Summary A total of 30 cases of 46,XX true hermaphroditism was analysed for Y-DNA sequences including the recently cloned gene for male testis-determination SRY. In 3 cases, a portion of the Y chromosome including SRY was present and, in 2 cases, was localised, to Xp22 by in situ hybridisation. Since previous studies have shown that the majority of XX males are generated by an X-Y chromosomal interchange, the Xp22 position of the Yp material suggests that certain cases of hermaphroditism can arise by the same meiotic event. The phenotype in the 3 SRY-positive cases may be caused by X-inactivation resulting in somatic mosaicism of testis-determining factor expression giving rise to both testicular and ovarian tissues. Autosomal or X-linked mutation(s) elsewhere in the sex-determining pathway may explain the phenotype observed in the remaining 27 SRY-negative cases.  相似文献

Rhizobia are symbiotic soil bacteria able to intracellularly colonize legume nodule cells and form nitrogen-fixing symbiosomes therein. How the plant cell cytoskeleton reorganizes in response to rhizobium colonization has remained poorly understood especially because of the lack of an in vitro infection assay. Here, we report on the use of the heterologous HeLa cell model to experimentally tackle this question. We observed that the model rhizobium Sinorhizobium meliloti, and other rhizobia as well, were able to trigger a major reorganization of actin cytoskeleton of cultured HeLa cells in vitro. Cell deformation was associated with an inhibition of the three major small RhoGTPases Cdc42, RhoA and Rac1. Bacterial entry, cytoskeleton rearrangements and modulation of RhoGTPase activity required an intact S. meliloti biosynthetic pathway for queuosine, a hypermodifed nucleoside regulating protein translation through tRNA, and possibly mRNA, modification. We showed that an intact bacterial queuosine biosynthetic pathway was also required for effective nitrogen-fixing symbiosis of S. meliloti with its host plant Medicago truncatula, thus indicating that one or several key symbiotic functions of S. meliloti are under queuosine control. We discuss whether the symbiotic defect of que mutants may originate, at least in part, from an altered capacity to modify plant cell actin cytoskeleton.  相似文献

Intravital fluorescence microscopy (IVM) is a powerful technique for imaging multiple organs, including the brain of living mice and rats. It enables the direct visualisation of cells in situ providing a real‐life view of biological processes that in vitro systems cannot. In addition, to the technological advances in microscopy over the last decade, there have been supporting innovations in data storage and analytical packages that enable the visualisation and analysis of large data sets. Here, we review the advantages and limitations of techniques predominantly used for brain IVM, including thinned skull windows, open skull cortical windows, and a miniaturised optical system based on microendoscopic probes that can be inserted into deep tissues. Further, we explore the relevance of these techniques for the field of parasitology. Several protozoan infections are associated with neurological symptoms including Plasmodium spp., Toxoplasma spp., and Trypanosoma spp. IVM has led to crucial findings on these parasite species, which are discussed in detail in this review.  相似文献

Species and cultivar differences in boron (B) uptake at low B availability and tolerance to high external B are known for many species but mechanisms explaining such differences remain obscure. Here we contrast B uptake and distribution between two cultivars of tomato and celery that differ significantly in their susceptibility to B deficiency. The celery cultivar S48-54-1 and tomato cultivar Brittle are known to be more susceptible to B deficiency (inefficient) than the closely related cultivars Emerson Pascal and Rutgers (efficient), respectively. B uptake and distribution was also compared in two wheat lines differing in tolerance to B excess (Chinese Spring, sensitive and Lophopyrum Amphiploid, tolerant). Results showed that there is no significant difference in either the specific uptake rate (IM) of 10B or the relative growth rate (RGR) between the efficient cultivar (Emerson Pascal) and less efficient cultivar (S48-54-1) of celery. However, the distribution of 10B among plant organs (leaves, stems and roots) of Emerson Pascal was different from S48-54-1. In Emerson Pascal more than 63% of accumulated B was present in the shoots while in S48-54-1 only 45% of accumulated B was present in shoots. In tomato plants, in addition to differences in B distribution among plant organs between the efficient (Rutgers) and less efficient (Brittle) cultivars, the specific uptake rate of 10B was significantly higher in the efficient cultivar. In wheat, the tolerant line (Amphiploid) took up less B than the less tolerant cultivar (Chinese Spring), and the pattern of B distribution among plant organs was different with a greater percentage of B found in roots of Chinese Spring compared to Amphiploid. Differences in sensitivity to B deficiency and excess amongst cultivars and species were a consequence of either reduced B uptake as in wheat (Amphiploid), a restriction in B translocation from roots to shoot as in celery (S48-54-1) or a combination of both process as in tomato (Brittle).  相似文献

