Molecular Characterization of a Barley Gene Induced by Cold Treatment   总被引:4,自引:1,他引:3  
A cDNA library was made from low positive temperature (6 ?C/2?C) grown barley shoot meristems. Several genes which are differentiallyexpressed, as measured by mRNA abundance, were selected fromthe library using a differential screen. This paper reportsan analysis of in vivo expression in several cultivars, theDNA sequence, copy number and chromosomal location of one gene(BLT14). In addition, genomic restriction fragment length polymorphismfor this gene in the 10 most widely UK-grown spring and 9 mostwidely UK-grown winter barley cultivars is analysed. Pearce, R. S. 1985. A frceze-fracture study of membranes ofrapidly drought-stressed leaf bases of wheat.—J. exp.Bol. 36: 1209-1221. Bases of expanding leaves were taken from well-watered or drought-hardenedwheat seedlings, and were progressively dehydrated (over ?–9h or, more slowly, for 24 h or 36 h) to between 76% and 5% ofthe water content of the turgid tissue. Damage was assessedby an ion-leakage test. The dehydrated tissues were freeze-fixedwithout rehydration. Patches free from intramembraneous particles(IMP) occurred in the plasma membrane, tonoplast and chloroplastenvelope of all the damaged leaf bases, and were mostly absentfrom undamaged tissues and controls. 15% of these patches appearedto have an ordered sub-structure. Lamellae with few or no IMP,were associated with some IMP-free patches of plasma membrane.Sometimes IMP-free patches and lamellae were associated withIMP-free folds. Groups of IMP-free lamellae occurred in thecytoplasm of the most severely stressed material. Vesicles andmembraneous sacs accumulated just below the plasma membranein some cells from stressed drought-hardened leaf bases. Depressions,‘lesions’ (mainly unusual circular discontinuities),and associated IMP-free patches, occurred in some plasma membranes,mostly in the stressed hardened tissues, including in non-damagedtissue. The results are related to an hypothesis previouslysuggested to explain damage due to extracellular freezing inwheat tissues: the stress causes cell dehydration and this inducesIM P-free patches leading to membrane reorganization (here expressedas IMP-free lamellae and folds) which results in leakage. A collaborative study of twenty-three laboratories was conducted to compare the relative effectiveness of three scales: two forms of magnitude estimation scaling and one form of a category scale in the measurement of hedonic response to a controlled stimulus. Responses from 553 individual judges show that all scales yield hedonic measurements that are very similar in both direction and magnitude of difference between the stimuli. No scale showed any clear superiority in reliability, precision, or discrimination. Selection of a scale must be based on considerations other than the simple form of response.  相似文献

A field experiment on winter wheat in autumn 1991 investigated the effect of the rhabditid nematode, Phasmarhabditis hermaphrodita, applied to soil at five dose rates (108 - 1010 infective larvae ha-1) immediately after seed sowing, on slug populations and damage to seeds and seedlings. The nematode was compared with methiocarb pellets broadcast at recommended field rate immediately after drilling and no molluscicide treatment. Slug damage to wheat seeds and seedlings was assessed 6 and 13 wk after drilling. Seedling survival increased and slug grazing damage to seedlings declined linearly with increasing log nematode dose. These two measures of slug damage were combined to give an index of undamaged plant equivalents, which also increased linearly with increasing log nematode dose. ANOVA showed that, after 6 wk, there were significantly more undamaged plant equivalents on plots treated with the two highest nematode doses (3 × 109 and 1 × 1010 ha-1) than on untreated plots, but the number of undamaged plant equivalents on methiocarb-treated plots was not significantly greater than that on untreated plots. Slug populations were assessed by refuge trapping and soil sampling. Deroceras reticulatum was the commonest of several species of slugs recorded. During the first 4 wk after sowing, significantly more slugs were found under refuge traps on plots treated with certain doses of P. hermaphrodita than under traps on untreated plots and more showed signs of nematode infection than expected from the prevalence of infection in slugs from soil samples, suggesting that the presence of P. hermaphrodita altered slug behaviour. Application of P. hermaphrodita had no significant impact on numbers or biomass of slugs in soil during a 27 wk period after treatment, except after 5 wk when slug numbers were inversely related to log nematode dose. However, by this time, numbers in soil samples from untreated plots had declined to levels similar to those in plots treated with the highest dose of nematodes. During the first 5 wk after treatment, c. 20% of slugs in soil samples from untreated plots showed symptoms of nematode infection. It is suggested that this represented the background level of infection in the experimental field rather than spread of infection from treated plots. The apparent lack of impact of P. hermaphrodita on slug numbers and biomass in soil suggests that its efficacy in protecting wheat from slug damage was through inhibition of feeding by infected slugs.  相似文献

