A strain of Stachybotrys atra isolated from a field case of stachybotryotoxicosis in Hungary was cultured in Hungary. All of the compounds toxic to brine shrimp were separated from the culture extract by solvent partition, column chromatography, and preparative thin-layer chromatography. Two of the toxic compounds were identified as verrucarin J and satratoxin H by comparison with pure standards resolved by high-pressure liquid chromatography and characterized by mass spectrometry. Two other toxic components were identified as roriden E and satratoxin G on the basis of their mass spectra. The fifth toxic compound was identified as a macrocyclic trichothecene based on the following findings: a positive 4-(p-nitrobenzyl)pyridine color reaction, hydrolysis resulting in verrucarol verified by combined gas chromatography-mass spectrometry, and a characteristic trichothecene proton-nuclear magnetic resonance spectrum. This macrocyclic trichothecene has a molecular ion (528) identical to satratoxin H, and its mass spectrum is similar; however, its Rf value on Silica Gel G differs.  相似文献

The cis and trans isomers of zearalenone [2,4-dihyroxy-6-(10-hydroxy-6-oxo-1-undecenyl)-benzoic acid mu-lactone] and zearalenol [2,4-dihydroxy-6-(6,10-dihydroxy-1-undecenyl)-benzoic acid mu-lactone] were tested for uterotropic activity in the white rat. The metabolites were administered through the oral route (per os) and by topical application to the freshly shaven skin on the back. cis-Zearalenone was significantly more active than trans when fed orally to the rats in the diet or when applied topically by skin application. However, the cis isomer of zearalenol was not significantly different than its trans isomer. trans-Zearalenone was less active than trans-zearalenol.  相似文献

Eight isolates of Fusarium roseum and three of Fusarium colmorum were found to produce deoxynivalenol in rice cultures. Deoxynivalenol was extracted with aqueous methanol (40%) and purified by partitioning with ethyl acetate and acetonitrile-petroleum ether (boiling point, 60--70 degrees C). The toxin was identified by gas chromatography/mass spectrometry and quantified by gas-liquid chromatography. High recoveries (80%) of deoxynivalenol were obtained from rice cultures, and as low as 0.250 microgram of the toxin per g was detected.  相似文献

The mycotoxins diacetoxyscirpenol, deoxynivalenol, and zearalenone, produced by Fusarium roseum, were found naturally occuring in mixed feed samples. In all cases analyzed, deoxynivalenol occurred together with zearalenone. The natural occurrence of zearalenone in sesame seed is reported for the first time. Strains of F. roseum isolated in various parts of the world form feed implicated in animal mycotoxicosis produced monoacetoxyscirpenol, diacetoxyscirpenol, deoxynivalenol, and zearalenone.  相似文献

 Field studies of gas exchange of Populus deltoides, Prosopis juliflora and Acacia auriculiformis showed large diurnal changes in net photosynthesis (A) and stomatal conductance (gs) during autumn. P. deltoides and P. juliflora undergo pronounced midday depression in A and gs while A. auriculiformis showed a one-peak response. Several factors indicative of photosynthetic performance were found to be reversibly affected during afternoon decline. These include (i) decrease in initial slope of the CO2 response curve (carboxylation efficiency), (ii) substantial increase in CO2 compensation point and (iii) decrease in overall quantum yield of photosystem II. The phenomenon can be duplicated in potted plants by simulating a typical daily pattern of PPFD and VPD. It is found that high VPD induces significant decline in A and gs at moderate temperature and saturating PPFD (800 μmol m–2 s–1) whereas these parameters are only marginally affected at high PPFD and low VPD. Fluorescence data show that the tree species under study have a high capacity for safe dissipation of excessive excitation energy. The activation of photorespiration, as evident from an increase in CO2 compensation point, maintains constant internal CO2 concentration (Ci) which may aid in minimizing photoinhibition during stomatal closure at midday. In case of P. deltoides and P. juliflora the stomata seem to be quite sensitive to the changes in humidity whereas this does not appear to be essential in case of A. auriculiformis because of its phyllode structure that endows it with mechanisms for conserving water without undergoing large-scale stomatal changes. The midday depression in net photosynthetic rate (PN) and stomatal conductance (gs) in Prosopis juliflora was studied in relation to two key enzymes of carbon metabolism. Diurnal gas exchange measurements carried out in autumn on P. juliflora showed a pronounced depression in PN and gs along with a decrease in apparent carboxylation efficiency (CE*) during midday. The activities of sucrose-phosphate synthase (SPS) and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) also showed large diurnal fluctuations. Initial RuBPCO activity (that present in vivo) and total activity (fully carbamylated activity) increased gradually with increase in irradiance, in the morning reaching a maximum by 08:00 h. The RuBPCO activity declined sharply during midday due to decrease in initial activity. The drop in the % activation of RuBPCO indicated that the deactivation of RuBPCO was achieved via a decarbamylation mechanism. There was a marked similarity in the diurnal patterns of SPS activity and the PN. During a diurnal rhythm, SPS activity increased after irradiation, reaching a maximum at 08:00 h and then declined during midday. Diurnal fluctuations in SPS activity could be due to the changes in the amount of protein (changes in Vmax) as well as to the changes in kinetic properties (changes in Vlim). Hence the midday decline in photosynthesis is closely associated with the regulation of RuBPCO and SPS activities. A suitable method was developed to efficiently incorporate 13C-labeled acetate into zearalenone by using solid cultures. Periodic feeding of the label during the zearalenone production phase significantly increased the label incorporation for the singly labeled acetate.  相似文献
