Siddiqi, M. Y. and Glass, A. D. M. 1987. Regulation of K+ influxin barley: Evidence for a direct control of influx by K+ concentrationof root cells.—J. exp. Bot. 38: 935–947. The kinetics of K+ (86Rb+) influx into intact roots of barley(Hordeum vulgare L. cv. Fergus) seedlings having different combinationsof root and shoot [K+], different growth rates and differentroot:shoot weight ratios were studied. K+ influx was stronglycorrelated with root [K+]; shoot [K+], growth rates, and root:shoot ratios appeared to have little effect on K+ influx. Adetailed study showed that both Vmax and Km for K+ influx wereaffected by root [K+] but not by shoot [K+]. We have suggestedthat factors such as growth rates and root: shoot ratio mayaffect K+ influx indirectly primarily via their influence onroot factors such as root [K+]. We have reiterated that othertypes of kinetic control, e.g. increased or decreased synthesisof ‘carrier systems’, may operate in addition todirect (allosteric?) control of K+ influx by root [K+]. Net accumulation of Cl by intact barley plants was virtuallyeliminated in roots and reduced by 40% in shoots when externalmedia (0.5 mol m–3 CaSO4 plus 0–5 mol m–3KCI) were supplemented with 0.25 mol m Ca(NO3)2. Plasmalemma36Cl influx (oc) was shown to be insensitive to externalNO3- in plants which had previously been grown in solutionslacking –3, but oc became sensitive to NO3-after a lagperiod of 3–6 h. Kinetic analyses revealed that the inhibitionof 36C1 influx by external NO3- was complex. At 0.25mol m–3 NO3- the Vmax for Cl influx was reducedby greater than 50%, with insignificant effects upon Km. At0.5 mol m–3 NO3- there was no further effect upon Vmaxbut Km for influx increased from 38±5 mmol m–3to 116±26 mmol m–3. By contrast, Cl effluxwas found to be insensitive to external NO3-. A model for theregulation of Cl influx is proposed which involves bothnegative feedback effects from vacuolar NO3- +Cl) concentrationand (external) NO3- inhibition of Cl influx at the plasmalemma.These combined effects serve to discriminate against Claccumulation, favouring NO3- accumulation, when the latter ionis available. Such observations are inconsistent with recentproposals for the existence of bona fide homeostats for chlorideaccumulation in higher plants. Eight cvs of barley (Hordeum vulgare L.) were separately plantedwith Wild Oats (Avena fatua L., genetically pure line CS40)in a sand culture with two external K+ concentrations. Substantialdifferences were observed among barley cvs in their abilityto compete with wild oat. The variety Fergus was highly competitiveat both high and low [K+]e, whereas Steptoe was competitiveonly at high [K+]e, and Compana was only weakly competitivewith wild oat. The differences between barley cvs were relatedto their previously reported efficiencies of K+ uptake and utilization. Interactive effects of K+ and N (principally NH4+) on plant growth and ion uptake were investigated using hydroponically grown rice (Oryza sativa L. cv. M202) seedlings by varying the availability of NH4+ or NO3? and K+ during an 18d growth period, a 3d pretreatment period and during flux measurements. Plants grew best in media containing 100 mmol m?3 NH4+ and 200mmolm?3 K+ (N100/K200), followed by N2/K200 < N100/K2 < N2/K2. 86Rb+(K+) fluxes were increased by exposure to N during the 18 d growth period and the 3 d of pretreatment, but decreased by the presence of NH4+ during flux measurements. This inhibition was a function of prior N/K provision and the [NH4+]0 present during flux determinations. NH4+ was least inhibitory to 86Rb+(K+) influx in high-N/low-K plants. Pretreatments with K+ failed to stimulate NH4+ uptake, and the presence of K+ in the uptake solutions reduced NH4+ fluxes only in high-N/low-K plants.  相似文献

