A 96-well microtiter plate most-probable-number (MPN) procedure was developed to enumerate hydrocarbondegrading microorganisms. The performance of this method, which uses number 2 fuel oil (F2) as the selective growth substrate and reduction of iodonitrotetrazolium violet (INT) to detect positive wells, was evaluated by comparison with an established 24-well microtiter plate MPN procedure (the Sheen Screen), which uses weathered North Slope crude oil as the selective substrate and detects positive wells by emulsification or dispersion of the oil. Both procedures gave similar estimates of the hydrocarbon-degrader population densities in several oil-degrading enrichment cultures and sand samples from a variety of coastal sites. Although several oils were effective substrates for the 96-well procedure, the combination of F2 with INT was best, because the color change associated with INT reduction was more easily detected in the small wells than was disruption of the crude oil slick. The method's accuracy was evaluated by comparing hydrocarbon-degrader MPNs with heterotrophic plate counts for several pure and mixed cultures. For some organisms, it seems likely that a single cell cannot initiate sufficient growth to produce a positive result. Thus, this and other hydrocarbon-degrader MPN procedures might underestimate the hydrocarbon-degrading population, even for culturable organisms. 相似文献

Crude oil is a complex mixture of several different structural classes of compounds including alkanes, aromatics, heterocyclic polar compounds, and asphaltenes. The rate and extent of microbial degradation of crude oil depends on the interaction between the physical and biochemical properties of the biodegradable compounds and their interactions with the non-biodegradable fraction. In this study we have systematically altered the concentration of non-biodegradable material in the crude oil and analyzed its impact on transport of the biodegradable components of crude oil to the microorganisms. We have also developed a mathematical model that explains and accounts for the dependence of biodegradation of crude oil through a putative bioavailability parameter. Experimental results indicate that as the asphaltene concentration in oil increases, the maximum oxygen uptake in respirometers decreases. The mathematically fitted bioavailability parameter of degradable components of oil also decreases as the asphaltene concentration increases. 相似文献

Pearl millet downy mildew (DM) incidence, severity and yield losses of two pearl millet varieties (local and improved) due to the disease were determined in the field. Significant differences in the disease incidence and severity were recorded in the plots sown with metalaxyl-treated seeds and those sown with non-treated seeds, indicating the efficacy of the fungicide on the fungus. Yield losses due to non-treatment of seeds with metalaxyl was 40.88 and 45.39% in a local variety and 43.00 and 18.60% in an improved variety in the 2000 and 2001 cropping seasons respectively. Significant differences between plots sown with metalaxyl-treated and those sown with non-treated seeds were obtained for other yield components such as 1000-grains weight, panicle length and weight. 相似文献

Background
Organisms are capable of developing different phenotypes by altering the genes they express. This phenotypic plasticity provides a means for species to respond effectively to environmental conditions. One of the most dramatic examples of phenotypic plasticity occurs in the highly social hymenopteran insects (ants, social bees, and social wasps), where distinct castes and sexes all arise from the same genes. To elucidate how variation in patterns of gene expression affects phenotypic variation, we conducted a study to simultaneously address the influence of developmental stage, sex, and caste on patterns of gene expression in Vespula wasps. Aerobically grown enrichment cultures derived from hydrocarbon-contaminated seawater and freshwater sediments were generated by growth on crude oil as sole carbon source. Both cultures displayed a high rate of degradation for a wide range of hydrocarbon compounds. The bacterial species composition of these cultures was investigated by PCR of the 16S rDNA gene using multiple primer combinations. Near full-length 16S rDNA clone libraries were generated and screened by restriction analysis prior to sequence analysis. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) was carried out using two other PCR primer sets targeting either the V3 or V6-V8 regions, and sequences derived from prominent DGGE bands were compared to sequences obtained via cloning. All data sets suggested that the seawater culture was dominated by alpha-subgroup proteobacteria, whereas the freshwater culture was dominated by members of the beta- and gamma-proteobacteria. However, the V6-V8 primer pair was deficient in the recovery of Sphingomonas-like 16S rDNA due to a 3' terminal mismatch with the reverse primer. Most 16S rDNA sequences recovered from the marine enrichment were not closely related to genera containing known oil-degrading organisms, although some were detected. Anaerobic biodegradation of vegetable oil in freshwater sediments is strongly inhibited by high concentrations of oil, but the presence of ferric hydroxide relieves the inhibition. The effect of ferric hydroxide is not due to physical or chemical interactions with long-chain fatty acids (LCFAs) that are produced as intermediates during metabolism of vegetable-oil triglycerides. The anaerobic biodegradation of canola oil and mixtures of acetic and oleic acids, two important intermediates of vegetable-oil metabolism, were investigated using sediments enriched on canola oil under methanogenic and iron-reducing conditions to determine whether the effect of ferric hydroxide has a biological basis. Sediments enriched under both conditions rapidly and completely converted canola oil to methane when the initial oil concentration was relatively low (1.9 g oil/kg sediments), but the biotransformation was strongly inhibited in sediments enriched under methanogenic conditions when the initial concentration was 19 g/kg (<30% of the oil-derived electron equivalents were transferred to methane in a 420-day incubation period). Sediments enriched under iron-reducing conditions, however, completely transformed canola oil to methane in about 250 days at this initial oil concentration. The anaerobic biotransformation of mixtures of acetate and oleic acid followed a similar pattern: the rate and extent of conversion of these electron-donor substrates to methane was always higher in sediments enriched under iron-reducing than under methanogenic conditions. These results suggest that enrichment on canola oil in the presence of ferric hydroxide selects a microbial community that is less sensitive to inhibition by LCFAs than the community that develops during enrichment under methanogenic conditions. 相似文献

Manganese induces the central nervous system injury leading to manganism, by mechanisms not completely understood. Chronic exposure to manganese generates oxidative stress and induces the mitochondrial permeability transition. In the present study, we characterized apoptotic cell death mechanisms associated with manganese toxicity in rat cortical astrocytes and demonstrated that (i) Mn treatment targets the mitochondria and induces mitochondrial membrane depolarization followed by cytochrome c release to the cytoplasm, (ii) Mn induces both effector caspases 3/7 and 6 as well as PARP-1 cleavage and (iii) Mn shifts the balance of cell death/survival of Bcl-2 family proteins to favor the apoptotic demise of astrocytes. Our model system using cortical rat astrocytes treated with Mn would emerge as a good tool for investigations aimed to elucidate the role of apoptosis in manganism. 相似文献

Biostimulation by nutrient enrichment and phytoremediation were studied for the restoration of an acutely stressed freshwater wetland experimentally exposed to crude oil. The research was carried out along the shores of the St. Lawarence River at Ste. Croix, Quebec, Canada. The research determined the effectiveness of fertilizer addition in enhancing the biodegradation rates of residual oil. It further examined the rate at which the stressed ecosystem recovered with and without the addition of inorganic fertilizers and the role of nutrients in enhancing wetland restoration in the absence of healthy wetland plants. Chemical analysis of integrated sediment core samples to the depth of oil penetration within the experimental plots indicated that addition of inorganic nutrients did not enhance the disappearance of alkanes or PAHs. In surface samples, however, hydrocarbon disappearance rates were higher when the metabolic activity of wetland plants was suppressed by the removal of emergent plant growth. These results suggest that oxygen limitation plays a major role in preventing rapid biodegradation of hydrocarbons in anoxic wetland sediment. 相似文献

The fractional loss of 22 Na+ from frog sartorius muscle is increased when the tonicity of the external solution is reduced.
