We tested the hypothesis that glucocorticoids attenuate changes in arterial pressure and renal sympathetic nerve activity (RSNA) in response to activation and blockade of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors within the nucleus of the solitary tract (NTS). Experiments were performed in Inactin-anesthetized male Sprague-Dawley rats treated for 7 +/- 1 days with a subcutaneous corticosterone (Cort) pellet or in control rats. Baseline mean arterial pressure (MAP) was significantly higher in Cort-treated rats (109 +/- 2 mmHg, n = 39) than in control rats (101 +/- 1 mmHg, n = 48, P < 0.05). In control rats, microinjection of AMPA (0.03, 0.1, and 0.3 pmol/100 nl) into the NTS significantly decreased MAP at all doses and decreased RSNA at 0.1 and 0.3 pmol/100 nl. Responses to AMPA in Cort-treated rats were attenuated at all doses of AMPA (P < 0.05). Responses to the AMPA-kainate receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) were also significantly reduced in Cort-treated rats relative to control rats. Blockade of glucocorticoid type II receptors with mifepristone significantly enhanced responses to CNQX in both control and Cort rats. We conclude that glucocorticoids attenuate MAP and RSNA responses to activation and blockade of AMPA receptors in the NTS.  相似文献

We deployed sediment traps adjacent to two active hydrothermal vents at 9°50'N on the East Pacific Rise (EPR) to assess the variability in bacterial community structure associated with plume particles on the timescale of weeks to months, to determine whether an endemic population of plume microbes exists, and to establish ecological relationships between bacterial populations and vent chemistry. Automated rRNA intergenic spacer analysis (ARISA) indicated that there are separate communities at the two different vents and temporal community variations between each vent. Correlation analysis between chemistry and microbiology indicated that shifts in the coarse particulate (>1 mm) Fe/(Fe+Mn+Al), Cu, V, Ca, Al, (232) Th, and Ti as well as fine-grained particulate (<1 mm) Fe/(Fe+Mn+Al), Fe, Ca, and Co are reflected in shifts in microbial populations. 16S rRNA clone libraries from each trap at three time points revealed a high percentage of Epsilonproteobacteria clones and hyperthermophilic Aquificae. There is a shift toward the end of the experiment to more Gammaproteobacteria and Alphaproteobacteria, many of whom likely participate in Fe and S cycling. The particle-attached plume environment is genetically distinct from the surrounding seawater. While work to date in hydrothermal environments has focused on determining the microbial communities on hydrothermal chimneys and the basaltic lavas that form the surrounding seafloor, little comparable data exist on the plume environment that physically and chemically connects them. By employing sediment traps for a time-series approach to sampling, we show that bacterial community composition on plume particles changes on timescales much shorter than previously known.  相似文献

Many organisms reproduce in temporary aggregations where estimates of colony size can be made by direct counts. When individuals are not synchronous, however, early breeders depart before the last arrive, so counts underestimate the total breeding population. We present a model describing a colony's census as a function of arrival, breeding tenure, and the correlation between them, and we use it to illustrate how variance in arrival and tenure affect the census. Counts of breeding female northern elephant seals ( Mirounga angustirostris ) from 1975 to 2007 were used to test the model. Four of the model's parameters—population size, mean and variance of arrival date, and the correlation between arrival date and breeding tenure—could be estimated from census data using a Bayesian approach; prior estimates of two other parameters—mean tenure and its variance—had to be used to avoid overparameterization. The model's predictions fit observed censuses well and produced reliable estimates of population size and arrival behavior, showing that the maximum census was 8%–16% below the total number of breeding females. This method could be used for estimating abundance in any asynchronous aggregation, given independent information on one of the defining distributions: arrival, tenure, or departure.  相似文献

