Mesodermal-inducing substances were isolated from the swim bladders of carp ( Carassius auratus ). These substances, extracted with 8 M urea, can induce the development of early gastrula ectoderm cells of newts into mesodermal tissues, such as muscles, notochords, mesenchyme, renal tubules, and so on, to a high degree. Isoelectric focusing was used to isolate them as a first purifying step, and five component parts with different pH ranges were obtained. Each part was tested for mesodermal-inducing activity. On the basis of the extent and percentage of the induced tissues, two of them (parts C and D with pH ranges 6.0 to 7.0 and 7.0 to 9.0, respectively) were found to have high inducing activity, and the others had lower activity. The substances present in part C showed only low mesodermal-inducing activity, when diluted with gamma-globulin 100-fold. Floral syndromes and pollination of three species of Phyllostachys bamboos were studied in Central China in 1999 and 2000. All were protogynous. Stigmas were receptive and had pollen deposited on them 2 days before anther dehiscence. The period of anthesis in the three bamboos was 3 days. Individual pollen grains of the three species were similar in size (30–40 μm in diameter) and had features typical of wind-pollinated plants. The ratios of pollen to ovules (p/o ratio) in P . nidularia , P . heteroclada and P . nuda were 6500, 12 700 and 33 000, respectively. Mean pollen loads on each flower (one ovule) of these three species were 7.3, 8.8 and 9.4 grains, respectively. Pollen transfer in P . heteroclada and P . nuda depended on wind, and no flower visitors were seen in the field. However, in P . nidularia , Apis cerana Fab. was a frequent pollen collector observed from 1200 to 1330 h. The visits undertaken by thousands of honeybees resulted in a large number of pollen grains being released from the anthers in a short time (10–15 min) in one day, which accelerated and synchronized the release of pollen from the anthers that seemed to enhance the chance of pollination. Given that honeybees played an indirect role in pollen transfer this could partly explain the low p/o ratio in P . nidularia . Reviewing the literature, we found insect visits to flowers of bamboos were not infrequent phenomena. Non-host resistance is the most general form of disease resistance in plants because it is effective against most phytopathogens. The importance of hypersensitive responses (HRs) in non-host resistance of Nicotiana species to the oomycete Phytophthora is clear. INF1 elicitin, an elicitor obtained from the late-blight pathogen Phytophthora infestans , is sufficient to induce a typical HR in Nicotiana species. The molecular mechanisms that underlie the non-host resistance component of plant defence responses have been investigated using differential-display polymerase chain reaction (PCR) in a model HR system between INF1 elicitin and tobacco BY-2 cells. Differential-display PCR has revealed that Cdc27B is down-regulated in tobacco BY-2 cells after treatment with INF1 elicitin. Cdc27B is one of 13 essential components of the anaphase-promoting complex or cyclosome (APC/C)-type E3 ubiquitin ligase complex in yeast. This APC/C-type E3 ubiquitin ligase complex regulates G2-to-M phase transition of the cell cycle by proteolytic degradation. In this study, we investigated the roles of this gene, NbCdc27B , in plant defence responses using virus-induced gene silencing. Suppression of NbCdc27B in Nicotiana benthamiana plants induced defence responses and a gain of resistance to Colletotrichum lagenarium fungus. Elicitin-induced hypersensitive cell death (HCD) was inhibited mildly in plants silenced with tobacco rattle virus::Cdc27B. The first five variable microsatellite DNA loci for mangrove crab, Scylla paramanosain, were developed. Allelic variation and other characteristics at these loci were examined in this species captured at the Urado Bay, Japan. The number of alleles per locus ranged from 24 to 44. The expected heterozygosity across loci ranged from 0.900 to 0.999 and probability of identity (PI) ranged from 2.8 × 10?3 to 1.7 × 10?2. Therefore, these microsatellite markers could be useful for estimating effect of stock enhancement release and population genetic structure of mangrove crab. 相似文献

