蔗糖转化酶(invertase, INV)在植物生长发育和抵御胁迫中发挥着重要作用。研究从葡萄基因组数据库中鉴定出19个蔗糖转化酶基因,对基因结构和编码蛋白质的理化性质进行生物信息学分析,并利用qRT-PCR技术分析基因在不同激素和非生物胁迫条件下的表达特征,为进一步探索葡萄INV基因家族参与葡萄逆境响应提供了一定的理论依据。结果表明,(1)该基因家族编码蛋白的氨基酸长度在150~766 aa之间,理论等电点介于4.43~9.1之间,亚细胞定位预测发现其主要在细胞质中表达,此外液泡和细胞壁也存在部分基因表达;(2)共线性结果显示VvCINV与其他5个物种复制频率较高;(3)保守基序分析表明VvCwINV包含了所有的保守基序,且Glyco_32和Glyco_hydro_100是VvINV基因主要结构域;(4)组织特异性表达分析发现多数基因在葡萄生长发育进程中都有表达;(5)qRT-PCR分析结果显示,VvINV基因家族在叶片中对激素处理和非生物胁迫的响应出现上调,VvCINV1在50 mg/L GA3和10%PEG处理后上调表达极显著,VvCINV4在盐胁迫、ABA...  相似文献

Tissue homeostasis of skin is sustained by epidermal progenitor cells localized within the basal layer of the skin epithelium. Post‐translational modification of the proteome, such as protein phosphorylation, plays a fundamental role in the regulation of stemness and differentiation of somatic stem cells. However, it remains unclear how phosphoproteomic changes occur and contribute to epidermal differentiation. In this study, we survey the epidermal cell differentiation in a systematic manner by combining quantitative phosphoproteomics with mammalian kinome cDNA library screen. This approach identified a key signaling event, phosphorylation of a desmosome component, PKP1 (plakophilin‐1) by RIPK4 (receptor‐interacting serine–threonine kinase 4) during epidermal differentiation. With genome‐editing and mouse genetics approach, we show that loss of function of either Pkp1 or Ripk4 impairs skin differentiation and enhances epidermal carcinogenesis in vivo. Phosphorylation of PKP1's N‐terminal domain by RIPK4 is essential for their role in epidermal differentiation. Taken together, our study presents a global view of phosphoproteomic changes that occur during epidermal differentiation, and identifies RIPK‐PKP1 signaling as novel axis involved in skin stratification and tumorigenesis.  相似文献

1. While it is clear that land‐use change significantly impacts the taxonomic dimension of soil biodiversity, how the functional dimension responds to land‐use change is less well understood. 2. This study examined how the transformation of primary forests into rubber tree monocultures impacts individual termite species and how this change is reflected in termite taxonomic and functional α‐diversity (within site) and β‐diversity (among sites). 3. Overall, individual species responded strongly to land‐use change, whereby only 11 of the 27 species found were able to tolerate both habitats. These differences caused a 27% reduction in termite taxonomic richness and reduced taxonomic β‐diversity in rubber plantations compared with primary forests. The study also revealed that the forest conversion led to a shift in some termite species with smaller body size, shorter legs and smaller mandibular traits. Primary forests exhibited higher functional richness and functional β‐diversity of termite species, indicating that functional traits of termite species in rubber plantations are more evenly distributed. 4. The present study suggests that forest conversion does not merely decrease taxonomic diversity of termites, but also exerts functional trait filtering within some termite species. The results affirm the need for biodiversity assessments that combine taxonomic and functional indicators when monitoring the impact of land‐use change.  相似文献

Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor (LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+ T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+ T cells: LDLR interacts with the T-cell receptor (TCR) complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes (CTLs).Furthermore,we found that the tumor microenvironment (TME) downregulates CD8+ T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9 (PCSK9) which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+ T cells by alleviating the suppressive effect on CD8+ T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well.  相似文献