The physiological effects of foliar boron application (FB) on nitrogen metabolism and seed composition have not been well established in soybean [(Glycine max(L.)Merr.)]. Therefore, the effect of FB on nitrogen metabolism and seed composition was investigated. Nitrate assimilation was evaluated by measuring nitrate reductase activity (NRA) and nitrogen fixation was evaluated by measuring nitrogenase activity and natural abundance of 15N/14N. NRA were significantly (P?≤?0.05) higher in plants that received FB than the control plants. Higher rate of FB (One application of four times of commercial rate) inhibited nitrogen fixation as measured by natural abundance of 15N/14N ratio, but increased NRA. The higher activities of NR and nitrogenase by FB were accompanied with a higher B concentration in leaves. The significant (P?<?0.0001) enrichment of 15N/14N, accompanied with a higher rate of FB, suggested a possible mechanism where nitrate assimilation may compensate for the decrease in nitrogen fixation. FB increased seed protein by 13.7% and oleic acid by 30.9% compared to the control plants. This alteration was accompanied by a higher B concentration in leaves and seed. The results suggest that FB affects nitrogen metabolism and alters seed compositions, especially protein and unsaturated fatty acids.  相似文献

Purpose

Life Cycle Assessment (LCA) is the process of systematically assessing impacts when there is an interaction between the environment and human activity. Machine learning (ML) with LCA methods can help contribute greatly to reducing impacts. The sheer number of input parameters and their uncertainties that contribute to the full life cycle make a broader application of ML complex and difficult to achieve. Hence a systems engineering approach should be taken to apply ML in isolation to aspects of the LCA. This study addresses the challenge of leveraging ML methods to deliver LCA solutions. The overarching hypothesis is that: LCA underpinned by ML methods and informed by dynamic data paves the way to more accurate LCA while supporting life cycle decision making.

Methods

In this study, previous research on ML for LCA were considered, and a literature review was undertaken.

Results

The results showed that ML can be a useful tool in certain aspects of the LCA. ML methods were shown to be applied efficiently in optimization scenarios in LCA. Finally, ML methods were integrated as part of existing inventory databases to streamline the LCA across many use cases.

Conclusions

The conclusions of this article summarise the characteristics of existing literature and provide suggestions for future work in limitations and gaps which were found in the literature.

Plasmodium falciparum virulence is linked to its ability to sequester in post‐capillary venules in the human host. Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) is the main variant surface antigen implicated in this process. Complete loss of parasite adhesion is linked to a large subtelomeric deletion on chromosome 9 in a number of laboratory strains such as D10 and T9‐96. Similar to the cytoadherent reference line FCR3, D10 strain expresses PfEMP1 on the surface of parasitized erythrocytes, however without any detectable cytoadhesion. To investigate which of the deleted subtelomeric genes may be implicated in parasite adhesion, we selected 12 genes for D10 complementation studies that are predicted to code for proteins exported to the red blood cell. We identified a novel single copy gene (PF3D7_0936500) restricted to P. falciparum that restores adhesion to CD36, termed here virulence‐associated protein 1 (Pfvap1). Protein knockdown and gene knockout experiments confirmed a role of PfVAP1 in the adhesion process in FCR3 parasites. PfVAP1 is co‐exported with PfEMP1 into the host cell via vesicle‐like structures called Maurer's clefts. This study identifies a novel highly conserved parasite molecule that contributes to parasite virulence possibly by assisting PfEMP1 to establish functional adhesion at the host cell surface.  相似文献

Many biological processes are mediated by protein-protein interactions (PPIs). Because protein domains are the building blocks of proteins, PPIs likely rely on domain-domain interactions (DDIs). Several attempts exist to infer DDIs from PPI networks but the produced datasets are heterogeneous and sometimes not accessible, while the PPI interactome data keeps growing.We describe a new computational approach called “PPIDM” (Protein-Protein Interactions Domain Miner) for inferring DDIs using multiple sources of PPIs. The approach is an extension of our previously described “CODAC” (Computational Discovery of Direct Associations using Common neighbors) method for inferring new edges in a tripartite graph. The PPIDM method has been applied to seven widely used PPI resources, using as “Gold-Standard” a set of DDIs extracted from 3D structural databases. Overall, PPIDM has produced a dataset of 84,552 non-redundant DDIs. Statistical significance (p-value) is calculated for each source of PPI and used to classify the PPIDM DDIs in Gold (9,175 DDIs), Silver (24,934 DDIs) and Bronze (50,443 DDIs) categories. Dataset comparison reveals that PPIDM has inferred from the 2017 releases of PPI sources about 46% of the DDIs present in the 2020 release of the 3did database, not counting the DDIs present in the Gold-Standard. The PPIDM dataset contains 10,229 DDIs that are consistent with more than 13,300 PPIs extracted from the IMEx database, and nearly 23,300 DDIs (27.5%) that are consistent with more than 214,000 human PPIs extracted from the STRING database. Examples of newly inferred DDIs covering more than 10 PPIs in the IMEx database are provided.Further exploitation of the PPIDM DDI reservoir includes the inventory of possible partners of a protein of interest and characterization of protein interactions at the domain level in combination with other methods. The purpose of this article is to illustrate a new classification of resin based aesthetic materials laying on the characterization of their matrix and their filler morphology. Four samples per material have been prepared for SEM evaluation. Each sample has been treated with chloroform to dissolve its matrix in order to evidence the filler morphology. A general schema of four different matrix systems which characterize the material's level of hydrophobicity can be put in evidence. The subsequent filler analysis individuates a more complex schema based on filler size and construction. A new classification based on matrix nature and filler morphology has been proposed. Based on this concept mechanical and aesthetic characteristics of the materials can be presumed.  相似文献
