Summary Seed and transplanted adult plants from populations of Festuca rubra, collected from inland, salt-marsh and sand-dune sites were grown on culture solution with added sodium chloride. The growth of the populations of the three habitats was reduced differentially by salt. The salt marsh ecotype Festuca rubra ssp. litoralis was only slightly affected and the inland ecotype F. rubra ssp. rubra was severely retarded at 60 mM NaCl. The dune ecotype F. rubra ssp. arenaria had an intermediate tolerance. The tolerant ecotypes accumulated less sodium chloride as compared to the sensitive ecotype, suggesting that salt tolerance is caused in part by salt exclusion.In addition, the dune ecotype F.r. arenaria appeared to be more drought tolerant than the salt marsh ecotype. Abscission of salt-saturated leaves does not function as an adaptation to salinity in Festuca rubra.All three ecotypes accumulated proline with increased salinity. The response was most pronounced in the drought tolerant F.r. arenaria, indicating that proline accumulation is a response to osmotic stress rather than to ion-specific effects of salinity. The observed differences in salt tolerance may be explained by differential sensitivity to toxic effects of sodium chloride.The occurrence on a beach plain of closely adjacent populations of F.r. arenaria and F.r. litoralis, differing markedly in salt tolerance, is briefly discussed.  相似文献

Cultured bovine capillary endothelial (BCE) cells produce low levels of collagenolytic activity and significant amounts of the serine protease plasminogen activator (PA). When grown in the presence of nanomolar quantities of the tumor promoter 12-O-tetradecanoyl phorbol-13-acetate (TPA), BCE cells produced 5-15 times more collagenolytic activity and 2-10 times more PA than untreated cells. The enhanced production of these enzymes was dependent on the dose of TPA used, with maximal response at 10(-7) to 10(-8) M. Phorbol didecanoate (PDD), an analog of TPA which is an active tumor promoter, also increased protease production. 4-O-methyl-TPA and 4α-PDD, two analogs of TPA which are inactive as tumor promoters, had no effect on protease production. Increased PA and collagenase activities were detected within 7.5 and 19 h, respectively, after the addition of TPA. The TPA-stimulated BCE cells synthesized a urokinase-type PA and a typical vertebrate collagenase. BCE cells were compared with bovine aortic endothelial (BAE) cells and bovine embryonic skin (BES) fibroblasts with respect to their production of protease in response to TPA. Under normal growth conditions, low levels of collagenolyic activity were detected in the culture fluids from BCE, BAE, and BES cells. BCE cells produced 5-13 times the basal levels of collagenolytic activity in response to TPA, whereas BAE cells and BES fibroblasts showed a minimal response to TPA. Both BCE and BAE cells exhibited relatively high basal levels of PA, the production of which was stimulated approximately threefold by the addition of TPA. The observation that BCE cells and not BAE cells produced high levels of both PA and collagenase activities in response to TPA demonstrates a significant difference between these two types of endothelial cells and suggests that the enhanced detectable activities are a property unique to bovine capillary and microvessel and endothelial cells.  相似文献

In a greenhouse study, plants of three monocotyledonous and five dicotyledonous species, which occur in a Dutch dune grassland, were exposed to four levels of ultraviolet-B (UV-B) radiation. UV-B levels simulated up to 30% reduction of the stratospheric ozone column during summertime in The Netherlands. Six of the plant species studied in the greenhouse were also exposed to enhanced UV-B irradiance in an experimental field study. In the field experiment plants either received the ambient UV-B irradiance (control) or an enhanced UV-B level simulating 15–20% ozone depletion during summertime in The Netherlands. The purpose of both experiments was to determine the response of the plant species to UV-B radiation and to compare results obtained in the greenhouse with results of the field experiment. Large intraspecific differences in UV-B sensitivity were observed in the greenhouse study. Total dry matter accumulation of monocotyledons was increased, while dry matter accumulation of dicotyledons remained unaffected or decreased. The increase in biomass production of monocotyledons at elevated UV-B was not related to the rate of photosynthesis but to alterations in leaf orientation. In the greenhouse study, UV-B radiation also affected morphological characteristics. Shoot height or maximum leaf length of five out of eight species was reduced. In the field study only one species showed a significantly decreased maximum leaf length at enhanced UV-B. Possible reasons for this discrepancy are discussed. The absorbance of methanolic leaf extracts also differed between species. UV absorbance of field-grown plants was higher than greenhouse-grown plants. In the greenhouse study, the highest UV-B level increased UV-B absorbance of some species. In the field study however, this stimulation of UV absorbance was not observed. In general, results obtained in the greenhouse study were similar to results obtained in the field study. Difficulties in extrapolating results of UV-B experiments conducted in the greenhouse to the field situation are discussed.  相似文献

