Laboratory scale anaerobic upflow filter, sludge blanket and hybrid bed reactors were operated for 860 days in the treatment of high ammonia landfill leachate. Organic loading was gradually increased from 1.3 to 23.5 kg COD/m3 day in the start-up period and then fluctuated according to the COD concentration of raw leachate. To prevent free ammonia inhibition, influent pH was reduced to 4.5 after Day 181 and consequently COD removal efficiencies above 80% were achieved in all reactors. However, the anaerobic filter and hybrid bed reactor were generally found slightly more efficient and stable than the UASB reactor. In addition to conventional anaerobic reactor control parameters, the complementary techniques of denaturing gradient gel electrophoresis (DGGE), cloning and fluorescent in situ hybridization (FISH) were used to identify and compare the microbial profiles in the reactors at Day 830. Molecular analyses revealed that acetoclastic Methanosaeta species were prevalent in all reactors and configuration did not have an impact on microbial diversity in the long-term.  相似文献

Ethanol causes oxidative stress and tissue damage. The aim of this study was to investigate the effect of antioxidant carnosine on the oxidative stress induced by ethanol in the rat brain tissue. Forty male rats were divided equally into four groups as control, carnosine (CAR), ethanol (EtOH), and ethanol plus carnosine (EtOH + CAR). Rats in the control group (n = 10) were injected intraperitoneally (i.p.) with 0.9% saline; EtOH group (n = 10) with 2 g/kg/day ethanol, CAR group (n = 10) received carnosine at a dose of 1 mg/kg/day and EtOH + CAR group (n = 10) received carnosine (orally) and ethanol (i.p.). All animals were sacrificed using ketamine and brain tissues were removed. Malondialdehyde (MDA), protein carbonyl (PCO) and tissue carnosine levels, and superoxide dismutase (SOD) activities were measured. Endogenous CAR levels in the rat brain tissue specimens were significantly increased in the CAR and EtOH groups when compared to the control animals. MDA and PCO levels in the EtOH group were significantly increased as compared to the other groups (P < 0.05). CAR treatment also decreased MDA levels in the CAR group as compared to the control group. Increased SOD activities were obtained in the EtOH + CAR group as compared to the control (P < 0.05). CAR levels in the rat brain were significantly increased in the CAR, EtOH and CAR + EtOH groups when compared to the control animals. These findings indicated that carnosine may appear as a protective agent against ethanol-induced brain damage.  相似文献

Tuz Lake is a hypersaline lake located in Central Anatolia, Turkey. The lake and its salterns, Kaldirim and Kayacik, are the major sources of solar salt for industrial applications in Turkey, especially in the food and leather industries. Use of the crude solar salt often results in microbial deterioration of the products. We therefore initiated a thorough characterization of the microbial communities in Tuz Lake and its adjacent salterns, and we present here the results of investigations on diversity of extremely halophilic Archaea. Twenty-seven colonies of aerobic red or pink Archaea (family Halobacteriaceae) were selected according to colony shape, size, consistency and pigmentation, and characterized according to their phenotypic characteristics, polar lipid contents, and antibiotic sensitivities. Furthermore, 16S rRNA genes of the isolates were screened by DGGE analysis and partially sequenced. Phylogenetic analysis showed that most isolates belonged to the genera Haloarcula, Halorubrum and Halobacterium. Haloarcula was found to be dominant both in Tuz Lake and in the saltern samples. Halorubrum species were isolated from Tuz Lake and from the Kaldirim saltern, and Halobacterium species were recovered from Tuz Lake and from the Kayacik saltern. All strains showed various activities of hydrolytic enzymes (proteases, amylases, cellulases, and others), activities which are responsible for the detrimental effects of the crude salt in food and leather products.  相似文献

Inhibitory effects of free ammonia on Anammox bacteria   总被引:2,自引:0,他引:2  
Anammox bacteria can effectively treat high ammonia and nitrite concentrations under anoxic environments. However, the presence of high ammonia and nitrite concentrations may cause free ammonia and nitrous acid inhibition at high pH and temperature environments. In this study, the inhibitory effect of free ammonia on Anammox bacteria was investigated in a lab-scale upflow fixed-bed reactor with Kaldnes biofilm carriers. Results of continuous operation showed that inhibition was not observed in the Anammox reactor when the free ammonia concentration gradually increased up to 150?mg/L. However, Anammox activity suddenly dropped to 10 % when the free ammonia concentration reached to 190?mg/L. Nevertheless, high influent ammonia and nitrite concentrations up to 1,500 and 500?mg/L, respectively, did not noticeably inhibit the Anammox activity. Gradually decreasing Anammox activity was also supported by fluorescent in situ hybridization (FISH) analysis. FISH and 16S rRNA gene analysis results revealed that main Anammox organisms were phylogenetically related to Candidatus Kuenenia stuttgartiensis, Candidatus Jettenia asiatica and Candidatus Brocadia anammoxidans.  相似文献

