Adventitious roots are natural roots which develop from the differentiated cells of any organ such as leaf, root or stem. These roots are attractive source of phytochemicals due to their genetic and biosynthetic stability. Their stability offers new scope for the in vitro production of valuable secondary metabolites. In the past two decades, adventitious roots have been induced from different plant systems and the suspension cultures have been established. However, the special morphology of adventitious roots hampers the scale-up processes, which needs modifications with the existing bioreactors to facilitate proper mixing, aeration and which can overcome shear stress. Various modifications have been done with the stirred tank and air-lift bioreactors and successfully used for the cultivation of adventitious roots. The progress made in this area is tremendous, in few cases adventitious root cultures are succeeded up to commercial scale with step-wise viz. small scale to large-scale by establishing the standards with various parameters which affect the biomass accumulation and metabolite productivity. This review addresses the recent progresses made in this area and discusses the potential applications and limitations.  相似文献

During the years 1990-2003 a large number of investigations were conducted using rodents, cultured rodent and human cells, and freshly collected human blood lymphocytes to determine the genotoxic potential of exposure to radiofrequency (RF) radiation. The results of most of these studies (58%) did not indicate increased damage to the genetic material (assessed from DNA strand breaks, incidence of chromosomal aberrations, micronuclei and sister chromatid exchanges) in cells exposed to RF radiation compared to sham-exposed and/or unexposed cells. Some investigations (23%) reported an increase in such damage in cells exposed to RF radiation. The observations from other studies (19%) were inconclusive. This paper reviews the investigations published in scientific journals during 1990-2003 and attempts to identify probable reason(s) for the conflicting results. Recommendations are made for future research to address some of the controversial observations.  相似文献

Exosomes, which act as biological cargo vessels, are cell-released, phospholipid-enclosed vesicles. In eukaryotic cells, exosomes carry and exchange biological materials or signals for the benefit or detriment to the cells. Thereby, we consider exosomes to be molecular Palkis (carriers). Although exosomes are currently one of the most popularly researched cellular entities, they have remained largely enigmatic and warrant continued investigation into their structure and functions. These membraned vesicles are between 30 and 150 nm in diameter and are actively secreted by all cell types. While initially considered cellular “trash bags,” recent years have revealed exosomes to be dynamic and multi-functional vesicles that may play a crucial role in cancer development, progression and metastasis. Thereby, they have the potential to be used in development of therapeutic modalities for cancer and other diseases. As more research studies emerge, it’s becoming evident that exosomes are released by cells with a purpose and are representatives of certain cell types and disease conditions. Hence, they may also be used as biomarkers for the detection of cancer initiation, progression and organotropic metastatic growth of cancer cells. This review will focus on the recent developments achieved in identifying the role of exosomes in cancer development and progression as well as therapeutic implications. The review will also discuss the pitfalls of methodologies used for the extraction of exosomes.  相似文献

Larvicidal activity of acetone and petroleum ether extracts of four plants Murraya koenigii, Coriandrum sativum, Ferula asafoetida, Trigonella foenum graceum and synthetic larvicides Fenthion and Temephos used alone and in combination was carried out against A. aegypti larvae under laboratory conditions. Optimum values were found out by using different concentrations of each plant (ranged 25 ppm-900 ppm) and both synthetic larvicides (ranged 0.01-7.5 ppm). The synergistic study was carried out by using 0.05 ppm of Temephos and Fenthion with 25ppm of M. koenigii, F. asafetida, T. foenum graceum and 100 ppm of C. sativum. All the plants showed potential synergistic activity although showed comparatively poor larvicidal activity when tested individually.  相似文献

Metacid 50, the commercial grade of methyl parathion (O,O-dimethyl-O-4-nitrophenyl phosphorothionate), a commonly used organophosphorus insecticide, was tested for its genotoxicity in Swiss albino mice using the sperm abnormality assay. Sperms of albino mice were examined at two time intervals, 1 week and 5 weeks after a single acute oral treatment with the pesticide at four dose levels, viz., 75.0, 37.5, 18.75 and 9.375 mg/kg body weight corresponding to 1/2 LD50, 1/4 LD50, 1/8 LD50 and 1/16 LD50 values respectively. A dose-related statistically significant increase in the percentage of abnormal sperm observed indicates the genotoxic potency of methyl parathion.  相似文献

Human peripheral blood samples collected from three healthy human volunteers were exposed in vitro to pulsed-wave 2450 MHz radiofrequency (RF) radiation for 2 h. The RF radiation was generated with a net forward power of 21 W and transmitted from a standard gain rectangular antenna horn in a vertically downward direction. The average power density at the position of the cells in the flask was 5 mW/cm(2). The mean specific absorption rate, calculated by finite difference time domain analysis, was 2.135 (+/-0.005 SE) W/kg. Aliquots of whole blood that were sham-exposed or exposed in vitro to 50 cGy of ionizing radiation from a (137)Cs gamma-ray source were used as controls. The lymphocytes were examined to determine the extent of primary DNA damage (single-strand breaks and alkali-labile lesions) using the alkaline comet assay with three different slide-processing schedules. The assay was performed on the cells immediately after the exposures and at 4 h after incubation of the exposed blood at 37 +/- 1 degrees C to allow time for rejoining of any strand breaks present immediately after exposure, i.e. to assess the capacity of the lymphocytes to repair this type of DNA damage. At either time, the data indicated no significant differences between RF-radiation- and sham-exposed lymphocytes with respect to the comet tail length, fluorescence intensity of the migrated DNA in the tail, and tail moment. The conclusions were similar for each of the three different comet assay slide-processing schedules examined. In contrast, the response of lymphocytes exposed to ionizing radiation was significantly different from RF-radiation- and sham-exposed cells. Thus, under the experimental conditions tested, there is no evidence for induction of DNA single-strand breaks and alkali-labile lesions in human blood lymphocytes exposed in vitro to pulsed-wave 2450 MHz radiofrequency radiation, either immediately or at 4 h after exposure.  相似文献

The genotoxic potential of the jet fuels, Jet-A and JP-8, were examined in mice treated on the skin with a single dose of 240 mg/mouse. Peripheral blood smears were prepared at the start of the experiment (t = 0), and at 24, 48 and 72 h following treatment with jet fuels. Femoral bone marrow smears were made when all animals were sacrificed at 72 h. In both tissues, the extent of genotoxicity was determined from the incidence of micronuclei (MN) in polychromatic erythrocytes. The frequency of MN in the peripheral blood of mice treated with Jet-A and JP-8 increased over time and reached statistical significance at 72 h, as compared with concurrent control animals. The incidence of MN was also higher in bone marrow cells of mice exposed to Jet-A and JP-8 as compared with controls. Thus, at the dose tested, a small but significant genotoxic effect of jet fuels was observed in the blood and bone marrow cells of mice treated on the skin.  相似文献
