Interaction of rice dwarf virus outer capsid P8 protein with rice glycolate oxidase mediates relocalization of P8 总被引:2,自引:0,他引:2
Yeast two-hybrid and coimmunoprecipitation assays indicated that P8, an outer capsid protein of Rice dwarf phytoreovirus (RDV), interacts with rice glycolate oxidase (GOX), a typical enzyme of peroxisomes. Confocal immunofluorescence microscopy revealed that P8 was colocalized with GOX in peroxisomes. Time course analysis demonstrated that the localization of P8 in Spodoptera frugiperda cells changed from diffuse to discrete, punctuate inclusions during expression from 24 to 48 h post inoculation. Coexpression of GOX with P8 may target P8 into peroxisomes, which serve as replication sites for a number of viruses. Therefore, we conclude that the interaction of P8 with the GOX of host cells leads to translocation of P8 into peroxisomes and we further propose that the interaction between P8 and GOX may play important roles in RDV targeting into the replication site of host cells. Our findings have broad significance in studying the mechanisms whereby viruses target appropriate replication sites and begin their replication. 相似文献

In the 200 years since the Sumatran rhinoceros was first scientifically described (Fisher 1814), the range of the species has contracted from a broad region in Southeast Asia to three areas on the island of Sumatra and one in Kalimantan, Indonesia. Assessing population and spatial distribution of this very rare species is challenging because of their elusiveness and very low population number. Using an occupancy model with spatial dependency, we assessed the fraction of the total landscape occupied by Sumatran rhinos over a 30,345-km2 survey area and the effects of covariates in the areas where they are known to occur. In the Leuser Landscape (surveyed in 2007), the model averaging result of conditional occupancy estimate was or 2,371.47 km2, and the model averaging result of replicated level detection probability ; in Way Kambas National Park—2008: or 634.18 km2, and ; and in Bukit Barisan Selatan National Park—2010: or 819.67 km2, and . In the Leuser Landscape, rhino occurrence was positively associated with primary dry land forest and rivers, and negatively associated with the presence of a road. In Way Kambas, occurrence was negatively associated with the presence of a road. In Bukit Barisan Selatan, occurrence was negatively associated with presence of primary dryland forest and rivers. Using the probabilities of site occupancy, we developed spatially explicit maps that can be used to outline intensive protection zones for in-situ conservation efforts, and provide a detailed assessment of conserving Sumatran rhinos in the wild. We summarize our core recommendation in four points: consolidate small population, strong protection, determine the percentage of breeding females, and recognize the cost of doing nothing. To reduce the probability of poaching, here we present only the randomized location of site level occupancy in our result while retaining the overall estimation of occupancy for a given area. 相似文献

Anchoring PdCu Amorphous Nanocluster on Graphene for Electrochemical Reduction of N2 to NH3 under Ambient Conditions in Aqueous Solution 下载免费PDF全文
As an alternative approach for N2 fixation under milder conditions, electrocatalytic nitrogen reduction reaction (NRR) represents a very attractive strategy for sustainable development and N2 cycle to store and utilize energy from renewable sources. However, the research on NRR electrocatalysts still mainly focuses on noble metals, while, high costs and limited resources greatly restrict their large‐scale applications. Herein, as a proof‐of‐concept experiment, taking PdCu amorphous nanocluster anchored on reduced graphene oxide (rGO) as NRR catalysts, the optimum Pd0.2Cu0.8/rGO composite presents a synergistic effect and shows superior electrocatalytic performance toward NRR under ambient conditions (yield: 2.80 µg h?1 mgcat.?1 at ?0.2 V vs reversible hydrogen electrode), which is much higher than that of monometallic, especially noble metal, counterparts. The superior catalytic performance of alloy catalysts with low noble metal loading would strongly spur interest toward more researches on NRR catalysts in the future. 相似文献

Efficient and selective dehydrogenation of hydrazine borane (HB), a novel hydrogen storage material with very high hydrogen content (HB, 15.4 wt%), is a key challenge for a fuel‐cell‐based hydrogen economy. However, even using the noble metal catalysts for HB decomposition, the activities are still far from satisfying, to say nothing of non‐noble‐metal‐containing catalysts. In response, as a proof‐of‐concept experiment, herein, noble‐metal‐free NiFe–CeOx nanoparticles are successfully immobilized on an MIL‐101 support without surfactant by a simple liquid impregnation method. Unexpectedly, the resultant Ni0.5Fe0.5–CeOx/MIL‐101 catalyst shows good performance, including 100% H2 selectivity, 100% conversion, and record catalytic activity (351.3 h?1) for hydrogen generation at mild temperature, which is even better than most of the noble metal heterogeneous catalysts and might be attributed to the good dispersion and uniform particle size of the Ni0.5Fe0.5–CeOx nanoparticles due to steric restrictions effect of the MIL‐101 support. Additionally, extending MIL‐101 to some other important kinds of metal–organic framework (MOF) structures, the resultant NiFe–CeOx/MOF catalysts all show good catalytic activity toward HB decomposition, showing the universality of the MOF supported NiFe–CeOx catalysts. 相似文献

