We describe a computer tool to aid the discovery of new motifsin nucleic acid sequences. A typical use would be to analysea set of upstream regions from a family of related genes inorder to find possible control sequences. The heart of the methodis the creation of dictionaries of related subsequences. Thesedictionaries can then be analysed to look for the commonestor best-defined subsequences, those that occur in the highestnumber of different sequences, or for those in equivalent positionswithin the family. Aqueous extracts of smoke, derived from Themeda triandra, a fire-climax grass, and Passerina vulgaris, a fynbos plant, stimulated the growth of primary root sections of tomato roots in suspension culture. The optimal dilution for both extracts was 1:2000. Several of the fractions obtained from TLC separation of the Themeda and the Passerina extracts significantly promoted primary root growth. The auxins naphthaleneacetic acid (NAA), indolebutyric acid (IBA) and indoleacetic acid (IAA) were found to stimulate the growth of the primary root axis, with IAA and NAA significantly promoting lateral root number. Similarly, the naturally occurring cytokinins, zeatin and its derivatives (zeatin-O-glucoside; dihydrozeatin and zeatin riboside) stimulated primary root length. Zeatin and dihydrozeatin promoted secondary root growth, but only at very low concentrations.  相似文献

Decapitated tomato plants were supplied via the roots with [8-14C]-kinetin or [8-14C]-benzyladenine in a nutrient solution for a period of 24 h. After this time the root material, the root sap produced during the 24 h period and the nutrient solution remaining at the end of the experiments were analysed for cytokinins. HPLC techniques and chemical treatments were used to tentatively identify radioactive metabolites formed. Uptake of kinetin and benzyladenine by the roots was found to be limited but once within the root tissues metabolism was both rapid and extensive.At least 14 metabolites of kinetin were recovered from root tissue and root sap. Many of these appeared to be degradation products. There was, however, some evidence of formation of zeatin-like derivatives. Side-chain cleavage of the original kinetin which occurs rapidly is suggested as a possible route for the eventual production of these endogenous cytokinin forms.The benzyladenine taken up by the roots was apparently both ribosylated and glucosylated. No unmetabolized benzyladenine was detected in the root tissues after 24 h. Only very low levels of radioactivity were associated with the retention time of adenine, suggesting that in the case of benzyladenine side-chain cleavage is of limited importance.The significance of these reactions in relation to the potential use of cytokinins in the regulation of plant growth is discussed.  相似文献

The effects of the seaweed concentrate “Kelpak’ on the growth and yield of wheat grown under conditions of varying K supply were investigated. Kelpak had no significant effect on the yield of wheat receiving an adequate K supply, but significantly increased the yield of K stressed plants. The increase in yield was caused by an increase in both grain number and individual grain weight. Although the beneficial effects of seaweed concentrates have often been attributed to their cytokinin content, several lines of evidence suggested that this group of plant growth regulators may not be solely responsible for the observed effects of Kelpak on wheat. Irrespective of the physiological mechanism of action, Kelpak would appear to have considerable potential for increasing yield in K stressed wheat and may therefore reduce the requirement of wheat for K fertilization.  相似文献

The effects of the seaweed concentrate Kelpak on the growth and mineral nutrition of lettuce plants grown under conditions of varying nutrient supply were investigated. Kelpak significantly increased the yield and the concentration and amounts of Ca, K and Mg in the leaves of lettuce receiving an adequate supply of nutrients, but had little effect on nutrient stressed plants. Summary The development of the testa was studied inErythrina lysistemon using both light and electron microscopy. Cells of the outer epidermis of the outer integument divide anticlinally and undergo radial elongation to form a palisade layer. The outer tangential walls are thickened at an early stage, and deposition of fluted thickenings on the radial walls occurs at maturity. Palisade cells in the hilar region differentiate from sub-funicular tissue, and at maturity the outer ends of the cells undergo extensive deposition of secondary walls and associated lignification. The light line occurs at the junction between the outer, thickened portions of the cells and the inner, less thickened portions. An electron-translucent (suberised) cap develops in the outer tangential walls of the palisade cells at a late stage. Microtubules and dictyosomes are closely associated with the developing thickenings in palisade and tracheid bar, and the microtubules run parallel to the wall microfibrils. Differentiation of the tracheid bar coincides with final secondary wall deposition and lignification in the hilar palisade. The cells of the tracheid bar are dead at maturity, but are surrounded by sheaths of elongate parenchyma.  相似文献

Irrespective of their age, leaves of Ginkgo biloba metabolised applied 8 (14C) zeatin to compounds of similar chromatographic properties. Glucosylation is apparently not a normal feature of cytokinin metabolism in immature leaves. However, the application of zeatin to these leaves did result in the formation of metabolites which co-chromatographed with glucosylated cytokinins. As far as cytokinin metabolism is concerned therefore, this application of excess zeatin allowed immature leaves to behave as mature or senescing leaves. Overall metabolism was fastest in immature leaves. From the metabolites formed it would appear as if oxidation, which resulted in the formation of a metabolite which co-eluted with N-(purin-6-yl)glycine, was also important in immature leaves. In senescing leaves glucosylation was the major form of metabolism. Extraction and re-application of the polar metabolites (formed from zeatin) to mature leaves resulted in the formation of compounds which co-chromatographed with zeatin. This suggests that these compounds could serve as precursors for zeatin or could be hydrolysed to form zeatin.Very little of the applied radioactivity was exported from the leaves irrespective of their physiological age. When the metabolites, obtained after zeatin application to mature leaves, were extracted and reapplied to the leaves, export of radioactive material was much improved. The results suggest that should cytokinins such as zeatin be translocated to mature leaves of this deciduous gymnosperm their export from the leaves would be unlikely unless first metabolised. Soybean callus metabolised applied 6-furfurylamino (8-14C) purine very rapidly to polar compounds, some of which were retained on a Dowex 50 cation exchange resin, and were unaffected by alkaline phosphatase; while others were apparently phosphorylated and were detected in the aqueous fraction. Adenine was detected as an intermediate and it can be concluded that it was formed as a result of the rapid and efficient removal of the furfuryl side chain. The formed adenine was rapidly incorporated into the polar metabolites. 6-(2,3,4-Trihydroxy-3-methylbutylamino) purine, trivial name trihydroxy-zeatin (THZ), an oxidation product of zeatin was applied via the epidermis or cut end of Avena leaf segments. THZ did not retard senescence of the segments in either case. With epidermal application THZ remained unmetabolised while the cut end method of application resulted in its metabolism to more polar THZ metabolites of unknown identity. The Avena leaf tissue did not apparently cleave the isoprenoid side chain of the THZ molecule.  相似文献
