利用52个家蚕品种在春秋两个季节的试验资料,对万头茧层量及其构成因素作了通径分析。结果表明:不同环境下,诸因素对万头茧层量的作用不完全相同,主要表现为幼虫生命率和茧层率在不同季节中的作用相对地变化。结合育种实践指出:(1)春用蚕品种的选育应在保证生命率的前提下努力提高茧层率和全茧量,而夏秋用品种的选育应在保证茧层率的前提下努力提高生命率和全茧量。 (2)为提高选择效果,春蚕期育种应以选择茧层率为主,夏秋期育种应以选择生命率为主。  相似文献

Summary Infection of E. coli with the viruses T7 or T3 leads to a dramatic efflux of potassium ions. This ion efflux is caused by the virus particle since no concomitant protein synthesis is required. 影响农杆菌介导的柑桔基因转化因素研究   总被引:10,自引:0,他引:10  
以柑桔( Citrus) 中的枳壳、甜橙、柠檬、四季桔上胚轴为对象, 进行了农杆菌介导的柑桔衰退病病毒外壳蛋白基因转化影响因素研究, 最终将衰退病病毒外壳蛋白基因转入枳壳。研究结果表明: 以卡那霉素为选择试剂, 且外植体水平放置时, 枳壳的选择浓度为50μg/mL, 其它品种为20 ~30μg/mL。外植体与农杆菌的共培养时间3 d 最好, 70 ~100μmol/L的酚类化合物(As) 的添加对外植体Gus 瞬时表达阳性率有明显促进作用。转化所用外植体年龄以20 d 最好, 品种之间抗性芽产生率与Gus 阳性芽产生率明显不同。以质粒DNANcoⅠ酶切产物为探针, 进行了Southern blot 分析, 结果表明外源基因已稳定整合到了枳壳植株的核基因组中。  相似文献

Prion diseases are conformational diseases, many factors are involved in altering the conformation of prion, such as RNA, DNA, pH, and copper etc. However the neurotoxic mechanism of prion diseases is not clear yet. The aim of this study is to investigate the effect of the nucleoprotein complex of RNA and recombinant ovine prion protein (OvPrPC) on the cultured rat cortical neurons in vitro. Our previous study revealed that the nucleoprotein complex (OvPrPC-RNA) is characterized with high β sheet conformation and proteinase K resistance. Here we found that the OvPrPC-RNA induced marked neuronal cell death by the MTT (3-(4,5-dimethyl-thiazole -2-yl)-2,5-diphenyl –tetrazolium bromide) and TUNEL (TdT mediated biotin-dUTP nicked-end labeling) assay, and the neurotoxic effects were confirmed by testing the content of Bcl-2 Associated X protein (Bax) in the immunoprecipitation assay and Western blot assay. Compared to the control group, there is no significant difference of active Bax or total Bax after RNA alone treatment or OvPrPC alone treatment, but the OvPrPC-RNA induced significant increases of active Bax level, while the contents of total Bax had no obvious changes after OvPrPC-RNA treatment. The results suggested that OvPrPC-RNA is neurotoxic in vitro, which added further evidence to the current understanding of mechanism of cellular injury by RNA molecules for transformation of the PrPC to PrPSc.  相似文献

土壤盐渍化严重影响了甜土植物(特别是农作物)的生长速率和产量。植物根部是与土壤中Na^+接触的重要界面,也是吸收Na^+的主要器官,正日益成为甜土植物耐盐研究靶标。本文概述近10年来对Na^+进入甜土植物根部的2种途径(共质体和质外体)以及根部维持离子稳态的3种应对方式(Na^+的细胞外排、液泡区隔化和地上部分转运)新的认识和观点。通过阐述Na^+在甜土植物根部的转运机制,以期为提高农作物抗盐性以及在盐渍环境中的产量提供理论基础。  相似文献

研究了阿维菌素对广赤眼蜂不同发育阶段的毒性和实验种群动态的影响.结果表明,阿维菌素田间推荐剂量喷雾处理可显著降低米蛾卵内广赤眼蜂的羽化率;除卵期外,其它3种发育阶段经该药处理,羽化后的雌蜂寿命显著缩短,其中预蛹和蛹期处理雌蜂前翅畸形率高达43.3%~47.2%,影响程度随着其生长发育的进行呈增大趋势.根据药膜法剂量与效应测定结果,广赤眼蜂成蜂对阿维菌素比较敏感,LC50值为2.1991mg·L-1.温室盆栽试验表明,1.8%阿维菌素乳油对广赤眼蜂成蜂的持效期为10d,具有一定程度的残留毒性.广赤眼蜂的蛹经阿维菌素亚致死剂量LC35处理后,发育至成蜂,除羽化后的雌蜂产卵量显著减少外,雌蜂寿命、子代羽化率和子代性比与对照并无显著差异.生命表参数显示,LC35处理后的广赤眼蜂净增值率和世代平均历期明显地低于对照,但内禀增长率、周限增长率和种群倍增时间与对照差异不明显.可以认为阿维菌素亚致死剂量对广赤眼蜂的种群没有重要影响.  相似文献

Dectin‐1, a specific pattern recognition receptor for β‐1,3/β‐1,6‐glucans, is expressed mainly on phagocytes. Human dectin‐1 (hDectin‐1) and mouse dectin‐1 (mDectin‐1) were separately expressed on HEK293 cell surfaces for examination of the binding abilities of a synthetic particulate β‐glucan (pβG), a product extracted from Saccharomyces cerevisiae, in this study. The binding of zymosan‐FITC to hDectin‐1 and mDectin‐1 was inhibited by pβG at similar concentrations for 50% inhibition of binding (IC50). However, the kinetics of the time course and dose response to zymosan stimulation observed for U937 and J774A.1 differed. Superoxide anion production was increased in U937 but reduced in J774A.1 when cells were treated with pβG, zymosan, or laminarin, whereas ovalbumin endocytosis was enhanced in U937 and J774A.1 treated either with pβG, zymosan, laminarin, or barley‐glucan. These results indicate that the binding affinity of pβG to hDectin‐1 is similar to the binding affinity to mDectin‐1, and that stimulation by pβG as well as various forms of β‐1,3‐glucans on U937 and J774A.1 resulted in upregulation of cell activity and ovalbumin endocytosis. Additionally, other coreceptors on U937 and J774A.1 may be involved in directing different responses to superoxide anion production in these two types of cells. These results will likely contribute to further investigations on identifying the biological forms of β‐glucans capable of binding its specific receptor as the effective immunomodulators. © 2010 American Institute of Chemical Engineers Biotechnol.
