Hydrodynamic forces can affect survival as well as limit the movement of motile benthic animals. An animal's danger of dislodgement depends on the hydrodynamic forces it experiences in its microhabitat relative to the force required to dislodge it (tenacity) from the substratum. We measured water flow and substratum characteristics in two different habitats of the shore crab Pachygrapsus crassipes: a wave-swept rocky shore and an intertidal mudflat. The maximum water velocities and accelerations in the microhabitats of the crabs at the wave-swept site were three times and two times greater, respectively, than at the mudflat site. In the laboratory, we measured the tenacity of crabs of various sizes on different substrata, and also measured their drag, lift and added-mass coefficients. Using these data, we calculated the flow conditions under which crabs would be overturned or sheared off the substratum in their two habitats. The net horizontal force (drag plus acceleration reaction) required to dislodge a crab on a rugose rock substratum was an order of magnitude greater than on smooth rock and two orders of magnitude greater than on mud. Our calculations indicate that, under non-storm conditions, crabs will not be dislodged from the substratum in either the mudflat or the wave-swept habitat when grasping the substratum with maximum tenacity. Moving crabs have lower tenacity and our calculations predict that hydrodynamic forces will restrict the mobility of large crabs more than that of small ones on smooth, but not on rugose rock.  相似文献

The South American weakly-electric knifefish (Apteronotidae) produce highly diverse and readily quantifiable electrocommunication signals. The electric organ discharge frequency (EODf), and EOD modulations (chirps and gradual frequency rises (GFRs)), vary dramatically across sexes and species, presenting an ideal opportunity to examine the proximate and ultimate bases of sexually dimorphic behavior. We complemented previous studies on the sexual dimorphism of apteronotid communication signals by investigating electric signal features and their hormonal correlates in Apteronotus bonapartii, a species which exhibits strong sexual dimorphism in snout morphology. Electrocommunication signals were evoked and recorded using a playback paradigm, and were analyzed for signal features including EOD frequency and the structure of EOD modulations. To investigate the androgenic correlates of sexually dimorphic EOD signals, we measured plasma concentrations of testosterone and 11-ketotestosterone. A. bonapartii responded robustly to stimulus playbacks. EODf was sexually monomorphic, and males and females produced chirps with similar durations and amounts of frequency modulation. However, males were more likely than females to produce chirps with multiple frequency peaks. Sexual dimorphism in apteronotid electrocommunication signals appears to be highly evolutionarily labile. Extensive interspecific variation in the magnitude and direction of sex differences in EODf and in different aspects of chirp structure suggest that chirp signals may be an important locus of evolutionary change within the clade. The weakly-electric fish represent a rich source of data for understanding the selective pressures that shape, and the neuroendocrine mechanisms that underlie, diversity in the sexual dimorphism of behavior.  相似文献

Summary The DNA sequences of the mercuric resistance determinants of plasmid R100 and transposon Tn501 distal to the gene (merA) coding for mercuric reductase have been determined. These 1.4 kilobase (kb) regions show 79% identity in their nucleotide sequence and in both sequences two common potential coding sequences have been identified. In R100, the end of the homologous sequence is disrupted by an 11.2 kb segment of DNA which encodes the sulfonamide and streptomycin resistance determinants of Tn21. This insert contains terminal inverted repeat sequences and is flanked by a 5 base pair (bp) direct repeat. The first of the common potential coding sequences is likely to be that of the merD gene. Induction experiments and mercury volatilization studies demonstrate an enhancing but non-essential role for these merA-distal coding sequences in mercury resistance and volatilization. The potential coding sequences have predicted codon usages similar to those found in other Tn501 and R100 mer genes.  相似文献

A new method for preparing non-parenchymal rat liver cells (NPC) is described. The liver cell suspension, prepared by perfusing the liver with collagenase, was treated with enterotoxin from Clostridium perfringens for 15 min. The enterotoxin made the parenchymal cells leaky, and these cells could be separated from the NPC by centrifugation in a solution containing Nycodenz (20%, w/v). During the centrifugation, the NPC floated, while the parenchymal cells sedimented. The yield of NPC per liver (200 g rat) was about 250 X 10(6) cells. The NPC were further separated into endothelial cells, Kupffer cells and stellate cells by centrifugal elutriation. This method was particularly useful for preparing endothelial cells in high yield (100 X 10(6) cells per liver). Intravenously injected formaldehyde-treated albumin was selectively taken up by the endothelial cells. Isolated endothelial cells in suspension as well as in surface culture maintained their ability to endocytose this ligand.  相似文献

