SYNOPSIS. Tetrahymena pyriformis syngen 1, mating type II, (optimal growth temperature ∼ 37 C) ordinarily dies out in 5-14 days at 0-5 C. Dying cells were lumpy, suggesting membrane damage. By supplying crude soy lecithin, survival at 0-5 C was prolonged (after growth in peptone-yeast-dextrin) to at least 22 weeks. Crude soy sterols or sitosterol or stigmasterol, and antioxidant, e.g., Ionox 330 or ascorbylpalmitate, permitted survival of cells in suspension or in growth media for at least 16-22 weeks. These sterols are known to protect against triparanol toxicity, which suggested that triparanol, which blocks cholesterol synthesis in higher animals, might enhance cold-induced injury. Triparanol was more toxic at 0–5 than at 28 C for cell suspensions and cells in growth medium; this toxicity was annulled by crude soy lecithin or β-sitosterol, the only phytosterol tested. The synthetic medium intended as a control on the crude media became toxic at 0–5 C. Protection against cold damage is discussed as a means of elucidating the role of sterols—especially phytosterols—and other lipids in maintaining the integrity of the ciliate cell membrane.  相似文献

Translocation, distribution, metabolism and photolysis of hexachlorophene (HCP) were investigated in peanut plants (Arachis hypogea L., Spanish type) grown under standardized conditions and treated with l4C-ring-labeled HCP. Treatment time ranged from 0–114 days. Autoradiographic analyses were performed on all plants. Selected plant tissues were extracted and chromatographed, using both thin layer and gas liquid chromatography. No translocation of HCP was detected in the plant tissue. No HCP metabolites were found. Some HCP was lost from the leaves and inert controls at a specific rate per unit time. The rates were slightly different, being slower on the leaves than on the controls. At the end of the 114-day treatment and based on regression analysis of thin layer chromatographic plates, an average of 68% of the applied HCP remained unaltered on the treated plants and an average of 77% remained on the controls. This indicated that, respectively, 32 and 23% of the original HCP had been altered. This 9% difference was statistically significant. Upon further analyses of the above data, using gas chromatographic methods, as many as 14 peaks were found in the treated samples and the controls, including some parent material. Ultraviolet photolysis seems to be the mechanism responsible for alteration of the HCP on the treated plants and controls. Two photolysis products have been identified by gas liquid chromatography-mass spectral analysis. Twelve other electrophylic compounds have been found in various treated plant or control extracts. Further analyses will be necessary to verify the identification and quantification of the other degradation products.  相似文献

SYNOPSIS. In chemically defined media, pH 6.4–6.8, supplementation with glycerol permitted growth equal to that with a carbohydrate supplement. In a pH 5.4 medium described in detail, containing L-glutamic acid 0.25%, supplementation with L-serine (essential for this strain), especially when further supplemented with proline, likewise permitted growth nearly equal to that with carbohydrate. These results are discussed in relation to the question of which components of peptone act as substrates and the resemblances of the Tetrahymena pattern of substrate utilization to that of metazoa.  相似文献

生殖细胞移植在生物学、畜牧学及兴起的鱼类生物工程上都有很多用途。目前,采用基于基因组的育种方法已选育出多种带有所需遗传性状的鱼类新品系;但由于缺乏鱼卵与胚胎的低温保存技术,尚未找到长期保存其种质资源的方法。就鱼类生殖细胞移植的最新研究进展进行综述,同时针对鱼类生殖细胞移植及其低温保存技术研究中存在的问题进行讨论,并提出了鱼类生殖细胞移植的应用前景,以期能积极推动鱼类生物工程的研究,加速鱼类生殖细胞移植在产业中的应用。  相似文献

SYNOPSIS. Cryoprotectants were tested in both complex and semidefined media for the trypanosomatid Crithidia fasciculata. Near log-phase or end-of-log-phase cultures were frozen for 24–48 hr at ∼ -20 C, then warmed in air to room temperature. Immediate motility was correlated with viability. The best protectant of the 83 tested was glycerol at ∼ 10% (w/v). Survival without cryoprotectant was rare. Outstanding cryoprotectants (perhaps also useful solvents for drugs poorly soluble in water) were: ethylene glycol; 2,2'-dioxyethanol (diethylene glycol); 1,2,4-butanetriol; 1,4-cyclohexanediol; dimethylsulfoxide; propylene glycol; and N -acetylethanolamine. Several sugars were active, e.g., D-arabinose, sucrose, and sorbitol. Trypanosomes tolerated cryoprotectants much less; tolerance was better in growth media than in suspension media. Trypanosoma gambiense was grown in blood-enriched media + 2-2.5% glycerol, suspended in 20% (w/v) glycerol. then frozen; this permitted 3-week survival. T. conorhini survived 4 weeks after growth in media containing glycerol 2.5%+ ethylene glycol 4%+ rutin 1.0 mg per 100 ml.  相似文献

