Biodiesel is a renewable and environmental friendly energy source that can be produced via tranesterification from various oil crops such as soy bean, sunflower, palm, and algae. In this work, the microalgae Scenedesmus obliquus, S. armatus and S. bernadii, isolated from natural water basins, were enriched in modified Chu 13 medium. Only S. obliquus showed significant oil accumulation and was thus further cultivated in 3 L tubular photo-reactors under mixotrophic conditions (16:8 h light-dark cycle) at room temperature and varying CO2 (5, 10, and 15%) supply. The results indicated that S. obliquus can be grown under various CO2 concentrations. A maximum biomass of 2.3 g/L was achieved when 15% CO2 was used. The effect of salinity on oil storage was also considered, using sodium chloride (NaCl) solutions of varying concentrations (0.05, 0.2, and 0.3 M). Higher lipid contents were found in cells that were subjected to salt stress compared to those in conditions without salt stress. A maximum oil accumulation of 36% was observed within 15 days at 0.3 M NaCl. A biodiesel yield of up to 97.4% was obtained.  相似文献

Male germ cells of the greater bandicoot rat, Bandicota indica, have recently been categorized into 12 spermiogenic steps based upon the morphological appearance of the acrosome and nucleus and the cell shape. In the present study, we have found that, in the Golgi and cap phases, round spermatid nuclei contain 10-nm to 30-nm chromatin fibers, and that the acrosomal granule forms a huge cap over the anterior pole of nucleus. In the acrosomal phase, many chromatin fibers are approximately 50 nm thick; these then thickened to 70-nm fibers and eventually became 90-nm chromatin cords that are tightly packed together into highly condensed chromatin, except where nuclear vacuoles occur. Immunocytochemistry and immunogold localization with anti-histones, anti-transition protein2, and anti-protamine antibodies suggest that histones remain throughout spermiogenesis, that transition proteins are present from step 7 spermatids and remain until the end of spermiogenesis, and that protamines appear at step 8. Spermatozoa from the cauda epididymidis have been analyzed by acid urea Triton X-100 polyacrylamide gel electrophoresis for basic nuclear proteins. The histones, H2A, H3, H2B, and H4, transitional protein2, and protamine are all present in sperm extracts. These findings suggest that, in these sperm of unusual morphology, both transition proteins and some histones are retained, a finding possibly related to the unusual nuclear form of sperm in this species.  相似文献

This work revealed for the first time the possible use of a newly isolated Bacillus aryabhattai PKV01 for poly-β-hydroxyalkanoates (PHAs) production from fermentative sweet sorghum juice. Its growth and PHA production were investigated under different pH and nitrogen sources. Medium composition was optimized using statistical tools. The highest biomass and PHA content were reached at pH 6.5 with the use of urea. Plackett-Burman design was then applied to test the relative importance of medium components and process variables on cell growth and PHA production. Cell growth and PHAs production were affected by total sugar and urea and were subjected to optimize the sorghum juice medium using response surface methodology (RSM) via central composite design (CCD). The predicted optimal culture composition was achieved. Maximum dry cell weight and PHAs were obtained using a flask and almost double the amount was achieved using a bioreactor. After PHA recovery, the structure and thermal properties were characterised and revealed to be similar to the standard of poly-β-hydroxybutyrate (PHB).  相似文献

Five new C-methylated flavonoids (15) together with seven known compounds have been isolated after chromatographic separation of the hexane and CH2Cl2 extracts of the roots of Pisonia grandis (Nyctaginaceae). Structural identification of the new compounds was accomplished by spectroscopic methods. The radical scavenging and antimicrobial properties of the isolates were investigated.  相似文献
