Sucrose was found to have a modulating effect on the morphogenesisof Anagallis arvensis L. leaves cultured in a Murashige-Skoogmedium. Root formation and growth seem to be more independentthan other morphogenetic expressions. Roots formed without exogenoussugars at 25°C but sucrose seemed to be necessary at 32and 35°C. Sucrose at 3% improved shoot formation at 25°Cand had an inhibitory effect at 6%concentration and 35°C.Shoot growth (internode length) is inhibited by sucrose concentrationshigher than 3%. Sucrose could also replace light irradiancein regulating shoot and leaf growth. A higher sucrose concentration,than that required for roots and shoots formation, is necessaryfor flower and fruit formation, but sucrose could not replacethe photoperiod requirement for flowering in culture medium. The hypothesis that light- and oxygen-induced proteolysis inchloroplasts is mediated by active oxygen species was examined.In order to determine whether or not H2O2 and/or {dot}OH radicalsare involved in these degradative processes we compared thedegradation of proteins in isolated oat chloroplasts exposedto white light at 80 W m-2 with that in chloroplasts incubatedin darkness in the absence or presence of H2O2 or a {dot}OH-generatingsystem composed by ascorbic acid, FeCl3 and H2O2 (Asc-Fe-H2O2).Light enhanced the rate of degradation of at least 18 polypeptides,while proteolysis was almost negligible in darkness in the abscenceof additives. H2O2 had a very small effect. However, Asc-Fe-H2O2-treatedchloroplasts in darkness showed a pattern of protein degradationalmost identical to that observed in the light. A thylakoid-boundendopeptidase (EP), the activity of which increased under photooxidativeenvironmental conditions and treatment with an {dot}OH-generatingsystem, was partially purified and characterized as a serinetypeprotease. Treatments with inhibitors of serine-type proteaseprevented both light- and Asc- Fe-H2O2-induced proteolysis.EP was more active against both soluble and membranous proteinsthat had been pretreated with Asc-Fe-H2O2 than against untreatedproteins. An outbreak of paralysis among 16- to 20-week-old CFW Swiss Webster sentinel mice developed in one of our barrier facilities. Two months after arrival and over a period of four weeks, six of 400 mice purchased from an approved vendor, developed progressive hind limb paralysis without other clinical signs of disease. On the basis of the histopathologic changes and negative serologic test results, lymphoblastic lymphoma causing compression of the spinal cord was diagnosed. There were two leading features to this outbreak: its unusual epidemiologic presentation, and the localization of the lesions principally in the lumbar muscles. A presumptive diagnosis of retroviral infection with Abelson's murine leukemia virus (A-MuLV) was established on the basis of histopathologic and immunohistochemical findings. Little is known about retroviral status in many commercial colonies, and few users report presence of spontaneous lymphomas. This report points out complications derived from commercially available animals that carry endogenous retroviruses. It also emphasizes the need of diagnosing and reporting clusters of hind limb paralysis or lymphomas in mice to assess the prevalence and relevance of retroviral infections in commercial colonies.  相似文献

Summary Cultures ofMinthostachys mollis were established from nodal explants obtained under aseptic conditions. Explants were cultured on Murashige and Skoog (MS) half-strength medium containing 6-benzyladenine (BA), and/or naphthaleneacetic acid (NAA). Optimum numbers of healthy shoots were induced on media containing 0.05 μM NAA plus 2.2 μM BA; higher concentrations caused more hyperhydricity and less extension. Rooting was achieved on half-strength MS medium with 0.05 μM NAA. Plantlets were acclimatized and successfully transferred to soil. Essential oil composition of the regenerated plants were determined by gas chromatography/mass spectrometry and little differences were found in the essential oil composition with the plant grown in the wild.  相似文献