SUMMARY 1. The study presents adaptive and acclimatory changes observed in Cyprinus carpio communis L with respect to ovarian cycle and serum levels of electrolytes, calcium, total proteins and albumin as a consequence of wide temperature and sunshine variation in the natural environment of Dal lake in Srinagar, Kashmir (India).
2. The atmospheric temperatures in Srinagar show well-defined seasonal and diurnal variations. During the period of study, the mean monthly average temperature in summer (June-August) was 24°C whereas in winter (December-February) it fell to 3.6°C. The total hours of sunshine during the winter months were about half of those found in summer.
3. Under local thermal and photo conditions the fish was found to spawn once a year compared to biannual spawning in tropical chmates.
4. A significant decrease was found in serum sodium, potassium and chloride concentrations during the months of January and February when the mean minimum atmospheric temperatures reached −2.5°C and −l.2°C respectively.
5. The serum calcium levels showed a marked increase during the period corresponding to ovarian maturity and spawning of fish.
6. A significant rise in total serum protein concentration was induced by decreased environmental temperatures. Serum albumin levels were found to exhibit a sharp fall coinciding with the spawning period.  相似文献

Interacting effects of phosphorus, potassium and sodium on the growth and nutrient uptake in Stylosanthes fruticosa were studied in its native sandy soil and solution culture. High phosphorus in the medium had adverse effects on growth. High potassium level completely masked or lessened adverse effects of high phosphorus. High sodium level adversely affected growth only in combination with high phosphorus and low potassium. Shoot nutrient content data indicated that phosphorus per se may not inhibit growth. Selective uptake of potassium against sodium was eliminated by high phosphorus. It is suggested that high concentration of phosphorus in the medium effects membrane permeability and the effect of phosphorus on plant growth is primarily through its influence on uptake of other ions.  相似文献

Abstract. Rates of proton extrusion and potassium (86Rb) influx by intact roots of barley ( Hordeum vulgare cvs . Fergus, Conquest and Betzes) plants were simultaneously measured in short-term (15min) experiments. The nature and extent of apparent coupling between these ion fluxes was explored by manipulating conditions of temperature, pH and cation composition and concentration during flux determinations. In addition, the influence of salt status upon these fluxes was examined. At low K+ concentrations (0.01 to 1 mol m−3), H+ efflux and K+ influx were strongly correlated in both low- and high-K+ roots, although K+: H+ exchange stoichiometries were almost consistently greater than 2:1. At higher concentrations (1 to 5 mol m−3), H+ efflux was either reduced or remained unchanged while K+ influxes increased. In the presence of Na2SO4, rates of H+ extrusion demonstrated similar cation dependence, although below 10 mol m−3 Na2SO4, H+ fluxes were generally 50% lower than in equivalent concentrations of K2SO4. These observations are considered in the context of current hypotheses regarding the mechanisms of k+/H+ exchange.  相似文献

Abstract. A microcomputer-based system for the measurement of changes of ion activity (H+, NO3, K+) in the uptake solutions during ion absorption by roots of intact plants is described. Ion activities are measured by means of ion-specific electrodes in a multichannel programmable voltmeter (data acquisition/control unit) which is activated by means of a microcomputer. Incoming data are stored digitally on tape cassettes or floppy discs via the microcomputer. The speed of sampling and large numbers of samples which can be simultaneously measured and recorded make this an extremely versatile system which might be employed for measuring and recording any number of analogue voltage signals.  相似文献

Barley (Hordeum vulgare L.) varieties differed in their raponseto [K+]0, in terms of their utilization efficiencies (UE = freshweight. concentration of [K+]1–1). At low [K+]0, Compana,an efficient-non-responder demonstrated superior utilizationof absorbed K+. On the other hand, at high [K+]0, Fergus (anefficient responder) and BT 334 (an inefficient responder) hadhigher UE values for K+ than Compana which performed poorlyat this [K+]0. Kinetic parameters for K+ activation of the enzyme pyruvatekinase from 12 barley varieties, representing a range of UEvalues, were determined. Varieties showed substantial differencesin their Vmax values (P<0·01). Compana, an efficientvariety, had the highest Vmax (31 µmol g–1 freshwt. h–1) which was about 50% higher than that of Mingo,an inefficient variety. By contrast, Km values for the enzymeswere not significantly different among varieties The mean valuesfor all varieties (3·9±0·15 mol m–3K+) is far below the estimated cytoplasmic [K+] (100-200 molm–3). It is, therefore, unlikely that differences in theutilization of K+ by these varieties can be explained on thebasis of differential requirements for (K+) activation of theseenzymes.
