Glial fibrillary acidic protein-positive astrocytes, but not neurons or fibroblasts, support the differentiation of an oligodendroglial precursor cell expressing O4 antigen and vimentin into an O4 antigen-positive, but vimentin-negative oligodendrocyte. Further maturation into galactocerebroside (O1)-positive oligodendrocytes is, however, not achieved under the culture conditions used, neither in the presence of astrocytes nor neurons.  相似文献

A novel dried bacterial consortium of Lactobacillus rhamnosus yoba 2012 and Streptococcus thermophilus C106 is cultured in 1 L of milk. This fresh starter can be used for the production of fermented milk and other fermented foods either at home or at small-scale in rural settings. For the fresh starter, 1 L of milk is pasteurized in a pan that fits into a larger pan containing water, placed on a source of heat. In this water bath, the milk is heated and incubated at 85 °C for 30 min. Thereafter, the milk is cooled down to 45 °C, transferred to a vacuum flask, inoculated with the dried bacteria and left for at least 16 hr between 30 °C and 45 °C. For the purpose of frequent home production, the fresh starter is frozen into ice cubes, which can be used for the production of small volumes of up to 2 L of fermented milk. For the purpose of small-scale production in resource-poor countries, pasteurization of up to 100 L of milk is conducted in milk cans that are placed in a large sauce pan filled with water and heated on a fire at 85 °C for 30 min, and subsequently cooled to 45 °C. Next, the 100 L batch is inoculated with the 1 L freshly prepared starter mentioned before. To assure an effective fermentation at a temperature between 30 and 45 °C, the milk can is covered with a blanket for 12 hr. For the production of non-dairy fermented foods, the fresh starter is left in a cheese cloth for 12 hr, and the drained-off whey can be subsequently used for the inoculation of a wide range of food raw materials, including vegetables and cereal-based foods.  相似文献

Plant phenotypes can be affected by environments experienced by their parents. Parental environmental effects are reported for the first offspring generation and some studies showed persisting environmental effects in second and further offspring generations. However, the expression of these transgenerational effects proved context-dependent and their reproducibility can be low. Here we study the context-dependency of transgenerational effects by evaluating parental and transgenerational effects under a range of parental induction and offspring evaluation conditions. We systematically evaluated two factors that can influence the expression of transgenerational effects: single- versus multiple-generation exposure and offspring environment. For this purpose, we exposed a single homozygous Arabidopsis thaliana Col-0 line to salt stress for up to three generations and evaluated offspring performance under control and salt conditions in a climate chamber and in a natural environment. Parental as well as transgenerational effects were observed in almost all traits and all environments and traced back as far as great-grandparental environments. The length of exposure exerted strong effects; multiple-generation exposure often reduced the expression of the parental effect compared to single-generation exposure. Furthermore, the expression of transgenerational effects strongly depended on offspring environment for rosette diameter and flowering time, with opposite effects observed in field and greenhouse evaluation environments. Our results provide important new insights into the occurrence of transgenerational effects and contribute to a better understanding of the context-dependency of these effects.  相似文献

At present, the production of cement contributes approximately 8% to global carbon dioxide (CO2) emissions, whereas a further increase in the future is expected. To reduce these emissions, a prominent measure of the cement industry is to use materials such as coal fly ash (CFA) as a cement substitute, but the availability of qualified ash is limited. The development of “green cements” with lower CO2 emissions attracts attention within the cement industry and politics. Recently, a new invention for a calcium carbonate (CaCO3)‐based cement intends to use CFA as an alkaline resource during a manufacturing process based on CO2 mineralization converting anthropogenic CO2 into stable calcareous materials by using calcium‐bearing brines. In our systems analysis, we demonstrate that the alkalinity of CFA usable for mass production of cements is poor. The alkalinity of CFA primarily depends on the dissolution of free lime. For CO2 mineralization, the resulting alkalinity is just sufficient to precipitate the calcium supplied by the CFA itself, and the capacity of CO2 mineralization is low. Thus, CFA could only be used as a supplementary resource. Even at locations where exceptional calcium‐rich ash is available, very large amounts of ash must be processed. Globally, only a few million tonnes of calcium could be extracted in this way. Because qualified CFA is already used in the construction industry, the extraction of calcium from nonqualified, more polluted CFA is associated with a risk of unwanted environmental effects and implies an elaborated pollutant control.  相似文献

The expression of the neural cell adhesion molecules L1 and N-CAM and of their shared carbohydrate epitope L2/HNK-1 was studied during the development and after the transection of mouse sciatic nerves. During development, L1 and N-CAM were detectable on most, if not all, Schwann cells at embryonic day 17, the earliest stage tested. With increasing age, the immunoreactivity was reduced being confined to non-myelinating Schwann cells by post-natal day 10, at which stage the staining pattern resembled that seen in adult sciatic nerves. Double-immunolabelling experiments revealed a complete overlap between L1 and N-CAM antibodies. The L2/HNK-1 epitope was not detectable in developing sciatic nerves until the end of the 2nd post-natal week, when it appeared to be associated with the outer profiles of thick myelin sheets, as also seen in adult sciatic nerves. Three days after the transection of adult sciatic nerves, L1 antigen and N-CAM was detectable in more Schwann cells in the distal nerve end than in untreated control nerves. The peak level of the reappearance of L1 antigen and N-CAM in Schwann cells occurred between 2 and 4 weeks after transection. The reduction of L1-antigen expression to its normal adult level took more than a year, thus recapitulating normal development, but on a more protracted time scale. Similarly, the L2/HNK-1 epitope remained undetectable until the transected nerve had returned to its normal state of myelination, i.e. approximately 1 year after transection.  相似文献

Wireless photoelectrochemical (PEC) devices promise easy device fabrication as well as reduced losses. Here, the design and fabrication of a stand‐alone ion exchange material‐embedded, Si membrane‐based, photoelectrochemical cell architecture with micron‐sized pores is shown, to overcome the i) pH gradient formation due to long‐distance ion transport, ii) product crossover, and iii) parasitic light absorption by application of a patterned catalyst. The membrane‐embedded PEC cell with micropores utilizes a triple Si junction cell as the light absorber, and Pt and IrOx as electrocatalysts for the hydrogen evolution reactions and oxygen evolution reactions, respectively. The solar‐to‐hydrogen efficiency of 7% at steady‐state operation, as compared to an unpatterned ηPV of 10.8%, is mainly attributed to absorption losses by the incorporation of the micropores and catalyst microdots. The introduction of the Nafion ion exchange material ensures an intrinsically safe PEC cell, by reducing the total gas crossover to <0.1%, while without a cation exchange membrane, a crossover of >6% is observed. Only in a pure electrolyte of 1 m H2SO4, a pH gradient‐free system is observed thus completely avoiding the build‐up of a counteracting potential.  相似文献
