Although Charles Darwin wrote about flower polymorphism in the wild carrot, Daucus carota, the function of the conspicuous central dark floret is still unclear. We used video recordings to evaluate the functional significance of the dark central floret as a short‐distance signal for insects landing on the umbels and analyzed the location of landing points, landing orientation and visit duration. These parameters, as well as insect attraction did not differ on umbels with and without a dark central floret. Hence, we found no evidence for the role of the dark central floret in the pollination of D. carota. Umbels with a dark central floret, however, were parasitized significantly less often by the gall midge Kiefferia pericarpiicola than umbels without a dark central floret. We propose that the dark central floret may play a role in reducing parasite infestation by mimicking an already present gall or deterring oviposition of the gall midge by other means.  相似文献

Microsatellite markers for Schistosoma mansoni were developed using four genomic microsatellite‐enriched libraries. Microsatellites were observed in 65.4% of all sequences. Primer pairs were designed and tested for 23 loci. Eighteen loci produced amplification products, out of which 11 were polymorphic and were further characterized on 100 individuals of S. mansoni. Two to 19 alleles per locus were detected. The average values of expected and observed heterozygosities among the 11 loci were 0.79 and 0.59, respectively.  相似文献

A new species of Daptonema is described based upon morphological characters and 18S rRNA sequence. Daptonema matrona sp. nov. was collected in Pina Basin (north‐eastern Brazil). It differs from all other species of the genus by the presence of reduced cephalic setae and straight spicules. These features require an adaptation of the generic diagnosis. Moreover, the females are characterized by intra‐uterine development of the offspring, considered herein as their major autapomorphic feature. Molecular systematic analyses supported Daptonema matrona sp. nov. as a distinct genetic and evolutionary lineage. The four living genera of Ctenodactylidae (Rodentia) are the only survivors of a flourishing Tertiary group. In this paper, we describe a new Oligocene species from Ulantatal (Chinese Mongolia) that highlights the origins of crown ctenodactylids. This species, Helanshania deserta gen. et sp. nov. , is lophodont and displays semi‐hypsodont teeth, a dental pattern that is somewhat transitional between that of primitive Oligocene ctenodactylids and the later hypsodont genera. We perform here a cladistic assessment of the dental evidence for species produced by the successive radiations of the group. In order to get new data, to decipher homologies for the dental pattern of modern ctenodactylids, and to specify their dental replacement, we describe additional dental material of Ctenodactylus, Massoutiera, and Felovia. The phylogenetic analysis (using PAUP) considered 45 characters (mainly dental) and 31 species. The performed heuristic searches yielded 596 equally most parsimonious trees. Protataromys and Karakoromys are stem ctenodactylids and appear as the earliest offshoots of the Ctenodactylidae clade, which represents a well‐supported family rank. Within this family, the Tataromyinae appear paraphyletic, whereas the Ctenodactylinae sensu lato are a clade including the new taxon Helanshania. As such, a revision of the Tataromyinae is envisaged and a new subfamily is erected (Yindirtemyinae). Hordeum spontaneum , the ancestor of cultivated barley, is mainly distributed in the Mediterranean zone of the Near East as well as in the Irano-Turanian steppes and Saharo-Arabian region; these areas receive unpredictable small amounts and distribution of rain followed by long, hot and dry summers. Caryopses of seven local ecotypes of H. spontaneum were collected in Israel from natural populations near Sede Boker, Neve Yaar, Tabigha and Mount Hermon. Three generations of these ecotypes were grown in natural conditions at Sede Boker with additional irrigation. The germination of caryopses of the third generation was tested. Due to the self pollination and atelechoric system of seed dispersal of this species, genetic ecophysiological diversity of afterripening and salt tolerance was found between the ecotypes originating from different areas, even within local populations and patches of plants that grew on different types or depths of soil at a distance of only a few meters from each other. Afterripening is important to prevent the caryopses from germinating shortly after maturation after a late rain at the beginning of the dry summer. Germination of the different ecotypes is regulated by temperature and length of post-maturation dry storage, as well as temperatures during wetting, and salinity.  相似文献

Five microsatellite loci were isolated and characterized within the woolly mouse opossum (Micoureus paraguayanus), a Neotropical marsupial, using an enrichment cloning procedure. Between four and seven alleles were detected per locus, with expected heterozygosity ranging from 0.358 to 0.560. These microsatellites should provide useful markers in a variety of genetic analyses to examine parentage, inbreeding, population structure and population dynamics in fragmented forest habitats.  相似文献

Heterozygosity–fitness correlations use molecular measures of heterozygosity as proxy estimates of individual inbreeding coefficients (f) to examine relationships between inbreeding and fitness traits. Heterozygosity–fitness correlations partly depend on the assumption that individual heterozygosity and f are strongly and negatively correlated. Although theory predicts that this relationship will be strongest when mean f and variance in f are high, few studies of heterozygosity–fitness correlations include estimates of f based on pedigrees, which allow for more thorough examinations of the relationship between f, heterozygosity and fitness in nature. We examined relationships between pedigree‐based estimates of f, multilocus heterozygosity (MLH) and the probability of survival to hatch in song sparrow nestmates. f and MLH were weakly, but significantly negatively correlated. Inbreeding coefficient predicted the probability of survival to hatch. In contrast, MLH did not predict the probability of survival to hatch nor did it account for residual variation in survival to hatch after statistically controlling for the effects of f. These results are consistent with the expectation that heterozygosity–f correlations will be weak when mean and variance in f are low. Our results also provide empirical support for recent simulation studies, which show that variation in MLH among siblings with equal f can be large and may obscure MLH–fitness relationships.  相似文献

