The optimization of regeneration protocol for different genotypes increases the yield in transformation studies. Cotyledon explants of watermelon [Citrullus lanatus (Thunb) Matsum & Nakai] cv Crimson Sweet were cultured on MS medium containing combinations of benzyl adenine (BA) (0, 5, 10, 20 µM) and indole-3-acetic acid (IAA) (0, 0.5, 5 µM). Maximum shoot growth and subsequent rooting from explants on regeneration medium were obtained from the media containing 10 µM BA + 0.5 µM IAA and 20 µM BA (75 and 78%) by direct organogenesis, respectively. Histological analysis showed that cell division was observed in the epidermal and subepidermal layers. Protuberant structures were observed in tissues between 7 and 12 days in culture. Meristematic structures were observed after 12days in culture which later developed into buds. 相似文献

The effects of the calculated photostationary state of phytochrome (c) and the photon fluence rate on the elongation growth of the hypocotyl of light-grown seedlings of Cucumis sativus L. are examined. Two threshold responses to c are found at values of 0.06 and 0.43. At c = 0.06, there is no response at any fluence rate. In the c range 0.1 to 0.43, elongation growth does not respond to changes in c. Above the second threshold (c = 0.43), there is a strong response to changes in c. At all values of c at and above 0.1, there is a response to fluence rate. A linear relationship can be demonstrated between a factor comprised of the logarithm of phytochrome cycling rate (a fluence-rate-dependent process) and c, and the growth response. 相似文献

In Hank's balanced salt solution EL-4 ascites thymoma cells possessed endogenous respiration which was sufficient for the maintenance of their ATP level: pH decrease down to 6.0 had no effect either on endogenous respiration or the ATP level. Glucose had no influence on the respiration of EL-4 cells but inhibited that of Ehrlich ascites carcinoma (EAC) cells by 40% (Crabtree effect); respiration of the both cell lines was strongly (4-fold) inhibited after simultaneous addition of glucose, lactate and pH decrease. EL-4 cells had no endogenous glycolysis; EAC cells showed a low level of glycolysis only after pH decrease. Glucose addition led to activation of glycolysis (both inhibited 2-fold after a decrease of pH down to 6.0. The respiration inhibition at pH 7.3 and 6.0 caused no decrease of ATP depletion when glucose was present in the medium; this result may be due to suppression of ATP consumption. Incubation of EL-4 cells under respiration and glycolysis deficiency conditions resulted in a sharp ATP depletion; pH decrease delayed this depletion. 相似文献

Abstract High-throughput sequencing approach of the 16S rRNA gene was employed to evaluate the bacterial diversity inhabit in melted water, snow, soil, and rocks samples at the lower altitudes of the Laohugou glacial environment. Bioinformatics tools were used to process millions of Illumina reads for alpha and beta diversities of bacterial communities. The diversity indices such as Chao, Shannon, and Simpson were different in the collected samples and solid samples (soil and rocks) showed higher taxon richness and evenness. Taxonomic diversity was unexpectedly higher and the major portion of sequences was assigned to Proteobacteria, Actinobacteria, and Acidobacteria. Higher variation in community structure was reported at the class level and Alphaproteobacteria was dominant. The solid niches were occupied by a higher number of phyla compared with liquid. The physicochemical variables acted as spatial gradients and associated with the bacterial structural communities of the glacial ecosystem. Findings showed that both Proteobacteria and Actinobacteria in solid samples influenced the bacterial community structure in downstream liquid samples. Interestingly, the metagenomic biomarkers were higher in liquid samples. This study provides precious datasets to understand the bacterial community in a better way under the influence of spatial, physical and environmental factors. 相似文献

Paced QRS morphology predicts incident left ventricular systolic dysfunction and atrial fibrillation

Background
The prognostic significance of paced QRS complex morphology on surface ECG remains unclear. This study aimed to assess long-term outcomes associated with variations in the paced QRS complex.Methods
Adult patients who underwent dual-chamber pacemaker implantation with 20% or more ventricular pacing and a 12-lead ECG showing a paced complex were included. The paced QRS was analyzed in leads I and aVL. Long-term clinical and echocardiographic outcomes were compared at 5 years.Results
The study included 844 patients (43.1% female; age 75.0?±?12.1). Patients with a longer paced QRS (pQRS) duration in lead I had a lower rate of atrial fibrillation (HR 0.80; p?=?0.03) and higher rate of systolic dysfunction (HR 1.17; p?<?0.001). Total pacing complex (TPC) duration was linked to higher rates of ICD implantation (HR 1.18; p?=?0.04) and systolic dysfunction (HR 1.22, p?<?0.001). Longer paced intrinsicoid deflection (pID) was associated with less atrial fibrillation (HR 0.75; p?=?0.01), more systolic dysfunction (HR 1.17; p?<?0.001), ICD implantation (HR 1.23; p?=?0.04), and CRT upgrade (HR 1.23; p?=?0.03). Exceeding thresholds for TPC, pQRS, and pID of 170, 146, and 112?ms in lead I, respectively, was associated with a substantial increase in systolic dysfunction over 5 years (p?<?0.001).Conclusions
Longer durations of all tested parameters in lead I were associated with increased rates of left ventricular systolic dysfunction. Antibiotic resistance is recognized as a global threat to public health. The selection and evolution of antibiotic resistance in clinical pathogens were believed to be majorly driven by the imprudent use of antibiotics. However, concerns regarding the same, through selection pressure by a multitude of other antimicrobial agents, such as heavy metals, are also growing. Heavy metal contamination co-selects antibiotic and metal resistance through numerous mechanisms, such as co-resistance and cross-resistance. Here, we have reviewed the role of heavy metals as antimicrobial resistance driving agents and the underlying concept and mechanisms of co-selection, while also highlighting the scarcity of studies explicitly inspecting the process of co-selection in clinical settings. Prospective strategies to manage heavy metal-induced antibiotic resistance have also been deliberated, underlining the need to find specific inhibitors so that alternate medicinal combinations can be added to the existing therapeutic armamentarium. 相似文献

Agro-ecosystems constitute essential habitat for many organisms. Agricultural intensification, however, has caused a strong decline of farmland biodiversity. Organic farming (OF) is often presented as a more biodiversity-friendly practice, but the generality of the beneficial effects of OF is debated as the effects appear often species- and context-dependent, and current research has highlighted the need to quantify the relative effects of local- and landscape-scale management on farmland biodiversity. Yet very few studies have investigated the landscape-level effects of OF; that is to say, how the biodiversity of a field is affected by the presence or density of organically farmed fields in the surrounding landscape. We addressed this issue using the metacommunity framework, with weed species richness in winter wheat within an intensively farmed landscape in France as model system. Controlling for the effects of local and landscape structure, we showed that OF leads to higher local weed diversity and that the presence of OF in the landscape is associated with higher local weed biodiversity also for conventionally farmed fields, and may reach a similar biodiversity level to organic fields in field margins. Based on these results, we derive indications for improving the sustainable management of farming systems. 相似文献
