WHEN chromosomes pair at meiosis the bivalents so formed do not normally interlock. Heat-treatments can, however, induce bivalent interlocking in the locust Locusta migratoria. Only the longest bivalents interlock and usually only two are found per cell; two “rod” bivalents, with single chiasmata, two “ring” bivalents, each with two or three chiasmata, or one “rod” and one “ring” bivalent (Fig. 1a, b and c). Cold‐induced sweetening (CIS) is a serious post‐harvest problem for potato tubers, which need to be stored cold to prevent sprouting and pathogenesis in order to maintain supply throughout the year. During storage at cold temperatures (below 10 °C), many cultivars accumulate free reducing sugars derived from a breakdown of starch to sucrose that is ultimately cleaved by acid invertase to produce glucose and fructose. When affected tubers are processed by frying or roasting, these reducing sugars react with free asparagine by the Maillard reaction, resulting in unacceptably dark‐coloured and bitter‐tasting product and generating the probable carcinogen acrylamide as a by‐product. We have previously identified a vacuolar invertase inhibitor (INH2) whose expression correlates both with low acid invertase activity and with resistance to CIS. Here we show that, during cold storage, overexpression of the INH2 vacuolar invertase inhibitor gene in CIS‐susceptible potato tubers reduced acid invertase activity, the accumulation of reducing sugars and the generation of acrylamide in subsequent fry tests. Conversely, suppression of vacuolar invertase inhibitor expression in a CIS‐resistant line increased susceptibility to CIS. The results show that post‐translational regulation of acid invertase by the vacuolar invertase inhibitor is an important component of resistance to CIS.  相似文献

This work has examined sewage sludge of the following heavy metal concentrations (mg/kg): Cd-3.43; Co-5.25; Cu-131; Fe-51300; Mn-177; Ni-37.5; Pb-104; Zn-3300. Metals speciation by sequential extraction according to Tessier et al. (1979), and Rudd et al. (1988), and a procedure recommended by European Community Bureau of Reference (BCR) (Ure et al., 1993; Quevauiller et al., 1996; Davidson et al., 1999), as well as analysis of chemical forms of metals, have been carried out. It has been found that only Zn concentration is higher than the value permissible for agricultural sewage sludge application (2500 mg/kg). The results obtained by Tessier et al. [Crossref], [Web of Science ®] [Google Scholar], and BCR procedures (Ure et al., 1993; Quevauiller et al., 1996; Davidson et al., 1999) appeared to be consistent. A comparison of the sequential analysis and the analysis of chemical forms of metals indicates that the sum of metal concentrations for the exchangeable, carbonate and bound to Fe/Mn oxyhydroxides forms (found by Tessier et al., 1979, and BCR analyses (Ure et al., 1993; Quevauiller et al., 1996; Davidson, et al., 1999)) corresponds to the sum of sulfate, oxide, metallic and siliceous forms. The concentrations of the forms bound to organic matter or sulfides correspond to the sulfide form while the residue corresponds to the ferrate form. Preparative extraction of metals from the sewage sludge using sodium salt of ethylenediaminetetraacetic acid (EDTA-Na), sodium pyrophosphate (V) and ammonia water has also been investigated. As far as the examined leaching agents are concerned, EDTA-Na appeared to be the best. Single leaching with this agent results in the following metal concentrations remaining in the sludge (mg/kg): Cd-1.1; Co-2.1; Cu-105; Fe-17700; Mn-28.3; Ni-12.8; Pb-44; Zn-1200. They meet the requirements of Polish regulations concerning the use of sewage sludge as a soil fertilizer.  相似文献

Assessment of ecological status in U.K. rivers using diatoms   总被引:6,自引:0,他引:6  
1. The European Union's Water Framework Directive requires all water bodies to achieve ‘good ecological status’ by 2015 and this paper describes a rationale for defining ‘good ecological status’ based on diatoms, a significant component of the biological quality element ‘macrophyte and phytobenthos’. 2. A database of benthic diatom samples collected over the past 20 years was assembled. New sampling, specifically for this project, was undertaken during 2004 to supplement these data. In total 1051 samples were included in the database with matching environmental data. 3. ‘Reference sites’, relatively unimpacted by human activity, were selected from this database by a series of screening steps and these sites were used to develop a site‐specific reference typology. 4. Environmental variables not related to the pressure gradient were used to predict the ‘expected’ Trophic Diatom Index (TDI) values at each site. Site‐specific TDI predictions were used to generate ecological quality ratios (EQRs) ranging from ≥1, where the diatom assemblage showed no impact, to (theoretically) 0, when the diatom assemblage was indicative of major anthropogenic activities. 5. The boundary between ‘high’ and ‘good’ status was defined as the 25th percentile of EQRs of all reference sites. The boundary between ‘good’ and ‘moderate’ status was set at the point at which nutrient‐sensitive and nutrient‐tolerant taxa were present in equal relative abundance. An ecological rationale for this threshold is outlined in the paper.  相似文献

