Plants of two genotypes of Lolium perenne L. cv. S23 and a L.perenne ? L. multiflorum Lam. hybrid cv. Augusta were grownin flowing solution culture. N was suppled in one treatmentat 10 mmol m3 NO3 throughout (HN), and in another(LN) the N supply was terminated after 10 d for 11 d. When was re-supplied both LN and HN plants were leftentire or defoliated. The two genotypes showed similar responsesto all treatments. The concentration of N in shoot dry matterdeclined from 4.4% to 2.0% and in the root from 2.8% to 1.0%over the 11 d of N deprivation, with 95% of the initially present being assimilated during this period. LN plantsassimilated 10% more of their total uptake than did HN plants. The in vitro nitrate reductase activity(NRA) was 10- to 50-fold higher in the youngest fully-expandedleaves than in roots and declined in the leaves during N deprivation.Between 26 d after defoliation, there was a large increasein NRA in leaves of HN (but not LN) plants. After defoliationof HN plants, net uptake from 10 mmol m3 declined to negligible levels within 15 h, but in defoliatedLN plants it increased to levels similar to those of entireHN plants (1520 µmol h1 g1 fr. wt.root) within 8 h. When was re-supplied to entire LN plants, uptake of increased to levels similar to those of entire HN plants within 2.3 h, butdid not markedly exceed that of HN plants for at least 10 h.Net uptake of by LN plants during depletion of stirred static nutrient solutions containing 1.0 mol m3 lagged behind that by HN plants by 70100 min, but the maximum unit absorption rate was similar for LNand HN plants (57 µmol h1 g1 fr.wt. root). The nature of the short-term demand for uptake following recovery from the stresses of defoliation andN starvation is discussed. The effects of root temperature and form of N nutrition (NH4or NOJ) on the mineral composition of the plant, the balanceof inorganic cation-anion uptake and on the apparent net effluxof H +/OHions from the roots were studied with 49-d-oldoilseed rape (Brassica napusL. cv. Bien venu) in flowing solutionculture. Plants were pre-treated for 14 d at a root temperatureof 5 °C prior to constant root temperatures of 3, 7, 11or 17°C for 14 d, with a common shoot temperature of 20/15°Cday/night. Nitrogen was supplied as NH+44 or NO3 at 10mmol m3. Values of Q10 (7-17°C) for mean unit absorptionrates of all the major nutrient ions (K+ , Mg++ , NH+4, SO4,H2PO4, NO3), except Ca++, were > 2.0 over the first 5 d oftreatment but thereafter were < 1.5; the apparent effectof temperature on uptake rates diminished with time. Under NH+4nutrition, inorganic cation uptake (Mg+ + + K++Ca+ + +NH+4)exceeded inorganic anion uptake (SO4 4+ H2PO4) over 14d at all temperatures, with the proportion of cation uptakeas NH4 remaining constant (0.67-0-68) irrespective of root temperature.The net efflux of H + from the roots approximately balancedNH+4 uptake (1:1) over 14 d at each temperature and also balancedthe difference between the total uptake of inorganic cationsand inorganic anions. Under NO3 nutrition, the sum of the netefflux of OH and the change in the carboxylate contents of plantsover 14 d approximately balanced the sum of NO3 and SO 4reduced in the plant. The majority of the negative charge associatedwith the reduction of NO3 and SO 4 was apparently effluxedas OH, but this fraction was lower at low root temperatures.The results are discussed in terms of mechanisms that have beenproposed to regulate the internal pH of plants. Abstract Changes in the net uptake rate of K+ and in the average tissue concentration of K+ were measured over 14 d in response to changes in root temperature with oilseed rape (Brassica napus L. cv. Bien venu) and barley (Hordeum vulgare L. cv. Atem). Plants were grown in flowing nutrient solutions containing 2.5 mmol m?3 K+ and were acclimatized over 49 d (rape) or 28 d (barley) to low root temperature (5°C) prior to steady–state treatments at root temperatures between 3 °C and 25 °C, with common air temperature. Uptake of K+ was monitored continuously over 14 d and nitrogen was supplied as NH4++ NO?3 or NH+4 or NO?3. Unit absorption rates of K+ increased with time and with root temperature up to Day 4 or 5 following the change in root temperature. Thereafter they usually approached steady-state, with Q10? 2.0 between 7 °C and 17°C, although rates became similar between 7 °C and 13°C. Uptake of K+ by rape plants was invariably greater under NO?3 nutrition compared with NH+4. The percentage K+ in the plant dry matter increased with temperature from 2% at 3 °C to 4% at 25 °C in rape, but there was less effect of temperature on the average concentrations of K+ in the plant fresh weight or plant water content. Concentrations of K+ in the leaf water fraction of rape plants decreased with increasing root temperature, but in barley they increased with increasing root temperature. Concentrations of K+ in the root water fraction were relatively stable with respect to root temperature. The results are discussed in terms of compensatory changes in K+ uptake following a change in root temperature and the relationships between growth, shoot: root ratio and K+ composition of the plant. 相似文献