The phylogeny of Colocasiomyia (Drosophilidae) is analysed using data for 70 morphological characters, many of which are re‐evaluated from or added to those used previously, for an expanded taxon sample of 24 Colocasiomyia ingroup species. A special focus is put on three species, of which two have remained unresolved for their relationships to other Colocasiomyia species, and the other is a newly discovered species. The analysis results in a single, most parsimonious cladogram, in which a clade comprising the three focal species is recognized along with other clades recovered for the known species groups of Colocasiomyia. Based on this, a new species group—the gigantea group—is established, including Colocasiomyia gigantea (Okada), C. rhaphidophorae Gao & Toda, n.sp. and C. scindapsae Fartyal & Toda, n.sp. These species of the gigantea group breed on inflorescences/infructescences of the subfamily Monsteroideae (Araceae) exceptionally among Colocasiomyia species, most of which use plants of the subfamily Aroideae as their hosts. Colocasiomyia gigantea uses Epipremnum pinnatum (L.) Engler, C. rhaphidophorae uses Rhaphidophora hookeri Schott and C. scindapsae uses Scindapsus coriaceus Engler as their hosts. The host plants of the gigantea group are epiphytes and differ in the structure of spadix and the fruiting process from those of the Aroideae. To understand how the species of the gigantea group adapt to properties of their host plants, their reproductive ecology—most intensively that of C. gigantea—is investigated. The lifecycle of C. gigantea is characterized by its relatively slow embryonic development (taking approximately 6 days), the very long duration of the full‐grown first instar within the egg capsule (approximately three months) until dehiscence of host infructescence, and its relatively fast larval and pupal development (taking approximately 11 or 12 days). Some morphological adaptations and the reproductive strategy in terms of ‘egg size vs. number’ trade‐off are discussed in relation to their reproductive habits and peculiar lifecycles. 相似文献

A total of 32 wild Hokkaido sika deer (Cervus nippon yesoensis) were shot (13 in summer, nine in autumn and 10 in winter) in the Syari district, Shiretoko Peninsula of Hokkaido Island, Japan. The ingested foods, rumen fermentation parameters and microbes were determined to evaluate digestive strategy and food availability in each season. Ingested foods and ruminal characteristics greatly varied by season. Rumen digesta mainly comprised of graminoids in summer, graminoids and agricultural products in autumn, and bark and twigs in winter. Rumen pH showed seasonal differences (P<0.05) and was lowest in summer, highest in winter, and intermediate in autumn, reflecting the seasonal differences in ruminal concentration of total volatile fatty acids which were significantly lower (P<0.05) in winter than in summer and autumn. Acetate proportions were significantly higher in winter than in other seasons (P<0.05), while the opposite trend was seen in proportions of propionate and butyrate. Rumen ammonia levels showed significant seasonal differences (P<0.05), decreasing from summer to autumn to winter. Rumen protozoa levels in autumn and winter decreased to 28 and 10% of the levels observed in summer, respectively (P<0.05 for both). The rumen bacteria level in winter was lower (P<0.05) than that in autumn, but no difference was seen for the other seasonal comparisons. Gram negative cocci were present in significantly higher proportions in winter than in other seasons (P<0.05), while Gram negative curved rods were less frequently observed in winter (P<0.05). Based on these results, wild sika deer in this area are shown to survive with rumen microbial populations altered with the dietary conditions that vary greatly by season. 相似文献

Stem cell factor (SCF) has been suggested to be indispensable for the development of neural crest cells into melanocytes because Steel mutant mice (i.e., Sl/Sf1) have no pig-mented hairs. On the other hand, it has been demonstrated that the addition of endothelin 3 (ET-3) or TPA to neural crest cell cultures can induce melanocyte differentiation without addition of extrinsic SCF. In this study, we excluded the influence of intrinsic SCF by using SI/SI mouse embryos to study more precisely the effects of natural cytokines, such as extrinsic soluble SCF or ET-3, or chemical reagents, such as TPA or cholera toxin. We found that SCF is supplied within the wild-type neural crest explants and that ET-3 cannot induce melanocyte differentiation or proliferation without SCF. These results indicate that SCF plays a critical role in survival or G1/S entry of melanocyte progenitors and that SCF initially stimulates their proliferation and then ET-3 accelerates their proliferation and differentiation. TPA has the ability to elicit neural crest cell differentiation into melanocytes without exogenously added SCF but it is not as effective as SCF because many more melanocytes developed in the wild-type neural crest explants cultured with TPA. 相似文献