All infectious disease oriented clinical diagnostic assays in use today focus on detecting the presence of a single, well defined target agent or a set of agents. In recent years, microarray-based diagnostics have been developed that greatly facilitate the highly parallel detection of multiple microbes that may be present in a given clinical specimen. Gynura procumbens (Lour.) Merris one of medicinal plant which was carried out used as antioxidant, anticancer, anti-inflammatory, hepatoprotective, and antimicrobial. Many strategies were used to increase the production of biomass and valuable compounds. This study was to investigate the variation effect of growth regulators and immersion frequency on production of biomass and flavonoid contained of G. procumbens shoots culture in temporary immersion bioreactor. Stem nodes were used as an explants and induction of shoots were done in solid MS medium supplemented with many kinds of growth regulator. The best treatments were used to produce biomass and flavonoid compounds in temporary immersion bioreactor; there are combination of IAA 2?mg/L and BA 4, 6, 8?mg/L and immersion frequency (5 min each 3?h; 15?min each 12?h). Results showed that the growths of G. procumbens shoots in solid MS medium were influenced by supplementation of growth regulators. MS medium supplemented with single cytokinine (6 mg/L kinetin) and combination of auxin (IAA) and cytokinine (BA) caused increasing of shoots growth. Production of biomass of G. procumbens in temporary immersion bioreactor was achieved in long immersion interval (12?h) and highest flavonoid production was obtained in combination treatment of immersion frequency 15?min each 12?h and MS medium supplemented with IAA 2?mg/L, BA 8?mg/L. 相似文献

The change in stored carbon (C) stocks was assessed for a 700 km~2 area where forestcover decreased from 60% to 10% in the last 30 years. At the same time, the area under coffee increased from 7% to 70% with a gradual evolution from open "sun coffee" systems to multi-strata "shade coffee" systems that provide a partial compensation for C loss. The use of a generic tropi-cal forest rather than tree-specific allometric equation can lead to substantial (up to 100%) overes-timates of aboveground biomass depending on wood density and tree shape. The shoot: root ratio (biomass) of coffee shifted with age, from the 4∶1 value often assumed for tropical trees to 2∶1.Annual aboveground C stock accumulation rates during the establishment stage after slash-and-burn land clearing were 1, close to 2 or 3.5 Mg C ha~(-1)a~(-1) for sun coffee, shade coffee and fallowregrowth, respectively. Forest remnants, shade coffee and sun coffee had soil C stocks in the up-per 30 cm of the soil that were 79%, 60% or 45%, respectively, of the values expected for primary forest in Sumatra. Total C stock (time averaged, above-0.3 m in the soil) for forest, shade and sun coffee was 262, 82 and 52 Mg C ha~(-1), respectively. In the 1970-1984 period, while forest cover was reduced from 59.5% to 19.7%, the landscape lost on average 6.8 Mg C ha~(-1) a~(-1). In the1984-2000 period forest cover was further reduced to 12.6%, but the landscape lost only 0.39 MgC ha~(-1) a~(-1), as forest loss was partially compensated by an increase in shade coffee systems. Conversion of all current sun coffee to shade coffee systems while protecting the remaining forest,could increase average landscape level C stocks by 10 Mg ha~(-1) over a time frame of say 20 years,or 0.5 Mg C ha~(-1) a~(-1). 相似文献

An efficient plant regeneration procedure has been established from hypocotyl explants of the common ice plant, Mesembryanthemum crystallinum L, a halophytic leaf succulent that exhibits a stress-induced switch from C3 photosynthesis to crassulacean acid metabolism
(CAM). Somatic embryos were initiated and developed up to globular and heart stages in Murashige and Skoog (MS) media supplemented
with 3% sucrose, 0.6% bacto-agar, 80 mM NaCl, 5 μM 2,4-D and 1 μM kinetin. High frequency regeneration occurred when somatic
embryos were germinated on media that lacked 2,4-D. High cytokinin treatment suppressed normal growth of embryos and favored
abnormal embryo proliferation. Without growth regulators, regenerated plants rooted on MS medium with 100% efficiency. Mature,
regenerated plants were fertile and morphologically identical to seed-derived plants.