SEM studies of the spathe structures in the two closely related genera Cryptocoryne and Lagenandra show differences between the inner and outer surfaces, as well as in cell structures in the various parts of the spathe. The cell structure reveals patterns mat makes it possible to depict homologous structures of the spathe, even though the spathes of the two genera look different. The basal part of the kettle has a mucilage covering of the cells, interpreted as a hitherto unnoticed food source. The cells of the inner surface of the kettle and tube have downward pointing trichomes. On the second day of flowering these collapse and sink into the cell lumen, which is suggested to create a unique lattice-like structure that enables the insects to climb out of the kettle and tube. The cell structure of the flap shows that it is a prolongation and continuation of the spathe tube margin.  相似文献

Pregnancy loss is a common obstetric problem. Significant causes of miscarriage include genetic and epigenetic disorders of the embryo, immunological and endocrine factors, uterine malformations, improper embryo selection, and lifestyle. Perhaps a hitherto underappreciated cause of miscarriage may be an abnormal microbiota composition of the female reproductive system. Lactobacillus spp. is the most common bacteria within the reproductive tract. However, the protective role of Lactobacilli in the vagina has been well described in the literature, while it is still unknown what function Lactobacilli may have in the uterus. Moreover, new research shows that Lactobacillus spp. can have a role in miscarriage. However, both molecular and immunological mechanisms of host-Lactobacillus spp. interactions are not fully understood. Understanding these relationships will help address the importance and extent of the protective role of Lactobacillus spp. in miscarriage.  相似文献

Kin selection plays an important role in the evolution of social behaviour in terrestrial systems. The extent to which kin selection influences the evolution of social behaviour in marine systems is largely unexplored. Generally, it is considered that kin selection is irrelevant in marine systems, because it is assumed that the dispersing larval phase of marine organisms will break up kin associations. Here, we challenge this assumption and investigate the opportunity for kin selection in a coral reef fish: the humbug damselfish Dascyllus aruanus . This fish lives in groups composed of a large male and a number of smaller females and nonbreeders. We use 10 polymorphic microsatellite loci to assess the relatedness of 265 individuals from 35 groups. The mean coefficient of relatedness among group members is 0.01 ± 0.04, suggesting that individuals are not associated with close relatives. However, the distribution of pairwise relatedness of individuals within groups has an overabundance of positive values, and indicates that there might be 35 pairs of close relatives within groups. Further analyses reveal that close relatives likely are similar in size and small in size, suggesting that they might have recruited together. We conclude that it is possible for kin selection to operate in D. aruanus , but kin recognition will be a prerequisite for such selection. This study reveals that individuals can be associated with close relatives, and there is a hidden potential for kin selection, during certain parts of the life cycle of coral reef fishes.  相似文献

ABSTRACT Mortality due to fisheries interactions has been implicated as a contributor to population decline for several species of sea turtle. The incidental capture of sea turtles in the coastal gillnet fisheries of North Carolina, USA, has received much attention in recent years, and mitigation measures to reduce sea turtle mortality due to gillnet entanglement are a high priority for managers and conservationists. Efforts to evaluate effects of gillnet entanglement on sea turtle populations are complicated by the lack of information on health status of turtles released alive from nets and postrelease mortality. We obtained blood samples from green (Chelonia mydas) and Kemp's ridley (Lepidochelys kempii) sea turtles captured in gillnets for 20–240 minutes to assess the impacts of gillnet entanglement on blood biochemistry and physiological status. We measured concentrations of lactate, corticosterone, ions (Na+, K+, Cl-, P, Ca2+), enzymes (lactate dehydrogenase [LDH], creatine phosphokinase [CPK], aspartate aminotransferase [AST]), protein, and glucose in the blood and also performed physical examinations of turtles to document external indicators of health status (injuries, lethargy, muted reflexes). We evaluated the effects of entanglement time on blood biochemistry and to look for correlations between blood biochemistry and results of the physical examinations. We observed a significant increase in blood lactate, LDH, CPK, phosphorus, and glucose with increased entanglement time. Alterations in blood biochemistry were generally associated with a decline in health status as indicated by results of the physical examination. Although entanglement time plays an important role in determining the health status of sea turtles upon release from a gillnet, our results suggest that factors such as the depth and severity of entanglement may also have an effect on health status of turtles and the probability of postrelease survival. We were unable to set a maximum unattended gillnet soak time to minimize impacts on captured sea turtles, and therefore recommend that fisheries managers continue to enforce the net attendance regulations currently in place in the lower Cape Fear River, North Carolina, during the summer months.  相似文献