Exudation of cations and of amino acids from detached oat seedling leaves ( Avena sativa L. cv. Victory) floated on water or solutions was measured under varied conditions. A small amount of exudation in the first 4 h, greater in the dark than in white light, is followed, one to 8 days later, by a relatively great increase which approximately accompanies senescence. This second increase is delayed by kinetin and is accelerated by abscisic acid or methyl jasmonate. both of which accelerate senescence. A nitrogen atmosphere (in darkness) causes immediate large exudation, mainly of potassium ions, and this is effectively delayed by light. There are two exceptions to the parallel between exudation and senescence; n-pentanol, which strongly delays senescence in darkness, nevertheless increases exudation both of potassium and of amino acids, probably due to a direct effect on the plasmalemma. Cycloheximide, which also delays senescence, increases the exudation somewhat. Kinetin prevents or delays exudation under all conditions. Thus, the permeability of the plasmalemma increases greatly along with other criteria of senescence, but this change is probably not the principal cause of the senescence syndrome. Some of the effects are considered to result from reduction in available energy and others from a direct influence on plasmalemma permeability.  相似文献

Changes in CuZn-SOD actvity and content in isolated wheat chloroplastsunder the light, and the involvement of protease(s) and/or activeoxygen species in this process were studied. Both SOD activityand content decayed with exposure time to photooxidative stress.Ascorbate, a H2O2 scavenger, prevented photooxidation-associatedinactivation of SOD, while benzoate, a OH scavenger, preventedSOD degradation. Wheat chloroplasts incubated in the dark didnot hydrolyze exogenous or endogenous SOD, either H2O2-pretreatedor not. Protease inhibitors did not prevent SOD degradationunder photooxidative treatment, suggesting that plastid protease(s)did not participate in this process. Purified chloroplast CuZn-SODwas exposed to H2O2 and or OH-generating systems. had no effect on either SOD activity or stability (estimated bynative PAGE). The genus Lotus comprises a heterogeneous group of annual and perennial species. Lotus japonicus (with MG20 and Gifu ecotypes) has been adopted as one of the model legumes in genetic and genomic studies. Other Lotus species, such us Lotus burttii and Lotus filicaulis, have also been used in genetic and genomic studies because of their capacity to produce fertile progenies in crosses with L. japonicus. In the present work, physiological responses to salt stress in four Lotus genotypes were evaluated on the basis of growth and associated parameters, such as photosynthesis, ions, relative water content, oxidative damage and antioxidant system responses, using two NaCl levels applied by acclimation for up to 28 and 60 d. Growth responses varied with plant developmental stage in the four Lotus genotypes. L. japonicus MG20 was found to be a salt-tolerant genotype, mainly when exposed to salt stress at the young plant stage. The capacity of Lj MG20 to sustain growth under salt stress was correlated with enhancement of Superoxide dismutase and Glutathione reductase activities, as well as with increases in total and reduced glutathione content and lower Na+ accumulation in leaves. These results suggest that enhancement of antioxidant responses in Lj MG20 contributed to improve salt stress tolerance at early stages. On the other hand, after long-term high NaCl stress treatment, L. filicaulis exhibited lower biomass reduction, lower oxidative damage and Na+ accumulation in leaves than the control treatment; hence, this genotype was considered salt-tolerant. These apparently ambiguous results remark that salt tolerance, as a development-related process, was differentially expressed among the Lotus genotypes and depended on stress duration and plant phenological stage.  相似文献

The regulation of senescence by oxygen-concentration, lightirradiance and H2O2 has been studied in leaf segments of Avenasativa L. cv. Suregrain. The development of the components of the senescence process,for example chlorophyll breakdown, proteolysis (as soluble aminoacids), hydroperoxides (as malondi-aldehyde) and permeability(as conductivity) is accelerated in light as the O2-tensionincreases. In darkness, 0.3% O2 accelerates increases in hydroperoxides,permeability and proteolysis and delays the chlorophyll break-down,but 0.0005% O2 delays all the components studied. In every casethe hydroperoxide content, permeability and proteolysis areclosely related. Any treatment inducing an increase in membranepermeability causes chlorophyll bleaching (photo-oxidation)if leaf segments are then treated with light in an atmospherecontaining oxygen. Light has a modulating effect on the senescenceprocess. An irradiance lower or higher than 40 W m–2 hasan accelerating effect on the senescence process.