Studies of sibling competition within brood hierarchies have rarely assessed simultaneously the effects of sex and rank in the brood hierarchy on traits other than offspring mortality and differential growth. We studied the expression of heat-shock proteins (Hsps) to assess the physiological stress response to different combinations of sex and position within competitive brood hierarchies in the black kite Milvus migrans (Bodd.), a sexually dimorphic raptor showing facultative siblicide. Senior males showed higher stress levels than did senior females and younger siblings of each sex as revealed by Hsp60 values. The analysis of Hsp70 levels indicated that nestlings from broods in which the senior chick was a male showed higher stress levels than did nestlings from broods in which the senior chick was a female. In addition, levels of Hsp60 were related negatively to nutritional condition expressed as levels of plasmatic albumin. This suggests that the sex of senior chicks may be key in determining their stress level and that of their siblings, which is probably associated with sibling competition by fighting within brood hierarchies. The comparatively higher stress levels of senior males (and their siblings) may be a consequence of their ability to exploit their potential advantage from being the head start while avoiding a possible competitive disadvantage from being the smaller sex, independent of environmental conditions determining the probability of brood reduction. Differential stress levels depending on sex and rank in the brood hierarchy may be a consequence of parental control of offspring behaviour through differential resource allocation (e.g. yolk androgens) or it may reflect adaptations of particular chicks (senior males) to enhance their competitive ability within brood hierarchies.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 88 , 383–390.  相似文献

The effect of constant and alternating temperatures on the life-history of Diglyphus isaea, as an ectoparasitoid of Liriomyza leafminers on tomato plants, was examined in the laboratory.
Parasitoid development and size were significantly affected by temperature and sex. Males showed a shorter development time and pupal size than female parasitoids. Both development time and pupil size in D. isaea differed according to the host species. Pupal size showed no consistent relationship with temperature. The lower thermal threshold for development and oviposition was determined. Fecundity with L. byroniae as host did not differ significantly between temperature regimes. Furthermore, larger females did not produce larger numbers of offspring. Development and reproduction under the alternating temperature regime did not differ from those at the comparable constant temperature, suggesting a rapid response to changes in temperature.
The intrinsic rate of increase (rm) and net reproduction (Ro) of D. isaea nearly doubled from 15°C to 25°C and 20°C, respectively, and generation time at 25°C was less than half that at 15°C. Because the parasitoid's population growth is higher than that of these pest insects at all temperatures, D. isaea is a promising candidate for seasonal inoculative biological control of Liriomyza on tomato crops in Scandinavian and western European glasshouses. Possible constraints to its effectiveness are discussed.  相似文献

Plants are altering their life cycles in response to current climatic change around the globe. More than 200 000 records for six phenological events (leaf unfolding, flowering, fruit ripening, fruit harvesting, leaf falling and growing season) of 29 perennial species for the period 1943–2003 recorded throughout Spain provide the longest temporal and the broadest spatial assessment of plant phenology changes in the Mediterranean region. The overwhelming majority of the 118 studied phenophases shifted their dates in recent decades. Such changes differed among phenological events. Leaf unfolding, flowering and fruiting are markedly advancing (?0.48, ?0.59 and ?0.32 days yr?1, respectively), but only since the mid‐1970s. Anemophilous have advanced more days their flowering than entomophilous. However, some species have delayed and others have advanced their leaf falling dates and as a result only a weak shift was observed in this event for the whole of the studied species (+0.12 days yr?1). The growing season lengthened by 18 days, which implies an increase of 8% in the life of annual leaves. Such an increase was achieved mainly through the advance of leaf unfolding dates in the spring, one of the most productive times of year for vegetation in the Mediterranean. Shifts in the plant calendar were accompanied as well by long‐term changes in the range of onset dates in 39% of studied phenophases. Leaf unfolding, flowering and growing season tended to reduce spatial variability, reflecting a faster and more synchronized onset (or duration) of phenophases across the study area. Changes in spatial variability may aggravate calendar mismatching with other trophic levels resulting from changes in dates. Because temporal responses differed markedly among species, calendar guilds of plants have changed, which suggests alterations of interspecific relationships in plant communities from Mediterranean ecosystems.  相似文献
