Tree decline and deaths have been observed among 15 to 20-year-old Scots pines (Pinus sylvestris L.) in a dry heath forest in southwestern Finland. The sudden decline in height growth, the dieback of leading shoots and the yellowing of needles in young shoots in the upper part of the tree are typical symptoms of the decline of these young pines. Needle ultrastructure and chemical composition of Scots pines with or without decline and fluctuations of them in different seasons were studied. Afflicted trees were found to suffer from a deficiency in calcium and magnesium with low concentration of foliar nitrogen and phosphorus observed in all the trees studied. Ultrastructural study revealed changes characteristic of different seasons and measured nutrient status of needles. A clear reduction of membrane system in chloroplasts, especially related to Mg deficiency, was observed in most samples. The symptoms related to N deficiency, the translucent appearance of the cytoplasm and chloroplast stroma, and the elongated chloroplasts, as well as swelling of mitochondria, indicating P deficiency, were also found in the needles sampled from this forest decline area. The present study showed that it is possible to detect specific nutrient deficiency symptoms in needle ultrastructure in field samples and for use as sensitive indicators of unbalanced nutrient status.  相似文献

We have used a gene replacement strategy to delete the previously isolated gene [(1987) EMBO J. 6, 2825-2833] for the cytochrome c oxidase subunit I from Paracoccus denitrificans. The resulting mutant was still able to synthesize active cytochrome c oxidase. This led us to look for another locus which could completely suppress the mutation. In this study we report the isolation of a second gene encoding subunit I. An open reading frame coding for cytochrome c 550 was found upstream from this gene. We suggest that there are isoenzymes of cytochrome c oxidase (cytochrome aa3) in this bacterium.  相似文献

This study deals with the effect of pine bark bugs (Aradus cinnamomeus Panzer) on the nutrition of young Scots pines (Pinus sylvestris L.). Soil and needle samples for analytical purposes were collected from a young pine stand growing on a dry barren mineral soil afflicted by pine bark bugs.The damage to vascular tissues caused by pine bark bugs disturbed the nutrition of the trees, especially in the top part of the crown. The foliar calcium, magnesium, manganese and sulphur concentrations were highly reduced. Scots pines suffered from a lack of calcium, magnesium, nitrogen and phosphorus. These deficiencies were secondary by nature, because no differences were observed between the nutrient concentrations of the underlying soil of the healthy and affected trees. The symptoms of trees damaged by pine bark bugs resembled most of all calcium deficiency symptoms.  相似文献

Retroanandamide (2f) and its 10 analogues (1a-e, 2a-e) were synthesized and evaluated for the cannabinoid receptor activation by a [35S]GTPgammaS binding assay using rat cerebellar membranes, and Chinese hamster ovary cell membranes expressing human CB2 receptors. The primary goal of the study was to develop cannabinoid receptor agonists having improved enzymatic stability compared to endogenous N-arachidonoyl ethanolamide (AEA). Furthermore, by reversing the amide bond of AEA, the formation of arachidonic acid would be prevented. Finally, an effect of the carbonyl carbon position on the cannabinoid receptor activity was explored by synthesizing retroanandamide analogues having different chain lengths (1a-e, C19; 2a-f, C20). All the synthesized compounds, except 2c, behaved as partial agonists for the both cannabinoid receptors. In rat brain homogenate, the reversed amides possessed significantly higher stability against FAAH induced degradation than AEA. Therefore, the reversed amide analogues of AEA may serve as enzymatically stable structural basis for the drug design based on the endogenous cannabinoids.  相似文献

