Methods are presented for modeling dose-related effects in proportion data when extra-binomial variability is a concern. Motivation is taken from experiments in developmental toxicology, where similarity among conceptuses within a litter leads to intralitter correlations and to overdispersion in the observed proportions. Appeal is made to the well-known beta-binomial distribution to represent the overdispersion. From this, an exponential function of the linear predictor is used to model the dose-response relationship. The specification was introduced previously for econometric applications by Heckman and Willis; it induces a form of logistic regression for the mean response, together with a reciprocal biexponential model for the intralitter correlation. Large-sample, likelihood-based methods for estimating and testing the joint proportion-correlation response are studied. A developmental toxicity data set illustrates the methods.  相似文献

Protein serine/threonine kinase casein kinase 2 (CK2) is a key player in cell growth and proliferation but is also a potent suppressor of apoptosis. CK2 has been found to be dysregulated in all the cancers that have been examined, including prostate cancer. Investigations of CK2 signaling in the prostate were originally initiated in this laboratory, and these studies have identified significant functional activities of CK2 in relation to normal prostate growth and to the pathobiology of androgen-dependent and -independent prostate cancer. We present a brief overview of these developments in the context of prostate biology. An important outcome of these studies is the emerging concept that CK2 can be effectively targeted for cancer therapy.  相似文献

Background

Protein translocation across the membrane of the Endoplasmic Reticulum (ER) is the first step in the biogenesis of secretory and membrane proteins. Proteins enter the ER by the Sec61 translocon, a proteinaceous channel composed of three subunits, α, β and γ. While it is known that Sec61α forms the actual channel, the function of the other two subunits remains to be characterized.

Results

In the present study we have investigated the function of Sec61β in Drosophila melanogaster. We describe its role in the plasma membrane traffic of Gurken, the ligand for the Epidermal Growth Factor (EGF) receptor in the oocyte. Germline clones of the mutant allele of Sec61β show normal translocation of Gurken into the ER and transport to the Golgi complex, but further traffic to the plasma membrane is impeded. The defect in plasma membrane traffic due to absence of Sec61β is specific for Gurken and is not due to a general trafficking defect.

Conclusion

Based on our study we conclude that Sec61β, which is part of the ER protein translocation channel affects a post-ER step during Gurken trafficking to the plasma membrane. We propose an additional role of Sec61β beyond protein translocation into the ER.  相似文献

Epidermal growth factor (EGF) mimicked the effect of insulin to activate glycogen synthase and stimulate glycogen synthesis in isolated rat hepatocytes. Both agents required glucose (greater than 5 mM) and had similar time courses of action. The maximum effect of EGF was approx. 70% of that of insulin, and the half-maximally effective concentrations were 9 nM and 4 nM respectively. Combinations of the two agents produced additive responses. EGF also resembled insulin in its ability to inhibit the effects of 0.1-1.0 nM-glucagon on cyclic AMP and glycogen phosphorylase in hepatocytes. The maximum effect of EGF was approx. 70% of that of insulin, and the half-maximally effective concentrations were approx. 5 nM and 0.5 nM respectively. EGF and insulin inhibited phosphorylase activation by exogenous cyclic AMP, and inhibited cyclic AMP accumulation induced by forskolin. They also inhibited phosphorylase activation provoked by phenylephrine, but not by vasopressin. EGF added alone rapidly activated phosphorylase and increased cytosolic [Ca2+], but the effects were no longer apparent at 5 min and were smaller than those of vasopressin. Insulin did not induce these changes. In hepatocytes previously incubated with myo-[3H]inositol, EGF did not significantly increase myo-inositol 1,4,5-trisphosphate. However, its ability to increase cytosolic [Ca2+] was blocked by neomycin, an inhibitor of phosphatidylinositol bisphosphate hydrolysis. It is concluded that some, but not all, of the effects of EGF in liver are strikingly similar to those exerted by insulin, suggesting that these agents may have some similar mechanisms of action in this tissue.  相似文献

We report the nucleotide sequence of a cloned cDNA, pMTS-3, that contains a 1-kb insert corresponding to mouse thymidylate synthase (E.C. 2.1.1.45). The open reading frame of 921 nucleotides from the first AUG to the termination codon specifies a protein with a molecular mass of 34,962 daltons. The predicted amino acid sequence is 90% identical with that of the human enzyme. The mouse sequence also has an extremely high degree of similarity (as much as 55% identity) with prokaryotic thymidylate synthase sequences, indicating that thymidylate synthase is among the most highly conserved proteins studied to date. The similarity is especially pronounced (as much as 80% identity) in the 44-amino-acid region encompassing the binding site for deoxyuridylic acid. The cDNA sequence also suggests that mouse thymidylate synthase mRNA lacks a 3' untranslated region, since the termination codon, UAA, is followed immediately by a poly(A) segment.   相似文献