This study reports changes in the plant's chemical composition and the decomposition of this plant material under enhanced solar UV-B radiation. Calamagrostis epigeios, a dominant grass species in the dune grassland in The Netherlands, was grown outdoor on an experimental field under ambient and enhanced solar UV-B (5 and 7.5 kJ m-2 day-1 UV-BBE, respectively), corresponding to about 15% stratospheric ozone depletion. After one growing season aerial plant parts were harvested. The decomposition of this harvested leaf material was studied in a dune grassland and on the above mentioned experimental field under ambient (5 kJ m-2 day-1 UV-BBE) and enhanced (7.5 kJ m-2 day-1 UV-BBE) radiation, using litter bags. The chemical quality of the leaves grown under enhanced solar UV-B changed. There was an increase in the leaf content of lignin, while no significant changes occurred for the content of -cellulose, hemicellulose and tannins under enhanced UV-B. In the field, the rate of decomposition of leaf material grown under enhanced UV-B (with an increased content of lignin) was reduced. The content of lignin of the decomposing leaf material increased, but less under exposure to enhanced UV-B. The latter may be explained by photodegradation of the lignin. The consequences of enhanced UV-B radiation for carbon fluxes in the dune grassland ecosystem are discussed.  相似文献

Protein farnesyltransferase (PFTase) is a zinc metalloenzyme that catalyzes the posttranslational alkylation of the cysteine in C-terminal -Ca(1)a(2)X sequences by a 15-carbon farnesyl residue, where C is cysteine, a(1) and a(2) are normally aliphatic amino acids, and X is an amino acid that specifies selectivity for the farnesyl moiety. Formation of a Zn(2+) thiolate in the PFTase. peptide complex was detected by the appearance of an absorbance at 236 nm (epsilon = 15 000 M(-1) cm(-1)), which was dependent on the concentration of peptide, in a UV difference spectrum in a solution of PFTase and the peptide substrate RTRCVIA. We developed a fluorescence anisotropy binding assay to measure the dissociation constants as a function of pH for peptide analogues by appending a 2',7'-difluorofluorescein to their N-terminus. The electron-withdrawing fluorine atoms allowed us to measure peptide binding down to pH 5.5 without having to correct for the changes in fluorescence intensity that accompany protonation of the fluorophore. Measurements of the pK(a)s for thiol groups in free and bound peptide indicate that peptide binding is accompanied by formation of a zinc thiolate and that binding to PFTase lowers the pK of the peptide thiol by 3 units. In similar studies with the betaY310F mutant, the pK(a) of the thiol moiety was lowered by 2 units upon binding, indicating that the hydroxyl group in the conserved tyrosine helps stabilize the bound thiolate.  相似文献

In this article, I argue that the practice of forced disappearance of persons on the part of paramilitary groups has become linked to specific processes of globalization. Global flows related to biopolitics, global crime networks, and dehumanizing imaginations reproduced by mass media together constitute a driving force behind forced disappearances. Drawing on ethnographic fieldwork in the Colombian city of Medellín, I analyze how these global flows interact with local armed actors, helping create a climate conducive to forced disappearance. These mechanisms in Colombia show similarities to those in some African and Asian countries. Gaining insight into the mechanisms behind forced disappearance may help prevent it from occurring in the future. Enhancing social inclusion of residents, unraveling the transnational crime networks in which perpetrators are involved, and disseminating rehumanizing images of victims all contribute to curbing the practice of forced disappearance.  相似文献

The response of faba bean seedlings to the combined effects of increased atmospheric CO2 concentrations ([CO2]) and solar UV-B irradiance was studied using open-top chambers transparent to UV-B radiation. The purpose of the study was to determine whether effects of increased [CO2] on growth and physiology are modified by the present solar UV-B fluence rate in the Netherlands. Seedlings were exposed to 350 or 700 micromoles mol-1 CO2. At both [CO2], solar UV-B irradiance was either present or reduced using polyester foil opaque to UV-B radiation. To obtain information on the time dependence of increased [CO2] and UV-B radiation effects, three harvests were performed during the experiment. CO2 enrichment resulted in increased biomass production at all harvests. At the final harvest, UV-B radiation did not affect biomass production but a significant decrease was observed after 14 d of treatment. A reduction of the UV-B fluence increased shoot length at both [CO2] throughout the experiment. UV-B radiation slightly altered biomass allocation. Plants grown at reduced levels of UV-B radiation invested less biomass in flowers and more in stem material compared to plants grown at ambient UV-B levels. CO2 enrichment resulted in a stimulation of net photosynthesis after 26 and 38 d of treatment. UV-B reduction did not alter this response. After 26 d of treatment, photosynthetic acclimation to CO2 enrichment was observed, which was probably the result of accumulation of carbohydrates in the leaves. After 38 d, photosynthetic acclimation was no longer present. The UV absorbance of methanolic leaf extracts was increased by CO2 enrichment only. Both CO2 enrichment and solar UV-B reduced the transmittance of radiation through intact attached leaves. Interaction between [CO2] and UV-B radiation was limited to UV-A transmittance of leaves. Under prevalent experimental conditions, UV-B radiation did not affect the measured physiological parameters. Most open-top chambers used for climate change research are constructed of materials which do not transmit UV-B radiation. Our results indicate that part of the 'chamber effects' on plant height often described in the literature might be explained by the absence of solar UV-B radiation in these chambers.  相似文献