Leachate treatment is a challenging issue due to its high pollutant loads. There are several studies on feasible treatment methods of leachate. In the scope of this study, high organic content of young leachate was eliminated using an upflow anaerobic sludge blanket (UASB) and a membrane bioreactor (MBR) in sequence and effluent of the system was given to single reactor for high activity ammonia removal over nitrite (SHARON) and anaerobic ammonia oxidation (Anammox) reactors to remove nitrogen content. All reactors were set up at lab scale in order to evaluate the usage of these processes in sequencing order for leachate treatment. COD and TKN removal efficiencies were over 90 % in the combined processes which were operated during the study. The biodegradable portion of organic matter was removed with an efficiency of 99 %. BOD5 concentration decreased to 50 mg/L by UASB and MBR in sequence even the influent BOD5 concentration was over 8,000 mg/L. Although high nitrogen concentrations were observed in raw leachate, successful removal of nitrogen was accomplished by consecutive operations of SHARON and Anammox reactors. The results of this study demonstrated that with an efficient pretreatment of leachate, the combination of SHARON–Anammox processes is an effective method for the treatment of high nitrogen content in leachate.  相似文献

Tuzkoy is a large and unpolluted salt mine in Central Anatolia, Turkey. High quality salt in this mine may be supplied to the table salt market after a simple process instead of complicated refining. However, the extremely halophilic microbial content of the salt has not yet been investigated which may result in detrimental effects in industrial processes such the food and leather industries, if unprocessed salt is directly used. Therefore, to identify and characterize the microbial contaminants in Tuzkoy salt, chemical and microbial analyses were conducted on salt crystal samples collected from three different locations of the mine. Generally 105–106 colony forming units of extremely halophilic microorganisms were detected per gram of salt samples. Twelve colonies were selected randomly for further characterization. Phenotypic characterization, lipid analysis, antibiotic susceptibility tests and positive PCR amplification results with Archaea-specific primers confirmed that all strains were halophilic Archaea belong to family Halobacteriaceae. According to denaturing gradient gel electrophoresis (DGGE) results, 10 comparatively different strains were selected for DNA sequencing. DNA sequences and phylogenetic analyses revealed that the isolated strains were mainly species of the genera Halobacterium, Haloarcula, Natrinema and Halorubrum. In addition enzymatic activity tests were also conducted to evaluate the salt quality for industrial applications. Results of gelatinase, caseinase, amylase, cellulase and lipase activity tests revealed that the isolated strains produced hydrolytic enzymes, which could cause deterioration in salt-treated food and hide. It may be pointed out that cellulase activity in halophilic Archaea has not been reported previously. Moreover, β-galactosidase activity has been reported in some Haloferax and Halorubrum species but not in the genus Halobacterium. For a successful nitrogen removal, Anammox process needs to be established in line with a stable partial nitritation pretreatment unit since wastewater influent is mostly unsuitable for direct treatment by Anammox. Partial nitritation is, however, a critical bottleneck for the nitrogen removal since it is often difficult to maintain the right proportions of NO2-N and NH4-N during long periods of time for Anammox process. This study investigated the potential of Anammox-zeolite biofilter to buffer inequalities in nitrite and ammonium nitrogen in the influent feed. Anammox-zeolite biofilter combines the ion-exchange property of zeolite with the biological removal by Anammox process. Continuous-flow biofilter was operated for 570 days to test the response of Anammox-zeolite system for irregular ammonium and nitrite nitrogen entries. The reactor demonstrated stable and high nitrogen removal efficiencies (approximately 95 %) even when the influent NO2-N to NH4-N ratios were far from the stoichiometric ratio for Anammox reaction (i.e. NO2-N to NH4-N ranging from 0 to infinity). This is achieved by the sorption of surplus NH4-N by zeolite particles in case ammonium rich influent came in excess with respect to Anammox stoichiometry. Similarly, when ammonium-poor influent is fed to the reactor, ammonium desorption took place due to shifts in ion-exchange equilibrium and deficient amount were supplied by previously sorbed NH4-N. Here, zeolite acted as a preserving reservoir of ammonium where both sorption and desorption took place when needed and this caused the Anammox-zeolite system to act as a buffer system to generate a stable effluent.  相似文献
