Samples of microphytobenthos from the Tagus estuary were analysed for photosynthetic pigments by spectrophotometry and High
Performance Liquid Chromatography (HPLC). Chlorophyll a values obtained with HPLC and spectrophotometry methods presented a highly significant positive correlation for both spectrophotometric
methods used (with and without the correction for pheopigments), but this relationship depended on the type of sediment. We
concluded that spectrophotometric methods give reliable Chl-a values, being suited for routine analysis, when a vast number of replicates is needed. However, for the correct estimation
of pheopigments, HPLC analysis is indispensable. In the literature, Chl-a estimations are expressed per content (μg g−1) or concentration (mg m−2). We discuss the influence of sediment type on the results depending on the type of unit used, and propose a simple conversion

Samples of the microalgaBotryococcus braunii were submitted to supercritical fluid extraction with carbon dioxide at 40 °C and pressures of 12.5, 20.0 and 30.0 MPa. The extraction yield and the fraction of the hydrocarbons in the extracts both increased with pressure and at 30 MPa these compounds were obtained rapidly. This behaviour is associated with the localization of the hydrocarbons outside the cell wall. We monitored one group of muriquis, or woolly spider monkeys (Brachyteles arachnoides), over a 9-year period at Fazenda Montes Claros, Minas Gerais, Brazil. The group grew from 22 to 42 individuals due to the births of 21 surviving infants. Eight immigrations involving immature females were offset by emigrations and disappearances. The home range of the group expanded as the group size increased. The group traveled as a cohesive unit during the first 6 years of the study, but recently it has begun to show greater tendencies to fission temporarily into smaller subgroups. Six adult males from the other muriqui group at this site have simultaneously increased their associations with the main study group. These observations indicate that the group is in a state of transition which may lead, ultimately, either to its division into two smaller units or to a more fluid social structure. 相似文献

Xylella fastidiosa is a Gram-negative xylem-limited plant pathogenic bacterium responsible for several economically important crop diseases. Here, we present a novel and efficient protein refolding protocol for the solubilization and purification of recombinant X. fastidiosa peptidoglycan-associated lipoprotein (XfPal). Pal is an outer membrane protein that plays important roles in maintaining the integrity of the cell envelope and in bacterial pathogenicity. Because Pal has a highly hydrophobic N-terminal domain, the heterologous expression studies necessary for structural and functional protein characterization are laborious once the recombinant protein is present in inclusion bodies. Our protocol based on the denaturation of the XfPal-enriched inclusion bodies with 8M urea followed by buffer-exchange steps via dialysis proved effective for the solubilization and subsequent purification of XfPal, allowing us to obtain a large amount of relatively pure and folded protein. In addition, XfPal was biochemically and functionally characterized. The method for purification reported herein is valuable for further research on the three-dimensional structure and function of Pal and other outer membrane proteins and can contribute to a better understanding of the role of these proteins in bacterial pathogenicity, especially with regard to the plant pathogen X. fastidiosa. 相似文献