Is the ability to respond to minor perturbations a key to survival to major crisis? This idea was supported by previous studies on Late Devonian conodonts. Genera surviving the Frasnian/Famennian (F/F) crisis, at the end of the Upper Kellwasser anoxic event, had shown a response to a similar but minor event, the Lower Kellwasser (LKW). In contrast, the genus Ancyrodella went extinct at the F/F crisis. This study aims to assess whether this extinction was associated with the absence of response to the minor LKW event. For this purpose, the shape of the P1 platform elements of the conodont Ancyrodella curvata was quantified based on their 2‐D outlines. Conodonts were extracted from two outcrops documenting the Late Devonian Kellwasser events, Coumiac (France) and Mrirt (Morocco). Ancyrodella curvata is characterized by a marked allometric growth that may blur any other source of shape difference, like response to the LKW. Hence, a multivariate method was developed to disentangle size‐free from size‐related shape variations. No coherent temporal size variations emerged from the analysis, but allometry occurring along the growth of the element indeed appeared as the prevailing source of shape variation. By isolating size‐free variations, however, we were able to evidence a morphometric response of A. curvata to the LKW independent from the allometric pattern. Hence, the extinction of A. curvata at the F/F crisis cannot be attributed to an absence of response to minor events. The amount of shape change involved in the LKW shift is, however, of minor importance in comparison with the shape variation due to allometric growth. Alternate hypotheses have been further investigated. Ancyrodella curvata appears as a rare species in the assemblages, and its distribution worldwide suggests a rather specialist taxon. Considering survivorship curves (i.e. frequency distribution of the conodont as a function of its size) A. curvata was characterized by low and constant mortality rate. This may be a clue for a long growth and progressive investment in the reproduction. These arguments support the view that A. curvata was a rather specialist, rare and ‘K‐selected’ taxon. All these factors may not be exclusive and their unfavourable conjunction might have been the key to the extinction of A. curvata.  相似文献

Background

Cytokine-induced killer cells (CIKs) are an advanced therapeutic medicinal product (ATMP) that has shown therapeutic activity in clinical trials but needs optimization. We developed a novel strategy using CIKs from banked cryopreserved cord blood units (CBUs) combined with bispecific antibody (BsAb) blinatumomab to treat CD19+ malignancies.

Methods

CB-CIKs were expanded in vitro and fully characterized in comparison with peripheral blood (PB)–derived CIKs.

Results

CB-CIKs, like PB-CIKs, were mostly CD3+ T cells with mean 45% CD3+CD56+ and expressing mostly TCR(T cell receptor)αβ with a TH1 phenotype. CB-CIK cultures had, however, a larger proportion of CD4+ cells, mostly CD56?, as well as a greater proportion of naïve CCR7+CD45RA+ and a lower percentage of effector memory cells, compared with PB-CIKs. CB-CIKs were very similar to PB-CIKs in their expression of a large panel of co-stimulatory and inhibitory/exhaustion markers, except for higher CD28 expression among CD8+ cells. Like PB-CIKs, CB-CIKs were highly cytotoxic in vitro against natural killer (NK) cell targets and efficiently lysed CD19+ tumor cells in the presence of blinatumomab, with 30–60% lysis of target cells at very low effector:target ratios. Finally, both CB-CIKs and PB-CIKs, combined with blinatumomab, showed significant therapeutic activity in an aggressive PDX Ph+ CD19+ acute lymphoblastic leukemia model in NOD-SCID mice, without sign of toxicity or graft-versus-host disease. The improved expansion protocol was finally validated in good manufacturing practice conditions, showing reproducible expansion of CIKs from cryopreserved cord blood units with a median of 28.8?×?106 CIK/kg.

Discussion

We conclude that CB-CIKs, combined with bispecific T-cell–engaging antibodies, offer a novel, effective treatment strategy for leukemia.  相似文献

Caesalpinia echinata and C. ferrea var. ferrea have different seed behaviours and seed and fruit types. Comparison of the seed ontogeny and anatomy partly explained the differences in seed behaviour between these two species of Brazilian legumes; some differences were also related to fruit development. The seed coat in C. ferrea consisted of two layers of osteosclereids, as well as macrosclereids and fibres, to form a typical legume seed coat, whereas C. echinata had only macrosclereids and fibres. In C. echinata , the developing seed coat had paracytic stomata, a feature rarely found in legume seeds. These seed coat features may account for the low longevity of C. echinata seeds. The embryogeny was similar in both species, with no differences in the relationship between embryo growth and seed growth. The seeds of both species behaved as typical endospermic seeds, despite their different morphological classification (exendospermic orthodox seeds were described for C. echinata and endospermic orthodox seeds for C. ferrea ). Embryo growth in C. ferrea accelerated when the sclerenchyma of the pericarp was developing, whereas embryonic growth in C. echinata was associated with the conclusion of spine and secretory reservoir development in the pericarp.
