Background  

Rupture of the cap of a vulnerable plaque present in a coronary vessel may cause myocardial infarction and death. Cap rupture occurs when the peak cap stress exceeds the cap strength. The mechanical stress within a cap depends on the plaque morphology and the material characteristics of the plaque components. A parametric study was conducted to assess the effect of intima stiffness and plaque morphology on peak cap stress.  相似文献

The ability to maintain human fungiform papillae cells in culture for multiple cell cycles would be of considerable utility for characterizing the molecular, regenerative, and functional properties of these unique sensory cells. Here we describe a method for enzymatically isolating human cells from fungiform papillae obtained by biopsy and maintaining them in culture for more than 7 passages (7 months) without loss of viability and while retaining many of the functional properties of acutely isolated taste cells. Cells in these cultures exhibited increases in intracellular calcium when stimulated with perceptually appropriate concentrations of several taste stimuli, indicating that at least some of the native signaling pathways were present. This system can provide a useful model for molecular studies of the proliferation, differentiation, and physiological function of human fungiform papillae cells.  相似文献

Human cytomegalovirus (HCMV) remains the leading viral cause of birth defects and life-threatening disease in transplant recipients. All approved antiviral drugs target the viral DNA polymerase and are associated with severe toxicity issues and the emergence of drug resistance. Attempts to discover improved anti-HCMV drugs led to the identification of the small-molecular-weight compound AIC246 (Letermovir). AIC246 exhibits outstanding anti-HCMV activity in vitro and in vivo and currently is undergoing a clinical phase IIb trial. The initial mode-of-action studies suggested that the drug acts late in the HCMV replication cycle via a mechanism distinct from that of polymerase inhibitors. Here, we extend our mode-of-action analyses and report that AIC246 blocks viral replication without inhibiting the synthesis of progeny HCMV DNA or viral proteins. The genotyping of mutant viruses that escaped AIC246 inhibition uncovered distinct point mutations in the UL56 subunit of the viral terminase complex. Marker transfer analyses confirmed that these mutations were sufficient to mediate AIC246 resistance. The mapping of drug resistance to open reading frame UL56 suggests that viral DNA processing and/or packaging is targeted by AIC246. In line with this, we demonstrate that AIC246 affects the formation of proper unit-length genomes from viral DNA concatemers and interferes with virion maturation. However, since AIC246-resistant viruses do not exhibit cross-resistance to previously published terminase inhibitors, our data suggest that AIC246 interferes with HCMV DNA cleavage/packaging via a molecular mechanism that is distinct from that of other compound classes known to target the viral terminase.  相似文献

Squids of the family Onychoteuthidae are ecologically important in pelagic food webs and have been reported from every ocean except the Arctic. Although they are abundant and caught frequently as bycatch in fisheries, the biogeography of many species remains poorly understood. Species identification within the Atlantic Ocean is usually restricted to two species: Onychoteuthis banksii and Onykia carriboea. Here, we report the occurrence of four species of the family Onychoteuthidae (Onychoteuthis cf. banksii, Onykia carriboea, Walvisteuthis jeremiahi, and Onychoteuthis sp. AL 2) from the Sargasso Sea in the western Atlantic, identified using DNA barcoding (cytochrome c oxidase subunit I) and morphology. Our results have doubled the known onychoteuthid biodiversity in the Sargasso Sea, which has implications for the ecology of this oceanic region.  相似文献

We conducted a year-round mesozooplankton study in the Arctic Kongsfjord from August 1998 until July 1999 to investigate seasonal abundance and vertical as well as stage distributions of the prevalent taxa. It is the first investigation in Kongsfjord that covers the Arctic winter season and provides reasonable estimates also of small-sized copepod species. Abundant smaller copepods comprised Oithona similis, Pseudocalanus minutus, Microcalanus spp., Triconia borealis and Acartia longiremis. Among the larger copepods, Calanus finmarchicus, C. glacialis, C. hyperboreus and Metridia longa dominated. The thecosome pteropod Limacina helicina was also an important component. Abundance maxima occurred in November (988,669 ind. m?2) with one to two orders of magnitude higher numbers as compared to all other months (39,832–200,067 ind. m?2). The summers of 1998 and 1999 were characterized by intrusions of Atlantic water, but the community was not entirely dominated by advected boreal species. During winter, the majority of the mesozooplankton occurred below 100 m. Advection is the most likely reason for the accumulation of zooplankton at depth in winter, but local production may also contribute to high overwintering numbers. Much lower abundances of most species in spring suggest high winter mortality and emphasize the importance of sufficient reproductive success during the previous summer to ensure enough winter survivors as seed stock for the coming reproductive season. This study was conducted prior to the recent warming trend in the Arctic. Therefore, it provides valuable baseline data and allows comparing present and future states of the zooplankton community in Kongsfjord.  相似文献

It is generally accepted that inositol-1,4,5-trisphosphate (InsP3) plays a role in olfactory transduction. However, the precise mode of action of InsP3 remains controversial. We have characterized the conductances activated by the addition of 10 microM InsP3 to excised patches of soma plasma membrane from rat olfactory neurons. InsP3 induced current fluctuations in 25 of 121 inside-out patches. These conductances could be classified into two groups according to the polarity of the current at a holding potential of +40 to +60 mV (with Ringer's in the pipette and pseudointracellular solution in the bath). Conductances mediating outward currents could be further divided into large- (64 +/- 4 pS, n = 4) and small- (16 +/- 1.7 pS, n = 11) conductance channels. Both small- and large-conductance channels were nonspecific cation channels. The large-conductance channel displayed bursting behavior at +40 mV, with flickering increasing at negative holding potentials to the point where single-channel currents were no longer discernible. The small-conductance channel did not display flickering behavior. The conductance mediating inward currents at +40 to +60 mV reversed at +73 +/- 4 mV (n = 4). The current traces displayed considerable fluctuations, and single-channel currents could not be discerned. The current fluctuations returned to baseline after removal of InsP3. The power density spectrum for the excess noise generated by InsP3 followed a 1/f dependence consistent with conductance fluctuations in the channel mediating this current, although other mechanisms are not excluded. These experiments demonstrate the presence of plasma membrane InsP3-gated channels of different ionic specificity in olfactory receptor cells.  相似文献

The Brazilian Atlantic Forest biome is considered a hotspot of biodiversity. It is estimated that today the remaining primary vegetation covers only 7.5% of its original area. Bromeliad species are important components of this biome. Some of these species are endemic, like the highly endangered Vriesea reitzii. Tissue culture techniques have been often employed for the mass propagation and conservation of threatened bromeliad species. In the present work we describe a procedure for the micropropagation and in vitro conservation of V. reitzii. Seedling explants were cultured on MS and LPm liquid media supplemented with BA, NAA and GA3. The induction and multiplication of shoots were observed in the MS medium supplemented with NAA (2 μM) and BA (4 μM). The best conditions for maintenance and conservation of shoots were half-strength or MS medium. Shoot elongation was observed in MS medium supplemented with GA3 (10 μM). MS medium supplemented with NAA (1 μM) and BA (2 μM) enabled an efficient proliferation system. The acclimatization of shoots longer than 2 cm resulted in 100% survival rate.  相似文献

The synthesis of bile acid oligodeoxynucleotide conjugates via the 3-OH group of the bile acids is described. When used in vivo in rats, covalent conjugation of an oligodeoxynucleotide via a linker to cholic acid resulted in an increased biliary excretion of bile acid-oligodeoxynucleotide conjugates compared to unconjugated oligodeoxynucleotides.  相似文献
