Characteristics of ribosomal RNA synthesis in the blood lymphocytes of chronic lympholeukemia patients

A study was made of the number of silver grains over the nucleoli and of the content of ribosomes in the lymphocyte cytoplasm for six healthy persons and for 20 patients with chronic lymphatic leukemia. Besides ratios of compact, nucleolonemic and ring types of nucleoli were evaluated in addition to counts of the specific radioactivity of mature 28S rRNA in lymphocytes. In the majority of cases examined, cells with 1 or 2 nucleoli of compact and nucleolonemic types were seen dominating. The number of silver grains over the nucleoli in the control healthy persons did not differ from that in patients who did not receive any treatment, which contrasted with high value grain counts in the treated patients. The lymphocyte ribosome contents varied within the normal and decreased values in both the patient groups. The specific radioactivity in 28S rRNA leukemic lymphocytes was significantly lower in groups of patients with low ribosome contents than in those with the normal ones. The data suggest that in the leukemic cells with a high or unaltered activity of ribosome cistrons and low ribosome levels rRNA processing is broken.     

Complementary addressed alkylation of Escherichia coli 16S rRNA with 2',3'-O-[4-N-methyl-N-(2-chloroethyl)aminobenzylidene]m derivatives of oligodeoxyribonucleotides. IV. Determination of binding sites of 16S rRNA with benzylidene derivatives of d(pACCTTGTT)rA, d(pTTACGACT)rU, d(TTTGCTCCCC)rA

By site-directed alkylation of 16S rRNA with benzylidene derivatives of d(pACCTTGTT)rA (II), d(pTTACGACT)rU (III), d(pTTTGCTCCCC)rA (IV) (reagents (II)--(IV] followed by the RNase H treatment a number of 16S rRNA fragments have been obtained. Hybridisation of these fragments with restriction fragments of plasmid pKK 3535, containing operon rrnB of E. coli rRNAs, led to the identification of all reagents' binding sites in 16S rRNA. Good correlation is found between estimated stability of non-perfect 16S rRNA.oligodeoxyribonucleotide duplexes and the level of modification of this site with alkylating derivative of the same oligodeoxyribonucleotide. With high concentration of the reagents (II)--(IV) ((2-5) x 10(-5) M) the site-directed alkylation proceeds not only at the desired site but also at other sites corresponding to non-perfect duplexes between 16S rRNA and the reagents. It should be noted that the modification mainly occurs in the non-perfect duplexes, carrying mismatched bases at the termini. Influence of the secondary structure of 16S rRNA on the site-directed modification is discussed.     

The sperm structure of the gall-midges Anaretella and Lestremia (Insecta, Diptera, Cecidomyiidae)

The sperm structure of some dipteran flies belonging to the Lestremiini tribe have been examined. Anaretella cincta was shown to have an axoneme made of 20-21 microtubular doublets, disposed in a circle in a cross section and surrounding a mitochondrion. Other crystal-containing mitochondria flank the axoneme; a second species (Anaretella sp.) was provided with 21-22 axonemal doublets. Lestremia is characterized by a flattened axoneme, consisting of about 150 doublets arranged in 2 antiparallel rows and surrounding a few mitochondria. These mitochondria, in Lestremia sp., have a crystalline core that is missing in Lestremia cinerea. The structure of microtubular doublets is quite similar in the 2 related genera and a derivation of the flattened axoneme found in Lestremia from that circular of Anaretella is suggested. Sperm structure suggests that Lestremia cinerea is not a uniform species.

Occurrence and social structure of Baird's beaked whales,Berardius bairdii,in the Commander Islands,Russia

The social structure of Baird's beaked whales is completely unstudied, and it is unknown if either females or males form long‐term social associations or occur in stable groups. In this paper we summarize our observations of individually identified animals over the span of 6 yr to provide insight on their long‐term social structure. We have identified 122 whales, with 28 of them encountered three times or more and thus included in the analysis of social structure. We found that the whales exhibited nonrandom patterns of social associations with some individuals preferentially associating with each other. Whales with more scarred skin had higher maximum association coefficients, which indicates that older animals and/or males were more inclined to form stable associations. Cluster analysis with a modularity test for gregariousness divided the whales into four clusters. Whales from the same clusters did not always occur together, but some individuals retained stable associations over several years. The strength of social relationships decayed over periods of months, with between‐year relationships showing little deviation from what would be expected if association was random. Generally these findings do not correspond to a stable society with fixed groups but instead suggest a fission‐fusion society with some stable alliances.     

Immunostimulating and anticoagulating activity of fucoidan from brown algae Fucus evanescens of Okhotskoe sea]

Fucoidan--nontoxic sulfated polysaccharide was isolated from brown algae Fucus evanescens in Okhotskoe Sea. Chemical analysis of the compound was performed, it was shown that fucoidan is freely soluble in water and acid solutions. Immunotropic and anticoagulating properties of the compound were evaluated in comparison with heparin. It was demonstrated that fucoidan in wide range of doses stimulated phagocytic and bactericidic activity at leucocytes of mice peritoneal exudate. Heparin on the contrary demonstrated depressive effect on these functions at high dose. It was shown that fucoidan has dose-dependent anticoagulating activity in vitro and in vivo comparable with heparin activity. The results of investigation demonstrated possibility of fucoidan application as immunomodulating and anticoagulating agent of plant origin.
