The contribution of the Ca2+ sensor S100A1 to in vivo Alzheimer's disease (AD) pathobiology has not been elucidated although S100A1 regulates numerous cellular processes linked to AD. This study uses genetic ablation to ascertain the effects of S100A1 on neuroinflammation, beta-amyloid (Aβ) plaque deposition and Akt activity in the PSAPP AD mouse model. PSAPP/S100A1−/− mice exhibited decreases in astrocytosis (GFAP burden), microgliosis (Iba1 burden) and plaque load/number when compared to PSAPP/S100A1+/+ mice at six and twelve months of age. The presence of detectable S100A1 staining in human AD specimens is consistent with a detrimental gain of S100A1 function in AD. S100A1 ablation also reduced plaque associated and increased non-plaque associated PO4-Akt and PO4-GSK3β staining. S100A1·Akt complexes were undetectable in PC12 cells and AD brain tissue suggesting that S100A1 indirectly modulates Akt activity. In contrast, S100A1·RyR (ryanodine receptor) complexes were present in human/mouse AD brain and exhibited Ca2+-dependent formation in neuronal cells. This is the first direct demonstration of an S100A1· target protein complex in tissue/cells and identifies the RyR as a primary S100A1 target protein in the brain. Collectively, these data suggest that S100A1 inhibition may be a novel strategy for normalizing aberrant Ca2+ signaling in AD.  相似文献

The light brown apple moth, Epiphyas postvittana (Walker), is native to Australia and first was detected in California in 2006. In this study, we regularly sampled populations on Leptospermum laevigatum (Gaertn.) F.Muell. at two sites in San Francisco and on Arctostaphylos densiflora M.S. Baker at two sites in Santa Cruz over a 2-yr period to monitor the abundance, age structure, and voltinism of this potential pest in relation to degree-days. Our results showed that larval abundance declined at two sites, cycled with peaks in midsummer at one site, and remained steady at one site. Generations overlapped at all four sites with the full range of larval instars being present for most of the year, although populations during the winter were predominantly mid to late instars. Accumulated degree-days predict an average of 3.27 and 4.58 generations per year in San Francisco and Santa Cruz, respectively, which matched our observed peaks of late-instar larvae in the field remarkably well. This new information on light brown apple moth phenology in coastal California will be invaluable for the development of effective monitoring and management strategies for this new invader in the studied region.  相似文献

Host range studies of the parasitoid Allotropa sp. near mecrida (Walker) (Hymenoptera: Platygastridae) were conducted prior to applying for a permit to release it against the pink hibiscus mealybug, Maconellicoccus hirsutus (Green) (Hemiptera: Pseudococcidae), in southern California. Using M. hirsutus as a control, four mealybug species were tested as potential hosts. Allotropa sp. near mecrida did not successfully reproduce in any of the mealybug species tested, except for the target species, M. hirsutus. The parasitoid was found to host feed upon one of the non-target species within the confined test environment. Subsequently a USDA-APHIS importation and release permit was granted.  相似文献

Experimental and correlative evidence has steadily mounted over the past 30 years implicating spiders in the suppression of insect herbivore pests in crop fields. A large body of evidence has also shown that increasing agroecosystem vegetation diversity often influences the abundance of herbivores and their natural enemies. In previous experiments, the abundance of several species of spiders on grapevines in a raisin grape vineyard was twofold enhanced in vineyard plots vegetationally diversified with a cover crop. A concomitant reduction in the abundance of the leafhopper pest Erythroneura variabilis Beamer was observed on grapevines in the diversified plots, but a causal relationship was not established. In the present study, we simultaneously manipulated spider densities (in open‐vine spider exclusion and vine‐shoot enclosures) and ground cover to determine their relative impact on E. variabilis population dynamics. Open‐vine spider exclusion resulted in an average 35% increase in the density of E. variabilis the greatest effect with occurring during the first and second leafhopper generations. The negative impact of spiders on E. variabilis densities was corroborated with vine‐shoot enclosure experiments. Under the conditions of the present study, the cover crop per se did not affect the dynamics of E. variabilis populations on grapevines, despite a 1.6‐fold increase in spider densities on vines in cover crop plots, compared with vines in bare ground plots, probably due to insufficient spider enhancement and low overall E. variabilis abundance during the summer months. The cover crop had little effect on vine macronutrient status (and presumably vine water status). While this study provided further support for the hypothesis that vegetation diversity can enhance spider abundance, this enhancement does not always lead to lower pest densities, thus underscoring the complexity and variability that exists in interactions involving cover crop, spiders, and crop plants and their herbivore pests.  相似文献

