The health of a preimplantation embryo can be diagnosed in one or more cells biopsied from the conceptus. Here, we tried to evaluate the impact of biopsy of some trophectoderm cells from hatching mouse blastocysts on their further in vitro implantation and early egg cylinder formation. Of 374 blastocysts evaluated 112 hours after hCG, 34% initiated hatching with a small number of mural, polar, or intermediate trophectoderm cells. Half of 59 embryos that underwent induction of hatching by zona puncturing herniated some cells through this opening. After removal of cells with a glass microneedle from spontaneously hatching blastocysts, viability assessed by vital FDA staining was impaired, as well as the in vitro zona pellucida shedding and implantation. When polar trophectoderm cells were biopsied, a significantly lower number of embryos reached the egg cylinder stage.  相似文献

An unusual human retrovirus was isolated from two patients with persistent generalized lymphadenopathy who originate from West-Central Africa and are currently residing in Belgium. Although the virus shared a number of the same biological and morphological properties as human immunodeficiency retrovirus type 1 (HIV-1) and HIV-2, significant antigenic differences could be demonstrated. Several of the viral proteins also differed in molecular weight from the corresponding HIV-1 and HIV-2 proteins. Partial chemical cleavage of the most highly conserved viral proteins resulted in patterns which differed from those of HIV-1 and HIV-2. Furthermore, nucleic acid hybridization experiments were capable of discriminating between the virus types. Sequence analysis of the viral U3 region revealed a unique enhancer organization not found in other immunodeficiency viruses. The data indicated that the new isolate is more closely related to HIV-1 than to HIV-2 but clearly differs in a number of important respects.  相似文献

Long-term effects of atmospheric carbon dioxide concentration(ambient or 700 µmol mol–1) and air temperature(simulation of field conditions or + 4 C) on leaf photosyntheticrate were examined in Lolium perenne L. cv. Vigor, exposed tonatural illumination during winter. Photosynthetic capacitywas compared over a range of air temperatures and photon fluxdensities of photosynthetically active radiation which wererepresentative of winter climate (5–15 C and 0–500µmol m–2s–1), with CO2 level during measurementsimilar to that during the experimental period. Long-term exposureto increased air temperature reduced leaf CO2 fixation capacityby 23% (averaged over all measurement conditions), resultingfrom a decline in lightsaturated uptake rate, but not in incident-lightquantum efficiency. CO2- stimulation was largely absent in plantsgrown in ambient temperature, but pronounced in plants grownunder +4 C, where it compensated for two-thirds of the 23%drop. This enhancing effect of elevated CO2 level on leaf CO2uptake rate observed in the warmer treatment, was strongly dependenton measurement temperature, increasing from 5% at 5 C, to upto 32% at 15 C. Measurements of chlorophyll fluorescence anddry matter corresponded with the observed changes in assimilationcapacity, which could not be attributed to a deteriorated nitrogenstatus of the leaves as there was a similar N content on anarea basis. In a medium-sized population ofArnica montana, a threatened species in The Netherlands, the breeding system, reproductive success and genetic clonal structure were studied. Pollination experiments suggested thatA. montana is largely self-incompatible. Inbreeding depression was observed for seedling weight but not for fruit weight and germination rate. Although genetic variation is rather low in this population, the data suggest an outcrossing mating system. Analysis of the genotype of all mapped rosettes in a plot of 100 m2 indicated that dense clusters often consist of identical genotypes, suggesting a clonal structure. Open clusters frequently contained several different genotypes. This may be caused by limited fruit dispersal, since seedlings were found mainly within or in the near surroundings of the clusters.  相似文献

Agar gel electrophoresis and ultracentrifugation on continuous sucrose gradients revealed the presence of a 4S estradiol 'receptor' in cytosols of samples of human benign hyperplastic prostate tissue. High affinity (charcoal stability) and saturability (disappearance with excess competitor) binding characteristics of the molecular species concerned facilitated its clear distinction from similarly migrating serum albumin-steroid complexes. Our data, obtained with human serum, purified human albumin and albumin-enriched cytosol strongly suggest that agar gel electrophoresis, when used alone, may lack specificity for the quantification of estrogen 'receptors'. Radioligand binding to these molecules may be obscured by similarly migrating albumin-steroid complexes which fail to dissociate during electrophoresis. We advocate the inclusion of competitor experiments to improve the specificity of the method.  相似文献

Early succesion of newly created sown/unsown margin strips on ex arableland, managed by two cuttings per year with/without removal of cuttings was characterised by the replacement of annuals in favour of perennials, a steadily increase in the importance of monocots and a decrease in non N-fixing dicots. Mowing with removal of cuttings delayed this succession pattern. Sorenson's qualitative similarity index (based solely on species occurrence) revealed that species composition of the sown communities (in terms of species occurrence) became increasingly similar to the unsown plots. Furthermore convergence in vegetation composition between sown and unsown plots occurred also in terms of species importance as assessed by Sorenson's quantitative index (based on the combination of species occurrence and importance). Similarity in species importance (but not of species occurrence) was significantly enhanced by cutting with removal of cuttings. During the first 3 successional years, species diversity of sown and unsown communities converged in time, irrespective of mowing regime or location. The decrease in species diversity, number of sown wildflower species and wildflower density of sown communities was more pronounced under a mowing regime without removal of cuttings. The annual addition of mown roadside herbage significantly enhanced species richness but not the importance of dicots.  相似文献