Long-term vegetation changes in permanent plots along two transects from the low to the high salt marsh at the Boschplaat on Terschelling between 1953 and 1990, were analysed in relation to sedimentation and flooding. In both transects, the number of plant species decreased between 1953 and 1990. In lower parts of the two transects the dominant plant species of the vegetation changed from Puccinellia maritima in 1953/54 to Limonium vulgare in 1980 to Atriplex portulacoides in 1990. The increase in A. portulacoides between 1980 and 1990 coincided with an increase in the thickness of the silt layer, caused by a sedimentation rate of about 5 mm per year. This sedimentation led to an increase in elevation but not to large changes in flooding frequency between 1953 and 1990, probably due to a sea level rise during the same period.In the higher parts of the salt-marsh transects Elymus athericus strongly increased and this species became dominant in 1980 and 1990. In these parts of the salt marsh, a small increase in elevation, in thickness of the silt layer, and in flooding frequency occurred between 1953 and 1990.The described changes in vegetation are discussed in terms of general succession of salt-marsh vegetation in relation to flooding and sedimentation.  相似文献

Terrestrial plant species vary widely in their adaptation to (increasing) solar UV-B radiation. Among the various responses of higher plants to enhanced UV-B are increasing leaf thickness and increasing concentrations of UV-B absorbing compounds. In some (UV-B resistant) plant species increased leaf thickness and UV-B absorbance may form part of mechanisms protecting plants from UV-B damage. However, in UV-B sensitive plant species leaf thickness and UV-B absorbance may increase as well with enhanced UV-B radiation. In the latter case however, this response cannot prevent plant damage and disturbance. In the present field study the relationship between these plant parameters and a natural elevational UV-B gradient on the tropical island of Jamaica was described. Four plant species of the Blue Mountain Tropical Montane Forest, occurring on open forest sites along the roadside and paths were studied along an elevational gradient. Plant species studied are Redbush (Polygonum chinense), Wild ginger (Hedychium gardneranum), John Crow Bush (Bocconia frutescens) and White clover (Trifolium repens). The elevational sites were at 800, 1000, 1200, 1400 and 1600 m above sea level. Leaf thickness was measured of leaves of intact plants around midday in the field. Leaf disks (5 mm) were sampled and extracted with a methanol/HCl mixture. UV-B absorption of these leaf extracts was measured spectrophotometrically. For all species leaves from higher elevations were thicker than those from lower elevations. In addition, the absorption of UV-B of leaf extracts increased with increasing elevations. It is assumed that the calculated gradient of the UV-BBE from 800 m above sea level: 9.45 kJ m-2 day-1 to 9.75 kJ m-2 day-1 at 1600 m is related to the measured increase of leaf thickness and UV-B absorbing compounds. The responsiveness of these plant parameters to the elevational gradient does not necessarily imply that the plant species are UV-B resistant. One possibility is that the species studied, which are growing on open, disturbed sites on the forest floor and along mountain-roads, are relatively sensitive to UV-B. In addition to clear sky conditions, mist and clouds occur frequently in this tropical mountane forest at Jamaica. Also, the low nutrient status of the soil (low pH, nutrient deficiency) and the high content of polyphenols in leaves of many plant species of the tropical montane rain forest may relate to the marked response of the species studied with increasing elevation. The germination of seeds of seven plant species occurring in a dune grassland vegetation of the Netherlands, was studied at four levels of UV-B radiation simulating unto 45% stratospheric ozone reduction during April. With the exception of seeds of Senecio jacobaea, germination of the dune grassland species was not affected by enhanced UV-B irradiance. Although a clear UV-B fluence-response relationship was not observed, the germination rate of S. jacobaea seeds and maximal germination percentage were reduced at enhanced UV-B. Germination rate in the dark was higher than germination in the light for Oenothera biennis, Plantago lanceolata, Rumex obtusifolius and S. jacobaea. Total dry biomass accumulation of seedlings was not affected by increased UV-B radiation in any of the species tested. Clear-cut differences in UV-absorbance of methanolic extracts were observed between species. Enhanced UV-B irradiance stimulated UV-absorbance of seedling extracts of Holcus lanatus and Verbascum thapsus. A clear UV-B fluence-response relationship was observed for both species. The results indicate that germination of the studied plant species probably will not be adversely affected by the expected stratospheric ozone reduction in The Netherlands.  相似文献
