm–1. The calculation and scale-up of fermentation processes need kLa as one of the most important engineering data. There are two methods to determine kLa depending on power input, aeration rate and the properties of the fermentation broth: static with a balance between air supply and exit, dynamic gassing out with following the changes of dissolved oxygen concentration during periods of air off and a following air on. Within early intervals of fermentation time the data from both methods agree well, while for later time intervals the dynamic method always gives much lower values for kLa than static. The only explanations for this phenomenon are quick changes in the oxygen metabolism or an enzymatic storage of oxygen. For both gassing out and saturation period it is possible to calculate the same absolute amounts of this additional oxygen.  相似文献

The expression of gamma-glutamyl transpeptidase (GGT) is a specific property of the brain capillary endothelium that constitutes the blood-brain barrier. We report here the detection of GGT, not only in endothelial cells, but also in pericytes, demonstrating that a brain capillary-specific pericyte population exists. We raised antibodies to GGT using a porcine brain microvessel GGT-protein-A (staphylococcal protein A) fusion protein as antigen which was expressed in Escherichia coli. The immunohistochemical analysis of the subcapillary distribution of GGT in porcine brain cortex and cerebellum sections by both light and electron microscopy revealed the expression of GGT in the capillary-adjacent pericytes in addition to the GGT-positive endothelial layer. We confirmed these data for cultured porcine brain microvascular endothelial cells and pericytes. GGT immunofluorescence could be detected in both cell types in culture. Endothelial cells exhibited a weak staining, whereas pericytes were strongly positive for GGT. Due to the high phagocytotic activity of pericytes and their location on the abluminal surface of the microvessels, we propose a possible protective or detoxifying function of GGT in cerebrovascular pericytes.  相似文献
