The CLCA gene family produces both secreted and membrane-associated proteins that modulate ion-channel function, drive mucus production and have a poorly understood pleiotropic effect on airway inflammation. The primary up-regulated human CLCA ortholog in airway inflammation is hCLCA1. Here we show that this protein can activate airway macrophages, inducing them to express cytokines and to undertake a pivotal role in airway inflammation. In a U-937 airway macrophage–monocyte cell line, conditioned media from HEK 293 cells heterologously expressing hCLCA1 (with or without fetal bovine serum) increased the levels of pro-inflammatory cytokines (IL-1β, IL-6, TNF-α and IL-8). This effect was independent of the metalloprotease domain of hCLCA1. Primary porcine alveolar macrophages were similarly activated, demonstrating the effect was not cell line dependent. Similarly, immuno-purified hCLCA1 at physiologically relevant concentration of ~100 pg/mL was able to activate macrophages and induce pro-inflammatory response. This cytokine response increased with higher concentration of immuno-purified hCLCA1. These findings demonstrate the ability of hCLCA1 to function as a signaling molecule and activate macrophages, central regulators of airway inflammation.  相似文献

Galactomannan from Ambiguous Crazyweed (Oxytropis ambigua(Pall.) DC)   总被引:1,自引:0,他引:1  
A galactomannan with a molecular weight of 735 kDa was first isolated and purified from seeds of ambiguous crazyweed Oxytropis ambigua(Pall) DC (family Leguminosae) with a yield of 3.6%. Its aqueous solutions displayed an optical activity ([] D = 73.32°) and high viscosity ([] = 644 ml/g). Chemical analysis and 13C-NMR spectroscopy revealed the presence of D-mannopyranose and D-glucopyranose in the heteropolysaccharide at a molar ratio of 1.39 : 1. The linear backbone of its macromolecule consists of 1,4--D-mannopyranose residues. Single -D-galactose residues substitute 72% of the mannoses to form branches.  相似文献

The effect of sodium salicylate on the population dynamics of the rhizobacterium Pseudomonas aureofaciens BS1393 and its variant bearing the naphthalene biodegradation plasmid pBS216 was studied in the wheat rhizoplane and adjacent soil. Optimum salicylate concentration for the maintenance of the plasmid-bearing strain and for the normal growth of wheat was found to be 250 micrograms/g soil. When the soil was supplemented with salicylate, the population of P. aureofaciens BS1393(pBS216) in the wheat rhizoplane and adjacent soil was, respectively, 4- and 20-fold higher than that of the parent strain lacking the plasmid.  相似文献

High temperature can change the effects of intra- and intercellular regulators and therefore modify the cellular response to hypoxia. We investigated H(2)O(2) production by alveolar macrophages, isolated from adult male rats, which were incubated under conditions of oxygen deficiency and high temperature (experiment in vitro). The incubation of these cells for 2 hours at 10 % or 5 % oxygen led only to slight fluctuations in the H(2)O(2) level, while the rise of temperature from 37 degrees C up to 42 degrees C significantly increased its generation. Level of thiobarbituric acid-reactive substances (TBARS) underwent similar changes. Under these conditions the accumulation of H(2)O(2) was found to be caused mainly by its decreased cleavage rather than its enhanced production. This is indicated by decreased catalase and glutathione peroxidase activity together with a parallel absence of significant changes in superoxide dismutase (SOD) activity. Slight fluctuation of reduced glutathione level and the pronounced increase of glucose-6-phosphate dehydrogenase (G6PD) activity were detected. Strong (5 %) but not moderate (10 %) lack of oxygen led to a sharp increase in formation of cellular nitrite ions by alveolar macrophages. In general, our data showed that high temperature did not lead to any qualitative shifts of defined hypoxia-derived changes in oxidant/antioxidant balance in alveolar macrophages, but promoted sensitivity of cells to oxygen shortage.  相似文献

A series of octa-hexapeptide fragments of HLDF and their conjugates with hemin were obtained by solid phase peptide synthesis. A relationship between the structure and the nuclease activity of the compounds was established. The effect of various factors (medium pH, the presence of metal ions, complexons, reducers, and buffer composition) on DNA destruction with hemin peptides was studied. Preliminary information confirming an oxidative mechanism of this process was obtained. The cleavage of plasmid DNA under the action of hemin peptides was studied by the methods of electron microscopy, gel electrophoresis, and atomic force microscopy.  相似文献

Effect of environmental conditions on formation of Nicotiana tabacum L. megagametophyte was studied. It was established, that unfavorable temperatures can specifically modify the structure of embryo sacs (ES). At low temperature (9/5 °C), ES with a reduced number of cells or with egg-like synergide(s) can be formed; at high variable (40/25 °C) or constant (37 °C) temperatures, ES with excessive numbers of cells or with synergide-like egg cells arise. Total frequencies of the changed ES patterns varied from 8 up to 35 % per plant and depended on the plant genotype and conditions of exposure. The authors studied population composition of 14 strains of EL Tor vibrios. Of 7 strains of Ogava serological type the population composition was homogeneous in 2, and heterogeneous in 5. All the 7 strains of EL Tor vibrios of Inaba serological type had homogeneous population composition. Changes of the existence conditions of EL Tor vibrios of Ogava serological type, containing in their population 5% cells of Hikoshima serological tupe, led to separation of the cells of Inaba serological type; under the same conditions, in El Tor vibrios of Ogava and Inaba serological type with homogeneous population the latter was stable.  相似文献
