Many commercial preparations of [32P]orthophosphate contain radioactive impurities that interfere with binding and transport studies in biological systems. One type of impurity is micro-particulate whereas another may be pyrophosphate. Methods of removing these impurities from radiolabeled orthophosphate solutions are described.  相似文献

The position of Helicopsyche borealis (Hagen) (Trichoptera: Helicopsychidae) larvae on the substratum surface is dependent on the current regime but varies with larval size. All size classes of larvae chose significantly different positions on the substratum under high versus low current velocities. All size classes preferred exposed surfaces under low current velocities. Small larvae preferred the upper surfaces of substrata under low current velocities and were physically displaced under high current velocities. Larger larvae also occurred on upper surfaces, but were more evenly dispersed over all surfaces than smaller larvae, and tended to aggregate on down-stream faces of rocks during high flow.  相似文献

The tryptic phosphopeptide of hen egg white riboflavin-binding protein has been found to exist as a mixture of peptides which differ only with respect to the number of covalently bound phosphoryl groups. Anion-exchange chromatography was used to separate homologues of the tryptic phosphopeptide of egg white riboflavin-binding protein. Four peptide peaks were obtained and analyzed using plasma desorption mass spectrometry. Molecular ions obtained agree closely with calculated molecular weight values for phosphopeptides with 8, 7 and 5 phosphoryl groups. Amino acid analyses showed that the octa- and hepta-phosphorylated peptides were pure and had the same amino acid compositions.  相似文献

We have assessed the effects of in vivo administration of different classes of diuretic drugs on the expression of the peripheral-type benzodiazepine binding site (PBBS) in crude membranes derived from the cortex and outer medulla of rat kidney by saturation analysis with the PBBS-selective ligands [3H]RO5-4864 and [3H]PK 11195 in cortex and [3H]RO5-4864 in outer medulla. Administration for 14-15 days of furosemide, a drug that blocks NaCl-KCl coupled transport in the thick ascending limb of the loop of Henle, produced a significant doubling in the PBBS density (Bmax) in outer medulla, a region of the kidney rich in thick ascending limbs, and produced a lesser but significant increase in PBBS density in the cortex. Conversely, administration for 14-15 days of the carbonic anhydrase inhibitor acetazolamide, which acts predominantly in the proximal tubule, and hydrochlorothiazide, which acts predominantly in the early distal tubule, elicited statistically significant increases in PBBS density in renal cortex but not in renal outer medulla. Furthermore, all drug treatments were without effect on the equilibrium dissociation constants (Kds) of [3H]RO5-4864 and [3H]PK 11195 binding to cortical and outer medullary membrane preparations. These findings demonstrate that the PBBS can be selectively "up-regulated" in different regions of the kidney by diuretic drugs with different modes/sites of action.  相似文献

The pineal has been previously shown to be an important factor in the regulation of testicular function in photoperiodic mammals. The effects of lack or increase in pineal hormones on testicular hormonal receptors has, therefore, been examined. Pinealectomy decreased the concentration of testicular LH receptors in hamsters exposed to either a long or short photoperiod but had no effect on the concentration of testicular PRL receptors. In animals exposed to a short photoperiod, pinealectomy prevented testicular regression and the concomitant decreases in total LH and PRL receptor contents. Treatment for 12 weeks with either melatonin or 5-methoxytryptamine caused a decrease in testicular PRL receptor levels, whereas the only changes in LH receptor levels were due to melatonin-induced testicular regression. The present results indicate that some of the effects of pineal hormones on the testes are independent of the pineal-induced changes in testes mass and are the consequence of long-term action. Furthermore, testicular function appears to be affected by both the lack or the increase in pineal hormones.  相似文献

Polyphenol oxidase (PPO) appears to be ubiquitous in higher plants but, as yet, no function has been ascribed to it. Herein, we report on the localization of PPO based upon biochemical fractionation of chloroplast membranes in Vicia faba (broad bean) into various complexes and immunocytochemical electron microscopic investigations. Sucrose density gradient fractionations of thylakoid membranes after detergent solubilization reveals that PPO protein (by reactivity with anti-PPO antibody) and activity (based upon ability to oxidize di-dihydroxyphenylalanine) are found only in fractions enriched in photosystem II (PSII). Furthermore, of the PSII particles isolated using three different protocols utilizing several plant species, all had PPO. Immunogold localization of PPO on thin sections reveals exclusive thylakoid labeling with a distribution pattern consistent with other PSII proteins (80% grana, 20% stroma). These data strongly indicate that PPO is at least peripherally associated with the PSII complex.  相似文献

