Water influx accompanying the swelling of embryos during normal development of horseshoe crabs, Limulus polyphemus and Tachypleus tridentatus , following a rupture of the chorion, was analyzed. The increase in volume of perivitelline fluid during deveopment was about 90 percent of the increase in total embryo weight. Considerable water discharge was observed on drying the embryos in air and a reversible water influx occurred with a second immersion in sea water, even though the embryos died as a result of this treatment. Since the osmotic pressure of the perivitelline fluids decreased markedly during development until the end of swelling, a close correlation between swelling and osmotic pressure was recognized. These results indicate that certain osmoactive substances may be produced in the perivitelline fluid at the initial stage of swelling.  相似文献

Several axenic strains of pathogenic and nonpathogenic Entamoeba histolytica were tested for their capacity to digest native radioactive type I collagen gels and to produce liver abscesses when injected into the liver of newborn hamsters. The results demonstrate that the pathogenic strains of amebas (HM1:IMSS, HM3:IMSS, HM38:IMSS, and HK9) have a collagenolytic activity that closely correlates with their in vivo capacity to produce liver lesions. The nonpathogenic isolate (Laredo) did not show collagenolytic activity and failed to produce lesions in the liver of newborn hamsters. The results also demonstrate that type I collagen obtained from rodents and cats is degraded less by amebic collagenase than is bovine collagen, which is similar to human collagen. These findings suggest that species susceptibility to invasive infection may depend, among other factors, on the characteristics of the extracellular components of host tissues.  相似文献

  2. The optimum concentration of 2,4-D for elongationinductionwas about 1 µg/ml, both for unpeeled and peeledsegments.
  3. Using stress-relaxation analysis, mechanical propertiesof thecell wall were expressed by the parameters 1/1, To andTm. Unpeeledsegments were first treated with 2,4-D, then theepidermis waspeeled off. Parameters of the epidermal cell wallwere conspicuouslychanged by 2,4-D but those of the inner tissuewere not.
  4. Actinomycin D and cycloheximide inhibited 2,4-D-inducedchangesin cell wall parameters, as well as in elongation, ofunpeeledsegments apd of the epidermis.
  5. 2,4-D did not induceelongation of the isolated epidermis butpromoted that of peeledsegments. This promotion was smalleras compared with unpeeledsegments. 2,4-D did not significantlyinfluence the diffusionpressure deficit of peeled segmentsbut did increase their elasticand plastic extensibilities.
  6. We conclude that auxin primarilyinduces cell wall looseningof the epidermis, most likely throughnucleic acid and proteinsynthesis.

Gene co‐expression analysis has emerged in the past 5 years as a powerful tool for gene function prediction. In essence, co‐expression analysis asks the question ‘what are the genes that are co‐expressed, that is, those that show similar expression profiles across many experiments, with my gene of interest?’. Genes that are highly co‐expressed may be involved in the biological process or processes of the query gene. This review describes the tools that are available for performing such analyses, how each of these perform, and also discusses statistical issues including how normalization of gene expression data can influence co‐expression results, calculation of co‐expression scores and P values, and the influence of data sets used for co‐expression analysis. Finally, examples from the literature will be presented, wherein co‐expression has been used to corroborate and discover various aspects of plant biology.  相似文献

It is well known that the motility of spermatozoa in rainbow trout is suppressed by K+. We showed here that although trout sperm are completely immotile in medium containing 5 mM K+, motility was initiated by the subsequent addition of several mM Ca2+, suggesting that both K+and Ca2+are related to the process of the initiation of sperm motility. It was further found that K+channel blockers tetraethylammonium, nonyltriethylammonium, Ba2+and Cs+, as well as the Ca2+channel blocker verapamil, inhibited the initiation of sperm motility at doses at which these reagents inhibit chnnel-related functions in other cells. However, Na+channel blocker, tetrodotoxin and anion channel blocker 4, 4-diisothiocyatatostilbene-2, 2'-disulfonic acid inhibited the motility only at extremely high doses. These results suggest that transport of K+and Ca2+through ion channels at the plasma membrane of spermatozoa is the first event that triggers the initiation of sperm motility in rainbow trout.  相似文献

Leaf discs of Perilla frutescens var. crispa f. viridi-crispawere cultured on a defined medium to investigate factors influencingbud and root formation, callus induction, somatic embryogenesis,and floral bud formation. Addition of naphthalene-acetic acid(NAA) to the culture medium caused compact callus whereas 2,4-dichlorophenoxyacetic acid (2,4-D) promoted soft and friable callus formationon the surface of the explants. Benzyladenine, when appliedwith auxin, suppressed callus and root formation. Somatic embryogenesisoccurred, when the explants were first grown on nutrient mediumcontaining 2,4-D and organic elements, and then transferredto the 2,4-D free medium. Treatments with cytokinins, N-phenyl-N'-(4-pyridyl)urea and its derivatives induced bud formation. A low concentrationof NAA and naphthoxy-acetic acid promoted bud development. Occasionalfloral bud formation was observed depending on the originalleaf positions on mother plants from which the leaf discs wereexcised. A gradient of floral bud forming capacity along thestem was noted. In in vitro cultured stem segments of Torenia fournieri Lind.,the formation of adventitious buds can be induced when the culturemedium contains cytokinin. When long stem segments (2.0 cm ormore) were cultured with cytokinin, a large number of buds wereformed in the marginal regions, namely, within the limits of0.5 cm from the cut ends of explants, while only a few budswere initiated in the middle part of the explants. If a slightinjury was made transversely with a scalpel in the central partof an explant, a significant increase in the number of budswas noted within the limits of 0.5 cm from the wound site. Whena wounding treatment was given lengthwise to an explant, a largenumber of adventitious buds were formed over the entire surfaceof the explant compared to the control. Excision itself of explantsfrom mother plants and the additional wounding given to theexplants seemed to trigger the induction of adventitious buddifferentiation in Torenia stem segments. These wounding treatmentsdid not affect the uptake into explants and/or the distributionpattern of radioactive benzyladenine applied to the culturemedium. A cell wall fraction (pectic substances) of oat coleoptile segmentsfed with 14C-glucose contained more radioactivity under theeffect of auxin than did the control. When labeled segmentswere grown for 6 hr in auxin or glucanase solution the labelin the hemicellulose fraction decreased as growth increased.ß-1,3-Glucanase prepared from the culture of a fungus,Sclerotinia libertiana, induces elongation of segments of thepea stem and the oat coleoptile. Traces of cellulase and pectinmethylesterase contaminating the enzyme preparation are notresponsible for the stimulatory effect. Cellulase seemed tobe rather inhibitory and pectin methylesterase showed only aslight effect on coleoptile elongation. A possible relationshipbetween the metabolic turnover of hemicellulosic polysaccharideand cell wall extension is suggested. (Received February 5, 1968; )  相似文献