The intracellular movement, following uptake of 125I-labelled denatured serum albumin into nonparenchymal liver cells, was followed by means of subcellular fractionation. Isolated nonparenchymal rat liver cells were prepared by means of differential centrifugation. The cells were homogenized in a sonifier and the cytoplasmic extract subjected to isopycnic centrifugation in a sucrose gradient. The intracellular movement of the labelled albumin was followed by comparing the distribution profile of radioactivity in the sucrose gradient with those of marker enzymes for plasma membrane and lysosomes. The distribution profiles for radioactivity after the cells had been exposed to the labelled denatured albumin for different time periods indicated that the radioactivity was first associated with subcellular fractions of lower modal densities than the lysosomes. With time of incubation the radioactivity moved towards higher densities. After prolonged incubations in the absence of extracellular labelled denatured albumin the radioactivity peak coincided with that of the lysosomal marker β-acetylglucosaminidase. When the cells were treated with the lysosomal inhibitor leupeptin, degradation of the labelled albumin was decreased, resulting in a massive intracellular accumulation of radioactivity. The radioactivity peak coincided with the peak of activity for the lysosomal marker β-acetylglucosaminidase, suggesting lysosomal degradation.  相似文献

The ~30,000 hectare classical Costa Rican Parque Nacional Santa Rosa has used about 35 years and $107 million to be converted to the 169,000 ha government-NGO hybrid Área de Conservación Guanacaste (ACG). This semi-decentralized conservation entity has today a staff of ~150 paraprofessional resident Costa Ricans, biodeveloping at least 650,000 multicellular species (Eucaryotes) into perpetuity for ACG survival through being integrated with its local, regional, national, and international society. ACG began in 1985 as an ongoing exercise of landscape-level ecosystem rescue and restoration of a continuous swath from 6 km out in the Pacific ocean, across dry forested lowlands, up and over the volcanic Cordillera Guanacaste, and down into the rain-forested Caribbean lowlands. It is being impacted by climate change, yet its diverse ecosystems hold hope for major biodiversity survival, albeit in new community assemblages. It quickly became simultaneously a biophysical challenge and an administratively novel challenge in decentralized conservation in a democratic tropical country. ACG specializes at being managed by on-the-job stimulated and trained residents with minimal formal education, searching for ways to involve ACG in its society without damaging its wildness, and pioneering ways to render wild biodiversity to being a welcome member at society's negotiating table. It continues to pay its bills through government subsidy, generous donors, payments for services, project grants, and huge in-kind contributions from mutualisms. ACG hopes that the concept will spread south–south to other tropical countries while they still have some of their wild biodiversity with which to integrate.  相似文献

The semiaquatic weed Mimosa pigra has negative impacts on biodiversity, fishing, crop and livestock production, and tourism in most places where it has been introduced, established and proliferated. Many of the ecological impacts are well known, but its impacts on rural livelihoods are less well documented, especially in Africa. We mapped the distribution of M. pigra in eastern and southern Africa, and then compared that with its potential distribution based on an ecoclimatic niche model. Household interviews were conducted to assess the impacts of this weed on local livelihoods. Mimosa pigra was found to be invasive in western Ethiopia, around the shores of Lake Victoria and Lake Tanganyika, and along the Tanzanian coastline, northern Malawi, parts of Mozambique and along the Kafue River and in the Barotse floodplain on the Zambezi River in Zambia. According to respondents living along the Kafue River floodplains in Zambia, it has a negative impact on biodiversity, wildlife, livestock, crop production, fishing and mobility. Dense stands prevented the movement of people and livestock, limiting access to croplands, grazing lands and fishing areas. Fish catches have been reduced and fishing equipment damaged. All respondents agreed that their livelihood options would be considerably enhanced if M. pigra was removed from the landscape. Based on its current and potential impact, we therefore recommend that an integrated management plan be developed and implemented, including the appropriate use of biological control agents to reduce the negative impacts of the weed.  相似文献
