The glucan-binding lectin (GBL) ofStreptococcus sobrinus is cell associated, enabling the bacteria to be aggregated by -1,6 glucans. Glucans, such as amylose, pullulan, laminarin and nigeran, have no affinity for the lectin. High molecular weight -1,6 glucans (dextrans) readily aggregate the bacteria, whereas low molecular weight glucans inhibit the aggregation brought about by the high molecular weight species. Methylated glucan T-2000 (an -1,6 glucan with an average molecular weight of 2 × 106 Da) aggregated the bacteria very poorly when the extent of methylation (DS, or degree of substitution) was high, and less poorly when the DS was low. Similarly, methylated low molecular weight -1,6 glucan was a poor inhibitor of aggregation induced by the high molecular weight glucan T-2000. Because the methylation occurred primarily on the hydroxyl of C-2, it is suggested that the hydroxyl is needed for formation of the lectin-glucan complex. It appears that the GBL is not only stereospecific in interaction with glucans, but also regiospecific, interacting only with the underivatized -1,6-glucan.  相似文献

The Eurasian gall-forming weevil Ceutorhynchus cardariae Korotyeav (Coleoptera: Curculionidae) is a biological control candidate for the invasive Eurasian Lepidium draba L. (Brassicaceae) in the western USA. Among 157 nontarget plant species that have been tested, some North American Caulanthus and Streptanthus species, confamilial with Lepidium, were found to be at potential risk of attack by C. cardariae. Many Caulanthus and Streptanthus species grow on serpentine soils, which are characterized by low nutrient content and high concentrations of various combinations of heavy metals. Some of these species accumulate heavy metals, which have been shown to act as deterrents against insect herbivory. Standard pre-release host specificity tests with C. cardariae used plants propagated on horticultural soils, which could have inflated performance by C. cardariae on Caulanthus and Streptanthus species. To examine this possibility, we assessed the performance of C. cardariae on three Caulanthus species, the federally listed threatened and endangered Streptanthus glandulosus ssp. albidus, and Lepidium draba, on plants propagated in horticultural soil or in native serpentine soil. Our study showed that native serpentine soil influenced C. cardariae attack. All plant species, including L. draba, received less feeding damage and gall formation when grown in serpentine soil. In addition, feeding by C. cardariae was much less and fewer galls were formed on the confamilial species than on L. draba, regardless of soil type. Our data show that native confamilial species restricted to specialized soil types may be at less risk of herbivore attack than predicted based on tests conducted in horticultural soil.

Previously, we demonstrated that treatment of monocytic cells with IFN-gamma causes release of ribosomal protein L13a from the 60S ribosome and subsequent translational silencing of Ceruloplasmin (Cp) mRNA. Here, evidence using cultured cells demonstrates that Cp mRNA silencing is dependent on L13a and that L13a-deficient ribosomes are competent for global translational activity. Human monocytic U937 cells were stably transfected with two different shRNA sequences for L13a and clonally selected for more than 98% abrogation of total L13a expression. Metabolic labeling of these cells showed rescue of Cp translation from the IFN-gamma mediated translational silencing activity. Depletion of L13a caused significant reduction of methylation of ribosomal RNA and of cap-independent translation mediated by Internal Ribosome Entry Site (IRES) elements derived from p27, p53, and SNAT2 mRNAs. However, no significant differences in the ribosomal RNA processing, polysome formation, global translational activity, translational fidelity, and cell proliferation were observed between L13a-deficient and wild-type control cells. These results support the notion that ribosome can serve as a depot for releasable translation-regulatory factors unrelated to its basal polypeptide synthetic function. Unlike mammalian cells, the L13a homolog in yeast is indispensable for growth. Thus, L13a may have evolved from an essential ribosomal protein in lower eukaryotes to having a role as a dispensable extra-ribosomal function in higher eukaryotes.  相似文献

