Background aims

Multipotent mesenchymal stromal cell (MSC)-based medicines are extensively investigated for use in regenerative medicine and immunotherapy applications. The International Society for Cell and Gene Therapy (ISCT) proposed a panel of cell surface molecules for MSC identification that includes human leukocyte antigen (HLA)-DR as a negative marker. However, its expression is largely unpredictable despite production under tightly controlled conditions and compliance with current Good Manufacturing Practices. Herein, we report the frequency of HLA-DR expression in 81 batches of clinical grade bone marrow (BM)-derived MSCs and investigated its impact on cell attributes and culture environment.

Methods

The levels of 15 cytokines (interleukin [IL]-1β, IL-4, IL-6, IL-10, IL-17A, IL-17F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon-γ, soluble CD40 ligand and tumor necrosis factor-α) were determined in sera supplements and supernatants of BM-MSC cultures. Identity, multipotentiality and immunopotency assays were performed on high (>20% of cells) and low (≤20% of cells) HLA-DR+ cultures.

Results

A correlation was found between HLA-DR expression and levels of IL-17F and IL-33. Expression of HLA-DR did neither affect MSC identity, in vitro tri-lineage differentiation potential (into osteogenic, chondrogenic and adipogenic lineages), nor their ability to inhibit the proliferation of stimulated lymphocytes.

Discussion

Out of 81 batches of BM-MSCs for autologous use analyzed, only three batches would have passed the ISCT criteria (<2%), whereas 60.5% of batches were compliant with low HLA-DR values (≤20%). Although a cause–effect relationship cannot be drawn, we have provided a better understanding of signaling events and cellular responses in expansion culture conditions relating with HLA-DR expression.  相似文献

We describe the development of 13 variable microsatellites developed to investigate population structure and dispersal in the sea urchin Centrostephanus rodgersii. This species is the dominant grazing herbivore in southeast Australian coastal waters and has the ability to modify benthic community structure. The microsatellites we identified showed a range of allele numbers (4–21) and expected heterozygosity (0.32–0.91) in two sampled populations. Contrary to previous findings in free‐spawning marine invertebrates, genotype proportions in neither population deviated significantly from Hardy–Weinberg expectations.  相似文献

Aeschynomene sabulicola L.P. Queiroz & D. Cardoso is proposed as a new species of the section Ochopodium. This new species seems to be related to the series Viscidulae, as it presents herbaceous, slender branches, densely covered by viscid glandular hairs. However, it possesses fruits much larger than expected for this series. Aeschynomene sabulicola can be diagnosed by the erect habit, reaching c. 3.5 m high, inflorescences divaricate and horizontal with remote flowers, and fruits with only one subreniform article, this measuring 13–14 × 7–8 mm. 1. A 7‐year study was conducted in three hydrologically distinct sections within the highly regulated, lowland Campaspe River to investigate the influence of hydrology on temporal and spatial patterns in fish composition, abundance and recruitment. One section had 6 months, one section 2 months and one section no months of increased flow due to storage releases. The fish fauna of the less regulated, nearby Broken River served as a reference to which that of the Campaspe River was compared for the last 3 years of the study to allow insight into the relative effects of hydrology, barriers to movement and other environmental characteristics. The study included one high‐flow year, a moderate‐flow year and five low‐flow years. 2. A total of 16 fish species – 10 native and six alien – were caught in the Campaspe River, although of the native species, only three are considered to have self‐sustaining populations. The remaining species are either itinerants or a result of stocking. Alien species comprised approximately 64% of the total biomass of all fish caught. 3. Overall composition of the fish fauna did not differ significantly by year, but did by section of river. Species richness and the abundance of most of the dominant species also differed significantly by river section, but there was little inter‐annual variation in the abundance of any species, except for European perch and for common carp; the latter showing an increase in abundance following a high‐flow event during the spring of 2000 as a result of recruitment. 4. Overall faunal composition was not influenced by hydrology. However, multiple regression indicated that species richness, abundance of the dominant species and abundance of young‐of‐year (YOY) of golden perch, European perch and common carp all were influenced significantly by hydrological variables. The nature of the relationships was dependent on river section and hydrological season (‘winter’ or ‘spring/summer’). Of note was the result that the total abundance of fish and that of YOY common carp were significantly positively related to the number of spells above the threshold for movement upstream through the lower two weirs in the Campaspe River. Only one significant relationship between hydrological and fish‐related variables was found for the upper river section, whereas seven and five were found for the lower and middle sections respectively. 5. Comparisons with fish collected in the Broken River over 3 years suggest that the fauna of the Broken River is in a more natural state than that of the Campaspe River. Since the two rivers do not differ substantially in water quality, and since both contain significant weirs, which act as barriers to movement of fish, flow regulation is most likely to be the major reason for the poor state of the fauna in the Campaspe River.  相似文献