Mutation of the nuclear gene sid disables chlorophyll degradation during leaf senescence in the pasture grass Festuca pratensis. This study investigated the effect of the mutation on photosynthesis and on leaf and whole plant growth under a range of nitrogen regimes. When plants were cultivated in a static hydroponic system, the chlorophyll content of fourth leaves of the stay-green mutant Bf993 remained virtually unchanged from full expansion to complete senescence, while tissue of the wild-type (cv. Rossa) became completely yellow. The retention of chlorophyll in Bf993 was not associated with maintenance of photosynthetic activity as shown by rates of light-saturated CO2 fixation and apparent quantum efficiency. Higher levels of total N in senescing leaves of Bf993 than in Rossa indicated reduced nitrogen remobilization in the mutant. When using a range of [NH4NO3], dry matter production and tillering Mere lower for Bf993 at all but the highest [NH4NO3, which was supra-optimal for the wild type. In contrast to the static system, where fluctuations in N supply occurred, growth and [NO3?] uptake were similar in mutant and wild type when [NO3?] was continuously maintained by a flowing solution culture system. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 711 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 725 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 313C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.470.77 at 37 C, 092154at 1117 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 1125 C (095110 mmol N plant1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake.The % N in shoot dry matter declined with decreasing root temperature,from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m3at 3 C to 290 mol m3 at 25 C. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·030.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3] between 10100 mmol m3(RGR=0·15 0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Oilseed rape (Brassica napus L. cv. Bien venu) plants were grownin a system of flowing solution culture and pre-treated at roottemperatures of 3 ?C or 13 ?C for 7 d with or without N suppliedas 10 mmol m3 NH4NO3. Subsequently, N was re-suppliedand root temperatures were reversed for 7 d. Shoot temperatureswere common at 20/15 ?C day/night. Net uptake of , and K+, leaf area, root length, and transpirationwere measured and compared with control plants having root temperaturesof 3 ?C and 13 ?C throughout. Plants that were continuouslysupplied with N and pre-treated at 3 ?C showed a 50% increasein total uptake of , and measured at 13 ?C over 7 d compared with control plants at 3?C,but uptake of was 28% lower and uptake of was 43% higher than that shown by control plants at 13 ?C. Pre-treatment at 3 ?C did not enhance the subsequentuptake of total N or of K+ at 13 ?C relative to the 13 ?C control.Transpiration rates at 3 ?C were on average 70% of those at13 ?C. The concentration of total N in plants was halved after7 d without a supply of N, but total dry matter production wasnot significantly affected. N starvation also increased thetemperature sensitivity of subsequent uptake relative to that of uptake. After N starvation at 13 ?C the uptake of and measured at 13 ?C was 50% higher over 7 d than that measuredunder continuous N supply. In contrast, after N starvation at3 ?C the uptake of at 3 ?C was 70% less, whilst uptake was 50% more than the respective totals absorbed by plants that were continuously supplied withN at 3 ?C. After N starvation the uptake of was generally 4060% of the daily total N uptake, comparedwith 6080% in plants continuously supplied with N. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g1 d1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. Oilseed rape plants (Brassica napus L. cv. Bien venu) were grownin flowing nutrient solution and acclimatized over 49 d to lowroot temperature (5 ?C) prior to treatment at root temperaturesof 3, 7, 11 or 17?C for 14 d, with common air temperature of20/15?C day/night and solution pH 6.0. The effects of root temperatureon growth, uptake and assimilation of N were compared with Nsupplied either as or at 10 mmol m3. Plant fresh weight, dry weight and leafarea increased with root temperature. Q10 values for specificgrowth rates between 7?C and 17?C were 1.10 and 1.33, respectively,under and nutrition. Fresh weight and leaf area were favoured by nutrition, particularly at 11 ?C and 17 ?C. At each temperaturethe net uptake of over 14 d was similar to that of , increasing 4-fold between 3?C and 17 ?C. Unit absorption rates of increased over the first 3 or 4 d, to an extent that increased with roottemperature. Thereafter, near constant values were maintainedat 3, 7 and 11 ?C. Unit absorption rates of at 3 ?C and 7 ?C did not increase over the first 3 or 4 d. Plantsat 17?C responded differently from those at other temperatures,with steep increases in unit absorption rates of and after day 8.