A method is described for using young field slugs Deroceras reticulatum (Muller) in a bioassay study of biochemical resistance of potato (Solanum tuberosum L.) cultivars to slugs. Tuber parts or an artificial diet were provided as food sources. Comparisons were made of feeding, survival and weight gain between the susceptible cultivar Maris Piper and the resistant cultivar Pentland Dell. Biochemical analyses were made of these two cultivars and the resistant cultivars Stormont Enterprise and Majestic. Comparisons of tuber sections and peelings as food sources indicated factors affecting growth were located in the surface layers of the tubers. Phenolics and glycoalkaloids were concentrated in the surface layers but the amounts were similar in the susceptible and resistant cultivars and the bioassays indicated that neither acting alone could explain resistance. The amounts and distribution of free amino acids also did not correlate with resistance although when supplied in the artificial diet they partly inhibited feeding. Proteinaceous inhibitors of slug gut proteolytic enzymes were present throughout the tubers but were not concentrated in the surface layers and the amounts were similar in the different cultivars thus they too did not explain the difference in susceptibility between the cultivars. Bioassays using acetone extracts (low molecular weight substances) and acetone powders (high molecular weight substances) either alone or in combination indicated that the resistant cultivar Pentland Dell contained a high molecular weight substance which together with a low molecular weight substance from either the same cultivar or the susceptible Maris Piper could confer resistance. Bioassays using protein extracts supplied in the presence or absence of chlorogenic acid indicated that this mechanism could comprise enzymic oxidation of phenolics. Assays of phenolase confirmed this since activity was highest in the outer layers of the tubers and was highest in the three resistant cultivars. Thus the chief resistance factor identified was high phenolase activity acting rapidly on phenolics when the slug first bites the tuber surface. The quantity of phenolics per se did not control the resistance. Thus while phenolics must be available, resistance is compatible with low blackening on cutting the tuber.  相似文献

Seedlings of Triticum aestivum L. cv. Lennox were grown in natural(late autumn) or controlled environments differing in temperature,to give plants differing either only in growth adaptation oralso in hardiness. The frost hardiness of laminae and leaf baseswas measured using an ion leakage method. Pieces of laminaeand leaf bases were freeze-fixed without chemical cryo-protectantor chemical fixative. The number of particles per unit area of the E fracture faceof the plasma membrane was substantially reduced in laminaefrom the environment inducing most hardening, compared to unhardenedcontrols. This difference in frequency of particles on the Eface was not related to adaptation of growth to low temperature(in the leaf bases) and was not an artificial consequence ofchemical treatment prior to freeze-fixation. The density ofparticles on the P face of the plasma membrane was unaffectedby growth environment. More limited data suggested the densityof particles on either fracture face of the thylakoids was alsounaffected. Unlike an earlier report, particle-free areas ofplasma membrane were not found in hardened tissue. Environment did not affect (a) the width of the ‘mouth’at the plasma membrane-plasmodesma junction in the leaf basesor (b) the size of the nuclear pores. We have examined whether acclimation to cold may involve alteredgene expression by testing for the appearance of new mRNA, capableof in vitro translation, in shoot meristems of the temperatecereal, barley, grown at 6 ?C day/2? C night. New mRNA is foundafter only 2 d in the cold, when acclimation is detectable butnot complete. The altered pattern of gene expression persistsduring subsequent growth in the cold and is associated witha number of new mRNA molecules including a major mRNA whichencodes a polypeptide Mt 77000.