Oxidative stress is proposed to play a pivotal role in the development of hepatocellular carcinoma. With an accelerated metabolism cancer cells demand high reactive species accumulation to maintain their indiscriminate cell growth and proliferation. Here we wanted to see the status of reactive species in the chemically induced liver cancer. For this purpose swiss albino mice were exposed to DEN and CCl4 to develop an in vivo model of hepatocarcinoma. Depletion of cellular antioxidants regulated accretion of reactive species during the development of DEN + CCl4 induced tumor formation in hepatocytes. Currently available therapeutics for heptatocellular carcinoma is costly and coupled with certain bystander effects to the surrounding control cells. Therefore considering the antioxidant properties of SAC and berberine we treated DEN + CCl4 exposed mice after the development of liver tumor. Results effectively pointed out the usefulness of the alternative treatment with SAC and berberine in hepatoprotection. Replenishment of both enzymatic and non enzymatic antioxidant efficiently reduced accumulation of reactive species and that eventually closely associated with effective reduction in tumor number and size after drug treatment in DEN + CCl4 exposed mice.  相似文献

Protein syntheses mediated by cellular and viral internal ribosome entry sites (IRESs) are believed to have many features in common. Distinct mechanisms for ribosome recruitment and preinitiation complex assembly between the two processes have not been identified thus far. Here we show that the methylation status of rRNA differentially influenced the mechanism of 80S complex formation on IRES elements from the cellular sodium-coupled neutral amino acid transporter 2 (SNAT2) versus the hepatitis C virus mRNA. Translation initiation involves the assembly of the 48S preinitiation complex, followed by joining of the 60S ribosomal subunit and formation of the 80S complex. Abrogation of rRNA methylation did not affect the 48S complex but resulted in impairment of 80S complex assembly on the cellular, but not the viral, IRESs tested. Impairment of 80S complex assembly on the amino acid transporter SNAT2 IRES was rescued by purified 60S subunits containing fully methylated rRNA. We found that rRNA methylation did not affect the activity of any of the viral IRESs tested but affected the activity of numerous cellular IRESs. This work reveals a novel mechanism operating on a cohort of cellular IRESs that involves rRNA methylation for proper 80S complex assembly and efficient translation initiation.  相似文献

The frequency of dengue virus (DENV) infection has increased dramatically in the last few decades, and the lack of a vaccine has led to significant morbidity and mortality worldwide. To date, a convenient murine system to study human T cell responses to DENV has not been available. Mice transgenic for HLA are widely used to model human immune responses, and it has been shown that mouse-passaged DENV is able to replicate to significant levels in IFN-α/βR(-/-) mice. To cover a wide range of HLA phenotypes, we backcrossed IFN-α/βR(-/-) mice with HLA A*0201, A*0101, A*1101, B*0702, and DRB1*0101-transgenic mice. A DENV proteome-wide screen identified a total of 42 epitopes across all HLA-transgenic IFN-α/βR(-/-) strains tested. In contrast, only eight of these elicited responses in the corresponding IFN-α/βR(+/+) mice. We were able to identify T cell epitopes from 9 out of the 10 DENV proteins. However, the majority of responses were derived from the highly conserved nonstructural proteins NS3 and NS5. The relevance of this model is further demonstrated by the fact that most of the epitopes identified in our murine system are also recognized by PBMC from DENV-exposed human donors, and a dominance of HLA B*0702-restricted responses has been detected in both systems. Our results provide new insights into HLA-restricted T cell responses against DENV, and we describe in this study a novel murine model that allows the investigation of T cell-mediated immune mechanisms relevant to vaccine design.  相似文献

In this study a novel bone-targeting agent containing elements of the tricarbonylmethane system of ring A of tetracycline was developed and was shown to bind to the mineral constituent of bone, hydroxyapatite. Conjugation of this bone-targeting agent to estradiol resulted in a bone-targeted estrogen (BTE2-A1) with an enhanced ability to bind to hydroxyapatite. In an ovariectomized rat model of osteoporosis a partial separation of the skeletal effects of estradiol from the uterine effects was observed following subcutaneous administration of BTE2-A1. This novel bone-targeting estradiol delivery system has the potential to improve the safety profile of estradiol in the treatment of osteoporosis.  相似文献