An earlier study suggested that soils on the windward slope of Mount Haleakala are excessively waterlogged at low altitudes but become better drained upslope. We analyzed altitudinal changes in soil N turn over, vegetation mass, and foliar chemical composition of the dominant canopy species, Metrosideros polymorpha (Myrtaceae), on this slope. The basal areas and DBH2× height (an index for volume) of woody species ≥2 m in height increased with altitude (as opposed to a general trend elsewhere), peaked at 1800 m elev., and abruptly declined above this altitude. Foliar N and P concentrations of Metrosideros showed a similar altitudinal trend with DBH2× height values. Foliar Fe and Al concentrations steadily increased downslope, but were low again at 450 m, the lowest altitude, contrary to expectation based on waterlogging. Toxic forms of Fe and Al may retard root penetration in waterlogged soils and thus the uptake of these elements. The net N mineralization rate for topsoils incubated in situ was 78.6 μg/g/20d at 1800 m elev., while small amounts of N were immobilized at 450, 1200 and 2200 m elev. The net N mineralization rates of the topsoils, which were air-dried to their permanent wilting points and incubated at 26°C, were greater than those of the wet samples at 26°C, only for the lowest altitude. Conversely, the net N mineralization rates of the topsoils which were kept wet and incubated at the higher temperature of 26°C were greater (P < 0.1) than those of the in situ wet samples only for 1800 m elev. The rates were the same for 450 and 2200 m elev. where the soils were waterlogged and very dry, respectively; thus, N turn over was more strongly limited by moisture than by temperature in these cases. Nutrient availability improved with altitude up to 1800 m elev. in association with the moisture gradient, and appeared to be responsible for the observed pattern in vegetation mass.  相似文献

Several loci on theParacoccus denitrificans chromosome are involved in the synthesis of cytochromec oxidase. So far three genetic loci have been isolated. One of them contains the structural genes of subunits II and III, as well as two regulatory genes which probably code for oxidase-specific assembly factors. In addition, two distinct genes for subunit I have been cloned, one of which is located adjacent to the cytochromec 550 gene. An alignment of six promoter regions reveals only short common sequences.  相似文献

The susceptibility of Scots pine (Pinus sylvestris L.) sap- and heartwood against the wood decaying brown-rot fungus (Coniophora puteana) was investigated after long-term forest fertilization at three different sites in central Finland. Different wood properties: wood extractives, wood chemistry, and wood anatomy were used to explain sap- and heartwood decay. Scots pine sapwood was more susceptible to decay than its heartwood. In one site, sapwood seemed to be more resistant to wood decay after forest fertilization whereas the susceptibility of heartwood increased. Significant changes in the sapwood chemistry were found between treatment and sites, however, no relationship between wood chemistry and wood decay was observed in the factor analysis. The results of this study show that there was an inconsistent relationship between decay susceptibility and fertilization and the measured physical and chemical attributes of the wood were not consistently correlated with the decay rate.  相似文献

Synthetic oligonucleotide probes were used to clone two loci from the chromosomal DNA of Paracoccus denitrificans that contain the genes for cytochrome c oxidase (cytochrome aa3). One locus seems to contain four or five genes probably forming an operon. Two of these code for the oxidase subunits II and III. Three open reading frames are found between the COII and COIII genes. The other locus codes for the subunit I. A short open reading frame is found upstream of this gene. All three subunits of the Paracoccus enzyme show remarkable homology to the corresponding subunits of the mitochondrial cytochrome oxidase. Possible protein products of the open reading frames have not yet been identified.  相似文献

The gene coding for cytochrome oxidase subunit I (COI) was isolated from a genomic DNA library of the thermophilic bacterium PS3 and sequenced. The N-terminal of the COI protein was also sequenced to verify the initiation site of the reading frame. The deduced amino acid sequence of COI protein is composed of 536 amino acid residues and its molecular mass is 59,510. The protein is clearly homologous to the corresponding subunit in the mitochondrial cytochrome oxidase and similarly appears to have 12 trans-membrane segments. The proposed ligands to two hemes (cytochrome aa3) and a copper atom (CuB) in this protein (Holm et al. (1987) EMBO J. 6, 2819-2823) are conserved in the sequence.  相似文献