Summary The absorption and subsequent transport of foliar applied Rb86 labeled Rb Cl (10 mM) was studied on bean plants (Phaseolus vulgaris, L. cv. Black Seeded Blue Lake) exposed to physiologically tolerable levels of certain plant growth substances in the solution culture root media. Gibberellin A3 (10-5 M) increased Rb uptake but did not affect total translocation from the treated leaf. Translocation was directed to the upper vegetative parts and markedly reduced to the roots. Foliar influx of Rb and transport to the roots were greatly enhanced by 1-naphthaleneacetic acid (10-6 M) but mobilization of Rb into the leaves and upper stem was reduced. 2-Chloroethyltrimethylammonium chloride (10-3 M) and N,N-dimethylaminosuccinamic acid (3×10-4 M) decreased the mobility of Rb to the upper stem, increased it to the roots, and had no effect on initial uptake. Rb absorption was depressed by 2,4-dichlorobenzyltributylphosphonium chloride (10-5 M) with no effect on subsequent translocation. Both uptake and mobility were strikingly inhibited by N6-benzyladenine (10-6 M).These results suggest that absorption and the subsequent transport of foliar absorbed Rb are independent processes and that the distribution or mobilization of Rb in the various plant organs was not always a function of the chemically modified growth rate of the corresponding organ.Report No. COO-888-50 in cooperation with the Division of Biology and Medicine of the United States Atomic Energy Commission, Contract No. AT (11-1)-888. Centrifuged, unfertilized eggs of the sea urchin, Arbacia punctulata, have been studied with the electron microscope. Subcellular particles were stratified by centrifuging living cells, known to be normally fertilizable, for five minutes at 3,000 g. The layered subcellular particles, including cortical granules, 16 mµ RNP particles, pigment, yolk, mitochondria, and oil droplets, possess characteristic ultrastructural features by which they may be identified in situ. The clear zone contains 16 mµ particles, most of them freely dispersed, scattered mitochondria, and a few composite structures made up of annulate lamellae in parallel layers or in association with dense, spherical aggregates of the RNP particles. Free 16 mµ particles are found, in addition, throughout the cell, in the interstices between the stratified larger particles. They show a tendency to form ramifying aggregates resulting from certain types of injury to the cell. A few vesicular structures, found mainly in the clear zone, have attached RNP particles, and appear to be related to the ER of tissue cells. Other vesicles, bounded by smooth membranes, are found throughout the cell. These are extremely variable in size, number, and distribution; their total number appears to depend upon conditions of fixation. It is suggested that limited formation of such structures is a normal property of the ground cytoplasm in this cell, but that fixed cells with very large numbers of smooth surfaced vesicles have produced the latter as a response to chemical injury. A model of the ground cytoplasm is proposed whose aim is to reconcile the rheological behavior of the living cell with the ultrastructural features observed.  相似文献

Glucocorticoid receptors (GRs) are present at a high density in the nucleus of the solitary tract (NTS), an area of the dorsal hindbrain (DHB) that is critical for blood pressure regulation. However, whether these receptors play any role in the regulation of blood pressure is unknown. We tested the hypothesis that glucocorticoids act in the DHB to increase arterial pressure using two experimental strategies. In one approach, we implanted pellets of corticosterone (Cort) or sham pellets onto the DHB over the NTS. Compared with rats with sham pellets, rats with DHB Cort pellets had an increased (P < 0.05) mean arterial pressure (111 +/- 2 vs. 104 +/- 1 mmHg) and heart rate (355 +/- 9 vs. 326 +/- 5 beats/min) after 4 days. In the second approach, we implanted subcutaneous Cort pellets to increase the systemic Cort concentration and then subsequently implanted pellets of the GR antagonist mifepristone (Mif; previously RU-38486) or sham pellets onto the DHB. Two days of DHB Mif treatment reduced (P < 0.05) mean arterial pressure in those rats with elevated plasma Cort levels (118 +/- 2 vs. 108 +/- 1 mmHg for sham vs. Mif DHB pellets). Cort and Mif pellets placed on the dura had no effects on arterial pressure or heart rate, ruling out systemic cardiovascular effects of the steroids. DHB Cort treatment had no effects on plasma Cort concentration or adrenal weight, indicating that the contents of the DHB Cort pellet did not diffuse into the systemic circulation or into the forebrain areas that regulate plasma Cort concentration in concentrations sufficient to produce physiological effects. Immunohistochemistry for the occupied GRs demonstrated that the Cort and Mif from the DHB pellets were delivered to the DHB with minimal diffusion to the ventral hindbrain or forebrain. We conclude that glucocorticoids act in the DHB to increase arterial pressure.  相似文献