The role of Non‐POU‐domain‐containing octamer‐binding protein (NONO) in the formation and development of angiotensin II (Ang II)‐induced abdominal aortic aneurysm (AAA) in apolipoprotein E‐knockout (ApoE?/?) mice is still unknown. In Part I, the protein level of NONO was suggestively greater in the AAA tissues compare to that in the normal abdominal aortas. In Part II, 20 ApoE?/? male mice were used to examine the transfection efficiency of lentivirus by detecting GFP fluorescence. In Part III, mice were arbitrarily separated into two groups: one was the control group without Ang II infusion, and another was the Ang II group. Mice treated with Ang II were further randomly divided into three groups to receive the same volume of physiological saline (NT group), sh‐negative control lentivirus (sh‐NC group) and si‐NONO lentivirus (sh‐NONO group). NONO silencing suggestively reduced the occurrence of AAA and abdominal aortic diameter. Compare to the NT group, NONO silencing markedly augmented the content of collagen and vascular smooth muscle cells but reduced macrophage infiltration in AAA. In addition, knockdown of NONO also increased the expression of prolyl‐4‐hydroxylase α1, whereas also decreased the levels of collagen degradation and pro‐inflammatory cytokines in AAA. We detected the interface of NONO and NF‐κB p65, and found that NONO silencing inhibited both the nuclear translocation and the phosphorylation levels of NF‐κB p65. Silencing of NONO prevented Ang II‐influenced AAA in ApoE?/? mice through increasing collagen deposition and inhibiting inflammation. The mechanism may be that silencing of NONO decreases the nuclear translocation and phosphorylation of NF‐κB.  相似文献

The use of wood polymer composites (WPCs) in industries has been growing, and this is fueled in part by the use of WPCs in the construction industry. Rape straw flour (RSF)/high-density polyethylene (HDPE) composites as WPCs can also be applied in the construction industry. As a result, the durability of RSF/HDPE composites after natural exposure becomes a concern. Fourier transform infrared (FTIR) spectroscopy is used to monitor the development of degradation products, such as carbonyl groups and vinyl groups, and to determine changes in HDPE crystallinity. Differential scanning calorimetry (DSC) is also used to analyze changes in HDPE crystallinity. The results indicate that the carbonyl index was roughly lower for the RSF/HDPE composites than for the nano-SiO2/RSF/HDPE composites, and the concentration of vinyl groups was generally much larger for the RSF/HDPE composites than for the nano-SiO2/RSF/HDPE composites. At the same time, carbonyl index and vinyl index decrease with an increasing depth of composites, particularly decreasing strongly from 100 to 200 μm. The crystallinity of the samples increase because of natural aging, and the values of crystallinity derived by DSC are smaller than those derived by FTIR.  相似文献

Background

Brain tumor patients often associated with losses of the small-world configuration and neurocognitive functions before operations. However, few studies were performed on the impairments of frontal lobe low-grade gliomas (LGG) after tumor resection using small-world network features.

Methodology/Principal Findings

To detect differences in the whole brain topology among LGG patients before and after operation, a combined study of neurocognitive assessment and graph theoretical network analysis of fMRI data was performed. We collected resting-state fMRI data of 12 carefully selected frontal lobe LGG patients before and after operation. We calculated the topological properties of brain functional networks in the 12 LGG, and compared with 12 healthy controls (HCs). We also applied Montreal Cognitive Assessment (MoCA) in a subset of patients (n = 12, including before and after operation groups) and HCs (n = 12). The resulting functional connectivity matrices were constructed for all 12 patients, and binary network analysis was performed. In the range of , the functional networks in preoperative LGG and postoperative one both fitted the definition of small-worldness. We proposed as small-world network interval, and the results showed that the topological properties were found to be disrupted in the two LGG groups, meanwhile the global efficiency increased and the local efficiency decreased. in the two LGG groups both were longer than HCs. in the LGG groups were smaller than HCs. Compared with the Hcs, MoCA in the two LGG groups were lower than HCs with significant difference, and the disturbed networks in the LGG were negatively related to worse MoCA scores.

Conclusions

Disturbed small-worldness preperty in the two LGG groups was found and widely spread in the strength and spatial organization of brain networks, and the alterated small-world network may be responsible for cognitive dysfunction in frontal lobe LGG patients.  相似文献
