Ethylene effects on growing and gravireacting maize root segments   总被引:1,自引:0,他引:1  
The effects of ethylene pretreatments (500 nl/l for 1 h) and treatments (100 nl/1 to 1000 nl/l for 6 h) on elongation and gravireaction of apical maize root segments were tested in light and in the dark. Ethylene was found to affect weakly root elongation and gravireaction, but to induce strong curvatures for root growing vertically.  相似文献

The level of endogenous Indol-3-yl-acetic acid (IAA) measured by gas chromatography-mass spectrometry in the elongating zone of intact primary roots of Zea mays showed a good linear correlation with the growth rate of these roots. When they were treated with IAA, their relative elongation decreased; this indicates a supraoptimal content of endogenous IAA. However, the growth of some of the relatively rapidly extending roots was enhanced by such treatment. The effects of a synthetic growth promoter, 4-ethoxy-l-( p -tolyl)-S-triazine-2,6 (1H, 3H)-dione [TA], on growth and gravireaction of Zea mays L. (cv. LG 11) roots were investigated. In horizontal, intact roots, pretreatment with TA at 4 × 10−4 M inhibited the gravireaction. If the pretreated roots were rinsed with a buffer solution before incubation, the TA effect was reduced, indicating that a continuous presence of TA was necessary for its maximal activity. On the other hand, the TA pretreatment (1×10−5, 1×10−4 and 4 × 10−4 M ) promoted the elongation of these roots. The TA effect was stronger for illuminated roots than for those kept in darkness. TA also decreased the lateral curvature of half-decapitated roots maintained vertically in light. This indicates that the action of TA could be associated with some growth inhibiting substances produced or released in cap cells.  相似文献

The cell-wall polysaccharides from different parts of maize roots have been analysed. The arabinose, galactose and mannose contents are influenced by cell differentiation, whereas xylose, rhamnose and uronic-acid contents are not. In cap cells, the pectin content is low but rhamnose and fucose are present in larger quantities. The cell-wall polysaccharides from cells of the elongation zone and their respective regenerating protoplasts were also analysed. The walls of the protoplasts contained higher xylose and mannose levels and a much lower level of cellulose than the cells from which they were derived.  相似文献

Protoplasts were isolated from cortical cells of the elongating zone of maize (Zea mays L. cv. LG 11) roots and submitted to microelectrophoresis. Significant and transient differences in zeta potential between protoplasts from upper and lower root sides were compared with the gravireaction and the differential elongation of these roots. The maximum difference in the zeta potential was obtained between protoplasts from the upper and lower cortical cells after 90 min, exactly the time of gravipresentation for which the maximum rate of gravireaction was observed. In addition, this almost corresponded to the time for which the difference between the elongation rates of upper and lower sides of the extending zone began to increase. Consequently, the changes in the charges of the plasmalemma of the cortical cells from the growing part of roots could be more or less directly related to the root graviresponse.  相似文献

Four glycosidases were analyzed in 10 mm apical segments prepared from growing roots (15 mm) of Zea mays L. The pH optima were found to be 5.8 for -glucosidase, 4.4 for -galactosidase, 6.4 for -glucosidase and 6.0 for -galactosidase. The -glucosidase showed 4-fold higher activity than the -galactosidase. Apical segments from maize roots of LG 11 and Orla 264 varietiesgeoreacted, at least for the first few hours, only in light,while those of the Anjou 210 variety were georeactive both inlight and darkness. The energy charge in the apical end of thetwo light geo-sensitive (LG and Orla) maize roots significantlyincreased in the light. In contrast, in Anjou root tips, theenergy charge was already high in the dark and did not changesignificantly after light exposure. The time course of light-inducedchanges in adenine nucleotide contents in root tips was comparedwith varietal differences in georeactivity in light and darkness.The present data corroborate the hypothesis that a high energystate in the root tip is a prerequisite for the expression ofthe root reaction to gravity. The mode of expression of cellulase activity obtained by viscosimetricmeasurement is analysed. After testing different types of substrates,it appears that the best one is hydroxyethylcellulose used ata high degree of polymerisation and a high concentration. Comparisonof results obtained with cellulases from Trichoderma virideand extracted from Pisum sativum favours the validity of thedetermination proposed.