Species worldwide have begun to shift their range boundaries in response to climate change and other anthropogenic causes, with population declines at the trailing edge of a species' range often foreshadowing future changes in core parts of the range. Therefore, we analyzed a 30-year (1991–2019) data set for the California spotted owl (Strix occidentalis occidentalis) near its southern range boundary in southern California, USA, that included the largest regional population (San Bernardino Mountains) to estimate trends in territory occupancy and reproduction. We then assessed how these demographic rates were affected by habitat, wildfire, fuel treatments, and climate. Mean occupancy declined from 0.82 to 0.39 during our study, whereas reproductive output showed no temporal trends ( young/occupied territory). Territory extinction (extirpation) rates were relatively low in territories with more large trees (≥50 cm dbh), and colonization increased strongly with large tree density for low-elevation territories within the shrub-woodland ecotype but not for higher-elevation territories within mixed-conifer forest. High-severity wildfire had an adverse effect on occupancy: territory extinction rates steadily increased with the amount of high-severity fire within an owl territory during the previous 10 years, while colonization declined to nearly zero when ≥40% of a territory burned at high-severity during the previous 10 years. The effects of high-severity fire were unlikely to be confounded with post-fire fuel treatments, which primarily consisted of the removal, burning, or scattering of brush and small trees and snags (<40.6 cm dbh) and affected much smaller areas than high-severity fire. Of the 40 territories that received fuel treatments within 10 years of a fire, only 3 of them had post-fire fuel treatments that affected >5% of the territory, whereas average area burned at high severity for all 40 territories was 17%. Fuel treatments intended to modify fire behavior and reduce the likelihood of large, high-severity fires led to increases in territory extinction and colonization such that their net effect on occupancy was minimal. Our simulations of occupancy dynamics indicated that high-severity fire accounted for 9.6% of the observed decline in occupancy, whereas fuel treatments effectively accounted for none of the decline. Spotted owl reproductive output was lower at territories where fuel treatments occurred, but low- to moderate-severity fire resulted in much larger, population-level reductions in reproductive output (141 fewer young) from 2006–2019 than treatments (19 fewer young). Thus, the benefits of fuel treatments that reduce fire occurrence and severity appear to outweigh potential short-term costs to spotted owls and their habitat. Because high-severity fire only explained a modest amount of the long-term occupancy decline and much of the decline occurred in the 1990s before large fires occurred, additional factors are likely adversely affecting the owl population and merit further study. Nevertheless, the large observed population decline, limited evidence of owl dispersal among mountain ranges in the southern California metapopulation, and negative effects of increasingly large and severe fire suggest that California spotted owls at their southern range boundary are vulnerable to extirpation. In an era of climate change, owls in the core part of the range will likely become increasingly susceptible to warmer temperatures and increased severe fire activity in the future. Thus, the restoration of historical, low-severity fire regimes through fuels management while maintaining large trees is important to improving owl persistence.  相似文献

Normal synaptonemal complexes (SCs), consisting of two lateral elements and a central element, are present in wild-type, him-4 and him-8 mutant strains in both hermaphrodites and males of Caenorhabditis elegans. Thus, the increase in rate of nondisjunction in the him mutants is not related to aberrant SC morphology. The wild-type hermaphrodite has six SCs, as determined from 3-D reconstruction analysis of serial sections from electron microscopy. Thus, n = 6 and this confirms early reports based on cytological studies with the light microscope. Only one end of the SC is attached to the nuclear envelope while the other end is free in the nucleoplasm and there is no apparent bouquet formation. Either end of the SC can attach to the nuclear envelope. The pairing behavior of the XX bivalent is normal and occurs synchronously with the autosomes. Electron dense bodies, or knobs, are associated with the SC via the central element and displace the chromatin for a distance of 200 nm. Each pachytene nucleus of the wild-type hermaphrodite has six such structures that are randomly dispersed along the bivalents such that some SCs have one or two knobs while others have none. The relationship between near-infrared reflectance at 800 nm (NIRR) from leaves and characteristics of leaf structure known to affect photosynthesis was investigated in 48 species of alpine angiosperms. This wavelength was selected to discriminate the effects of leaf structure vs. chemical or water content on leaf reflectance. A quantitative model was first constructed correlating NIRR with leaf structural characteristics for six species, and then validated using all 48 species. Among the structural characteristics tested in the reflectance model were leaf trichome density, the presence or absence of both leaf bicoloration and a thick leaf cuticle (>1 μm), leaf thickness, the ratio of palisade mesophyll to spongy mesophyll thickness (PM/SM), the proportion of the mesophyll occupied by intercellular air spaces (%IAS), and the ratio of mesophyll cell surface area exposed to IAS (A(mes)) per unit leaf surface area (A), or A(mes)/A. Multiple regression analysis showed that measured NIRR was highly correlated with A(mes)/A, leaf bicoloration, and the presence of a thick leaf cuticle (r = 0.93). In contrast, correlations between NIRR and leaf trichome density, leaf thickness, the PM/SM ratio, or %IAS were relatively weak (r < 0.25). A model incorporating A(mes)/A, leaf bicoloration, and cuticle thickness predicted NIRR accurately for 48 species (r = 0.43; P < 0.01) and may be useful for linking remotely sensed data to plant structure and function.  相似文献
