Abstract

Populations of the genus Fusarium in wheat fields were studied within the crop-growing season at Qena area (Upper Egypt) using two different types of media (DCPA and DRBA) at 25°C. Fourteen Fusarium species were isolated during this study, namely F. anthophilum, F. aquaeductuum, F. chlamdosporum, F. dimerum, F. merismoides, F. moniliforme, F. oxysporum, F. poae, F. proliferatum, F. sambucinum, F. scripi, F. solani, F. sporotrichioides and F. subglutinans. Fusarium merismoides, F. oxysporum and F. sambucinum were the most common Fusarium species isolated from different wheat plant parts (rhizosphere and rhizoplane) as well as from the wheat fields (soil and air). Fusarium spp. rarely appeared at the beginning of the season and increased sharply between January to March and decreased slightly or sharply at the end of the season according to the type of media and isolation source.  相似文献

Alternaria alternata is the most common fungal pathogen of tomatoes in Upper Egypt. Morphological identification of this fungus is challenging; therefore, this study searched for new classification tools based on molecular techniques. Using a dilution plating method, 67 strains of A. alternata were isolated from 34 samples of rotten tomato fruits representing the Giza 80 and Edkawy cultivars. The collected strains were identified using the amplification products of the internal transcribed spacer (ITS) region, glyceraldehyde 3‐phosphate dehydrogenase (Gpd) and Alt a1, which is a gene involved in the production of most of the allergens produced by A. alternata. The screening revealed that A. alternata constituted more than half of the total fungi recovered from rotten tomatoes in this study. According to the phylogenetic analysis using these three loci, the collected strains clustered in accordance with the host cultivar type from which they had been isolated. Specific gene random primer polymerase chain reaction (SGRP‐PCR) techniques indicated that the A. alternata population in the tested region has a high genetic diversity. The pathogenicity test showed that most of the A. alternata isolates (67.2%) were highly pathogenic, and no correlation was found between the phylogenetic analysis and pathogenicity. In addition, the influence of the fungicide Disan 80% on the collected strains showed significant differences that were attributed to the source of isolation.  相似文献

This paper describes the synthesis of a series of quinolines graphted with hydrazones, pyrazoles, pyridazine, phthalazine, triazepinone, semicarbazide, and thiomorpholide moieties and four metal complexes. These derivatives were screened against Fusarium oxysporum and the red palm weevil (RPW) Rhynchophorus ferrugineus Oliver (coleopteran: Curculionidae) as palm pathogens. Only chlorinated quinolines were active against these organisms with hydrazones being good fungicides, while those modified with pyrazoles and pyrazines showed moderate insecticidal activity. A unique trihydroxylated hydrazone was active against both organisms, while another hydrazone, the most potent fungicide in this series, exhibited insecticidal activity only upon complexation with Zn2+ ions.  相似文献

The biodiversity of Trichoderma was studied in the Northern half of the Nile valley in Egypt. 20 strains were isolated from 9 different geographic locations, representing 19 different habitats, all with a pH between 7.3 and 8.4. Only T. harzianum (three ITS1/2 haplotypes and three RAPD-genotypes) and the anamorph of Hypocrea orientalis were found. One of the T. harzianum haplotypes (4 strains) is new. The occurrence of T. harzianum haplotypes and of H. orientalis appeared to be essentially independent of the habitat (pH, plant, soil type), and also did not correlate with biochemical properties (cellulase and chitinase activity) of the individual strains. These two taxa seem to be indigenous to the Nile valley, their presence not being influenced by the agricultural history of the soils.  相似文献

Twelve isolates of Pythium species (P. aphanidermatum, P. deliense, P. ultimum var. ultimum and P. ultimum var. sporangiiferum) from different hosts were compared from morphological, pathological and molecular viewpoints. Minimum, optimum and maximum temperatures of P. aphanidermatum and P. deliense were similar while those of P. ultimum var. ultimum and P. ultimum var. sporangiiferum were also similar. All tested isolates were highly virulent against cucumber seedlings with 100% damping-off. RAPD data using three different primers revealed that strains of P. ultimum var. ultimum and P. ultimum var. sporangiiferum are distinct from each other. This data can be used to separate those species from P. aphanidermatum and P. deliense. In contrast, RAPD data cannot be used to separate P. aphanidermatum and P. deliense. Sequence analysis of the ribosomal DNA internal transcribed spacers (ITS) was used to establish phylogenetic relationships among the tested isolates.  相似文献