Evaluation of several degree-day estimation methods in California climates   总被引:1,自引:0,他引:1  
 Procedures for estimating degree-day accumulations are frequently employed instead of the more accurate method of calculating degree-days from hourly temperature data because on-site temperature data are commonly restricted to daily minimum and maximum temperature records. Data from seven methods of estimating degree-days at each of nine locations during 2 years in California were compared by month to degree-day values calculated by hourly summation. Methods included three sine-wave approaches, three triangulation approaches and the averaging (i.e., rectangle) method. Results of the double-sine and corrected-sine (i.e., corrected for day length) methods were nearly identical to those of the single-sine method. The double triangulation and corrected triangulation methods produced very similar results to the single triangulation method. The averaging method and sine-wave methods deviated to a greater extent from degree-day accumulations calculated from hourly temperatures from November through February than did the triangulation methods. Degree-day estimations from the late spring and summer months were more similar to one another for all estimation methods than during the cooler months of the year. Since no advantages were noted in the more complicated double and corrected methods, the single triangulation method or the sine-wave method is preferred as they are less complicated procedures. Of the various temperature threshold cut-off methods evaluated, error levels were unaffected when estimating degree-days using the sine-wave method. The employment of a horizontal cut-off with the triangulation method did not significantly increase the amount of error in the estimation of degree-days. However, an increase in error was observed when employing the intermediate cut-off and vertical threshold cut-off techniques with the triangulation method for computing degree-days. Peripheral vasculature resistance can play an important role in affecting blood pressure and the development of cardiovascular disease. A better understanding of the genes that encode vasodilators, such as adenosine, will provide insight into the mechanisms underlying cardiovascular disease. We tested whether the adenosine monophosphate deaminase-1 (AMPD1) C34T gene polymorphism was associated with the vasodilatory response to ischemia in Caucasian females aged 18-35 years. Blood samples (n = 58) were analyzed for the C34T variant and resulted in the following genotype groups: CC (n = 45) and CT (n = 13). Mean blood pressure (MBP), heart rate, and forearm blood flow (FBF) measured by venous occlusion plethysmography were measured at baseline and at 1 (peak FBF), 2 and 3 min of vasodilation during reactive hyperemia following 5 min of arm ischemia. To control for interindividual variability in baseline FBF and forearm vascular resistance (FVR) the percent change in FBF and FVR were calculated for each min. The percent decrease in FVR was significantly greater in the CT compared to the CC genotype group (-40+/-4% vs. -24+/-3%, P = 0.01) during the 2nd min of reactive hyperemia. The percent increase in FBF tended to be greater in the CT compared to the CC genotype group (+69+/-9% vs. +42+/-9%, P = 0.07) during the 2nd min of reactive hyperemia after adjustment for percent body fat. Consistent with previous findings of increased production of adenosine during exercise in individuals carrying a T allele, our findings suggest that the AMPD1 C34T polymorphism is associated with vasodilatory response to ischemia in the peripheral vasculature because individuals with the T allele had a greater vasodilatory response to ischemia.  相似文献

Resident natural enemies can impact invasive species by causing Allee effects, leading to a reduction in establishment success of small founder populations, or by regulating or merely suppressing the abundance of established populations. Epiphyas postvittana, the Light Brown Apple Moth, an invasive leafroller in California, has been found to be attacked by a large assemblage of resident parasitoids that cause relatively high rates of parasitism. Over a 4-year period, we measured the abundance and per capita growth rates of four E. postvittana populations in California and determined parasitism rates. We found that at two of the sites, parasitism caused a component Allee effect, a reduction in individual survivorship at lower E. postvittana population densities, although it did not translate into a demographic Allee effect, an impact on per capita population growth rates at low densities. Instead, E. postvittana populations at all four sites exhibited strong compensatory density feedback throughout the entire range of densities observed at each site. As we found no evidence for a negative relationship between per capita population growth rates and parasitism rates, we concluded that resident parasitoids were unable to regulate E. postvittana populations in California. Despite a lack of evidence for regulation or a demographic Allee effect, the impact of resident parasitoids on E. postvittana populations is substantial and demonstrates significant biotic resistance against this new invader.  相似文献