The inactivation of simian rotavirus SA-11 and human rotavirus type 2 (Wa) by chlorine was compared at 4 degrees C by using single-particle virus stocks. Both virus types were usually more readily inactivated at pH 6.0 than at pH 8.0 when low chlorine concentrations (0.05 to 0.2 mg/liter) were used. A complete (5 log) reduction of both was obtained within 20 s at all pH levels when chlorine concentrations were increased to 0.3 mg/liter. Slight differences in the chlorine sensitivities of SA-11 and human rotavirus type 2 were noted but were not considered to be significant.  相似文献

Hyla chrysoscelis (2n=24) and H. versicolor (2n=48) are a diploid-tetraploid species pair of treefrogs. Restriction endonuclease mapping of ribosomal RNA (rRNA) gene repeat units of diploids collected from eastern and western populations reveals no differences within rRNA gene coding regions but distinctive differences within the nontranscribed spacers. A minimum of two physical maps is required to construct an rRNA gene map for the tetraploid, whose repeat units appear to be a composite, with about 50% of the elements resembling the western diploid population and about 50% resembling the eastern population. These results imply that this population of the tetraploid species may have arisen from a genetically hybrid diploid. Alternatively, the dual level of sequence heterogeneity in H. versicolor may reflect some type of gene flow between the two species. The coding region of the rRNA genes in the tetraploid differs from that in either diploid in about 20% of all repeat units, as exemplified by a BamHI site located near the 5 terminus of the 28 S rRNA gene. If the 20% variant class of 28 S rRNA gene coding sequences is expressed, then there must be two structural classes of ribosomes; if only the 80% sequence class is expressed, then a genetic control mechanism must be capable of distinguishing between the two different sequence variants. It is postulated that the 20% variant sequence class may be correlated with a partial functional diploidization of rRNA genes in the tetraploid species.This research was supported, in part, by NSF Grants CDP-8002341 and PRM-8106947 and by faculty research grants from Miami University to J.C.V.  相似文献

Morphometric, electrophoretic, and immunological procedures were used to probe the structural and physiological differences between triazine-resistant (R) and susceptible (S) isolines of canola (Brassica napus L.). The R biotype exhibited increased grana stacking and decreased amounts of starch compared to the S biotype. Likewise, characters associated with an increase in grana stacking (lower chlorophyll a/b ratio, increased chlorophyll a/b light-harvesting complex, and relatively lower amounts of the P700 chlorophyll a protein and chloroplast coupling factor) were all observed in the R isoline of canola. Proteins which occur with approximately equal frequency in grana and stroma lamellae (plastocyanin, cutochrome f) or present only in the stroma (ribulose 1,5-bisphosphate carboxylase/oxygenase) were not quantitatively different in the two biotypes. Gross anatomical parameters (volume of epidermis, palisade mesophyll, spongy mesophyll, and air space) were similar in the two isolines. Thus, the triazine-resistance mutation does not confer a shade-type anatomy despite the chloroplast changes that are characteristic of shade biotypes or shade adaptions. These data indicate that the differences in chloroplast structure noted previously in comparisons of nonisonuclear R and S weed biotypes reflect differences in the triazineresistance factor rather than characters unrelated to triazine resistance.  相似文献

Evidence for photosynthetic linear electron transport in guard cells was obtained with two sensitive methods of high spacial resolution. Light-dependent diaminobenzidine oxidation (an indicator of PSI) and DCMU-sensitive, light-dependent thiocarbamyl nitroblue tetrazolium reduction (an indicator of PSII) were observed in guard cell plastids of Hordeum vulgare L. cv Himalaya using electron microscopic cytochemical procedures. DCMU-sensitive Chl a fluorescence induction (an indicator of PSII) was detected in individual guard cell pairs of Vicia faba L. cv Longpod using an ultramicrofluorometer. At least for these species, we conclude these results are proof for the presence of PSII in guard cell chloroplasts, which until now has been somewhat controversial.  相似文献
