Fifteen polymorphic microsatellite DNA loci were isolated from barn swallow (Hirundo rustica) and optimized for future studies of radiation‐induced mutations in populations from Ukraine. The loci were screened for variability among 25 individuals from two populations. The primers amplified loci with relatively high numbers of alleles ranging from five to 32 alleles per locus and polymorphic information content from 0.481 to 0.951. Observed heterozygosity varied from 0.458 to 0.960. None of the loci showed deviations from Hardy–Weinberg equilibrium in either population.  相似文献

Abstract. Two-year-old potted sweet chestnut seedlings were grown at 350 ppm CO2 and 700 ppm, day and night in constantly ventilated tunnels during two full growing seasons, near Paris, France (48° N, 2° E). Enrichment with CO2 caused an unusual shoot growth response. After the end of July, stem elongation ceased in 62% of the CO2 enriched plants as compared with 37% in the control. The leaves of CO2-enriched seedlings showed early senescence, indicated by premature yellowing and a decrease in chlorophyll content. This was associated with nutrient dilution brought about by the rapid growth of these trees. The increase in total dry weight of the CO2-enriched seedlings was essentially the result of increase in the root dry weight (69%). Shoot weight decreased by 22% relative to the control. Total leaf area per enriched plant was 25% smaller than the control. This unusual pattern of growth and carbon allocation of the CO2 treated Chestnut trees emphasizes the concept of a response specificity within trees to an increase of atmospheric CO2.  相似文献

Elevated CO2 decreases seed germination in Arabidopsis thaliana   总被引:2,自引:0,他引:2  
The impact of elevated [CO2] on seed germination was studied in different genotypes of Arabidopsis thaliana from natural populations. Two generations of seeds were studied: the maternal generation was produced in the greenhouse (present-day conditions), the offspring generation was produced in two chambers where the CO2 concentration was either the present atmospheric concentration (about 350 ppm) or elevated (700 ppm). The seeds were tested for proportion of germinated seeds and mean germination time in both chambers to study the impact of elevated [CO2] during seed production and germination. Elevated [CO2] during maturation of seeds on the mother-plants decreased the proportion of germinated seeds, while elevated [CO2] during germination had no effect on the proportion of germinated seeds. However, when seeds were both produced and germinated under elevated [CO2] (situation expected by the end of next century), germination was slow and low. Moreover, the effect of the [CO2] treatment differs among genotypes of Arabidopsis: there is a strong treatment × genotype interaction. This means that there is ample genetic variance for a selective response modiying the effects of high levels of [CO2] in natural populations of Arabidopsis thaliana. The outcome at the community level will depend on what seeds are available, when they germinate and the resulting competition following germination.  相似文献

Extreme environmental perturbations are rare, but may have important evolutionary consequences. Responses to current perturbations may provide important information about the ability of living organisms to cope with similar conditions in the evolutionary past. Radioactive contamination from Chernobyl constitutes one such extreme perturbation, with significant but highly variable impact on local population density and mutation rates of different species of animals and plants. We explicitly tested the hypothesis that species with strong impacts of radiation on abundance were those with high rates of historical mutation accumulation as reflected by cytochrome b mitochondrial DNA base‐pair substitution rates during past environmental perturbations. Using a dataset of 32 species of birds, we show higher historical mitochondrial substitution rates in species with the strongest negative impact of local levels of radiation on local population density. These effects were robust to different estimates of impact of radiation on abundance, weighting of estimates of abundance by sample size, statistical control for similarity in the response among species because of common phylogenetic descent, and effects of population size and longevity. Therefore, species that respond strongly to the impact of radiation from Chernobyl are also the species that in the past have been most susceptible to factors that have caused high substitution rates in mitochondrial DNA.  相似文献

A knowledge of genetic structure in natural populations is often necessary for conservation and management purposes, especially in declining Pacific salmon populations. To test for genetic differentiation between nine populations of chinook salmon, Oncorhynchus tshawytscha, from south-western British Columbia, Canada, DNA was extracted from 603 fish and hybridized with a single-locus minisatellite probe. Multivariate statistical analyses of the resulting allele size data permitted successful overall population identification of 52% (individual population range: 24–78%; P < 0.005), indicating a high level of genetic differentiation among the nine populations. Two of the nine populations were further analysed using data from a second minisatellite locus. The discrimination success rate improved from 81.1% (one-locus analyses) to 90.0% (two-locus analyses), indicating the potential for greatly increased resolution gained by the addition of more loci. These results indicate that variation at minisatellite loci can be used for assessing population-level genetic structure, even with artificial gene flow.  相似文献

Abstract A method is described in which 1 year-old chestnut coppice was fed in situ with air highly enriched in 13CO2 (23%). After 3 days, 13C concentration increases in shoots were measured by mass spectrometry. Respiratory losses between 13C feeding and harvest were estimated using two different methods: (i) a model involving the temperature response of respiration and (ii) direct measurement of 13C content of the CO2 respired by the shoots during the night. Carbon allocation to roots was deduced by subtracting from the given amount of 13C, the amount remaining in shoots and the 13C respired by the shoots. The method was tested twice during the growing season. Very little carbon was allocated to roots in late July, but over 80% of assimilated 13C went to roots at the end of September. Despite some approximations in the 13C respiratory losses estimations, the method allowed evaluation of carbon allocation to roots with an error of about 5%.  相似文献

Individuals ofArabidopsis thaliana, collected in different naturalpopulations, were grown in controlled and elevated CO2in a glasshouse.Following germination, root growth of progeny of different linesof these populations was studied in control and elevated atmosphericCO2. No significant direct effect of atmospheric CO2concentrationcould be demonstrated on root growth. An important parentaleffect was apparent, namely that root length and branching weredecreased in seeds collected from a mother plant which had beengrown in elevated CO2. This was correlated with smaller seeds,containing less nitrogen. These parental effects were geneticallyvariable. Senescence is the deterioration of the phenotype with age caused by negative effects of mutations acting late in life or the physiological deterioration of vital processes. Birds have traditionally been assumed to senescence slowly despite their high metabolic rates, high blood sugar levels and high body temperature. Here we investigate the patterns of age‐related performance of sperm of a long distance migrant, the barn swallow Hirundo rustica, varying in age from 1 to 6 years, analysed by the computer‐assisted sperm analysis equipment. Sperm showed deteriorating performance in terms of linear movement, track velocity, straight line velocity and reduced proportions of rapidly moving, progressive and motile sperm with age. In a second series of experiments, we assessed performance of sperm from the same males in neutral medium and in medium derived from the reproductive tract of females in an attempt to test if sperm of old males performed relatively better in female medium, as expected from extra‐pair paternity being negatively related to male age, but not to female age. Older males showed consistently better performance in female medium than in neutral medium in terms of track velocity, straight line velocity and reduced proportions of rapidly moving, progressive and motile sperm, whereas young males showed better performance in neutral medium. These results provide evidence of declining sperm performance for important reproductive variables not only with age, but also with the sperm of old males performing differentially better in female medium than young males.  相似文献