Thirty soil-isolates of Streptomyces were analyzed to determine their antagonism against plant-pathogenic fungi including Fusarium oxysporum, Pythium aristosporum, Colletotrichum gossypii, and Rhizoctonia solani. Seven isolates showed antifungal activity against one or more strain of the tested fungi. Based on the 16S rDNA sequence analysis, these isolates were identified as Streptomyces tendae (YH3), S. griseus (YH8), S. variabilis (YH21), S. endus (YH24), S. violaceusniger (YH27A), S. endus (YH27B), and S. griseus (YH27C). The identity percentages ranged from 98 to 100%. Although some isolates belonged to the same species, there were many differences in their cultural and morphological characteristics. Six isolates out of seven showed chitinase activity according to a chitinolytic activity test and on colloidal chitin agar plates. Based on the conserved regions among the family 19 chitinase genes of Streptomyces sp. two primers were used for detection of the chitinase (chiC) gene in the six isolates. A DNA fragment of 1.4 kb was observed only for the isolates YH8, YH27A, and YH27C. In conclusion, six Streptomyces strains with potential chitinolytic activity were identified from the local environment in Taif City, Saudi Arabia. Of these isolates, three belong to family 19 chitinases. To our knowledge, this is the first reported presence of a chiC gene in S. violaceusniger YH27A.  相似文献

Keratinophilic fungi include true fungi that vigorously degrade keratin as well as a number of important human pathogenic dermatophytes. We identified 41 species and one variety belonging to 19 genera in 50 floor dust samples following culture on Sabouraud’s dextrose agar medium at 28°C. Dermatophytes and closely related fungi were represented by six species—Aphanoascus fulvescens, Aphanoascus sp., Arthroderma cuniculi, Chrysosporium lucknowense, Gymnoascus uncinatus and Trichophyton rubrum. There were 35 species and one variety of other fungal species, with members of Aspergillus and Penicllium being the most prevalent. Twenty-seven species and 1 variety belonging to 14 genera were identified from 24 dust samples gathered at 2-week intervals from male student housing at El-Kenose during January–December 2005 that had been cultured on Sabauraud’s dextrose agar at 28°C. The monthly counts of keratinophilic fungi showed irregular fluctuations, with a peak in April and the lowest point in November. Dermatophytes and closely related fungi were represented by Aphanoascus fulvescens, Aphanoascus sp., Chrysosporium lucknowense, Gymnoascus uncinatus, and Trichophyton rubrum. In conclusion, the study reports the incidences of some dermatophytes found in the floor dust of student houses of South Valley University, Egypt.  相似文献

Fungal endophytes were isolated from the leaves of Calotropis procera (Apocynaceae) collected from Taif region (Saudi Arabia). Thirty-three different taxa were recovered. The overall foliar colonization rate was 35.1%. A total of 161 isolates were obtained and identified into 33 distinct operational taxonomic units based on the sequencing of the internal transcribed spacer regions of the rRNA gene. The most prevalent fungi were Aspergillus flavus, Chaetomium globosum, Cochliobolus lunatus, Fusarium dimerum, F. oxysporum, and Penicillium chrysogenum. A total of 161 isolates were tested for antifungal activities against four plant pathogenic fungi (Alternaria alternata, Fusarium oxysporum, Botrytis cinerea, and Pythium ultimum), of which 33 isolates showed antifungal activity against at least one plant pathogenic fungi. Four isolates of Chaetomium globosum and three isolates of Myrothecium verrucaria showed the strongest antifungal activity. This study reported the occurrence of a much wider spectrum of fungi, when compared with previous work. Also, it confirmed the variation of different isolates from the same species in terms of antifungal activity.  相似文献

Fungal endophytes were isolated from twigs of Juniperus procera (Cupressaceae) collected from Taif region (Saudi Arabia). Twenty-six different taxa were recovered. The overall foliar colonization rate was 36%. A total of 144 isolates were obtained and identified into six distinct operational taxonomic units based on the sequencing of the internal transcribed spacer regions of the rRNA gene. The most prevalent fungi were Aspergillus fumigatus, Penicillium oxalicum, Preussia sp., Peyronellaea eucalyptica, Peyronellaea sancta and Alternaria tenuissima. A total of 144 isolates were tested for antibacterial and antifungal activities against Staphylococcus aureus, Klebsiella pneumoniae, Candida albicans and Fusarium solani in which 52 isolates showed antimicrobial activity against at least one of the tested microbes. Aspergillus fumigatus (7 isolates), Hypocrea lutea (4), Penicillium oxalicum (10) and Preussia sp. (5) presented the strongest antimicrobial activity. This study confirmed the variation of different isolates of the same species in terms of antibacterial activity. Also, it indicated that the endophytic fungi of Juniperus procera plants should be another potential source of bioactive antimicrobial agents.  相似文献

We investigated the occurrence and genetic diversity of Trichoderma in the river Danube national park, a primeval, riparian forest area located south-east of Vienna (Austria) which represents one of the last cases of an original European river-floodplain landscape. Forty-six strains were isolated and identified at the species level by analysis of morphological characters, by sequence analysis of their internal transcribed spacer regions 1 and 2 (ITS 1 and 2) of the rDNA cluster and--in some cases--a fragment of the translation elongation factor 1alpha (tef1) gene, and RAPD-analysis. Twenty-one strains were positively identified as T. harzianum, thirteen as T. rossicum, four as T. cerinum, two as T. hamatum, and one each as T. atroviride and T. koningii: four strains yielded two different ITS1 and 2 as well as tef1 sequence types, which were not alignable with any known species. Our studies show that they represent two new taxa of Trichoderma.  相似文献