1. Longitudinal gradients in the epilimnetic waters of stratified reservoirs provide a useful database to study changing environmental conditions. The spatial distribution, assemblage structure and specific adaptations of phytoplankton assemblages can be analysed along these gradients over short time scales. 2. Four reservoirs with a similar typology, located along an altitudinal gradient in the same eco‐region, were sampled along their longitudinal axes. In total, 19 sampling stations provided a trophic spectrum, ranging from oligo‐mesotrophy to hypertrophy, which was quantified by calculating the trophic state index of each sampling station in the four reservoirs. 3. Several patterns in phytoplankton assemblage structure were detected. Total chlorophyll‐a (Chl‐a), biovolume, abundance and the relative biomass contribution of the main algal groups (chlorophytes, cyanobacteria, cryptophytes and diatoms) were highly correlated with their location along the trophic gradient. 4. We also adopted the functional classification of Reynolds et al. (2002) : this effectively summarized differences among phytoplankton assemblages under varying resource‐limiting combinations, especially nutrients and underwater light climate. 5. In terms of relationships with the trophic gradient, diatoms and cyanobacteria exhibited significant opposing trends in both their relative chlorophyll contribution to total Chl‐a and biovolume. Chlorophytes were more abundant at an intermediate position along the trophic spectrum. 6. The identified patterns are consistent with models of self‐organization of phytoplankton assemblages. In particular, light availability was a strong determinant of size and shape diversity, especially in hypertrophic conditions, where ‘R‐strategist’, needle shaped species, dominated the system. In contrast, under decreased availability of nutrients and higher light extinction coefficients (Kd), the system was co‐dominated by C‐ and S‐strategist species, having shapes with a higher surface/volume ratio.  相似文献

The relationship between amino acid and sugar export to thephloem was studied in young wheat plants (Triticum aestivumL. ‘Pro-INTA, Isla Verde’) using the EDTA-phloemcollection technique. Plants grown with a 16 h photoperiod showeda rapid decrease in the concentration of sugars and amino acidsin the phloem exudate from the beginning of the dark period.When plants grown with a 16 h photoperiod were kept in the darkfor longer than 8 h the free amino acid content in leaves andexudate (on a dry weight basis) increased continually throughoutthe 72 h of darkness. During the first 24 h of darkness thesugars in the phloem exudate decreased to 30% of the initialvalue, and returned to the control level when plants were returnedto light. When plants grown under low light intensity for 10d were transferred to high light intensity, they showed an increasein leaf sugar content (dry weight basis) after 3 d but therewere no differences in leaf free amino acid content (dry weightbasis) compared to low-light plants. The sugar concentrationin the phloem exudate was increased by higher light intensities,but there was no difference in the amino acid concentrationof the phloem exudate, and thus the amino acid:sugar ratio inthe phloem decreased in the high-light plants. The present resultssuggest that amino acids can be exported to the phloem independentlyof the export of sugars.
