Non‐local population stocking can have adverse genetic effects on wild populations through loss of genetic diversity and introgressive hybridization. The grey partridge (Perdix perdix) has been an important European game species for centuries, widely subject to translocation and stocking. After c. 80 years of releasing reared grey partridges in Denmark, this study investigated whether an indigenous Danish grey partridge still existed. If so, they would (1) belong to the western European clade (W1) and (2) be more closely related to the historical, indigenous grey partridges than to farm‐bred partridges. These predictions were tested by analysing the variation in both the mitochondrial control region (CR1) and microsatellite markers in museum samples representing the ancestral indigenous Danish grey partridge, contemporary wild grey partridges and farmed grey partridges from the five largest farms in Denmark. Phylogeography and population structure analyses showed traces of the indigenous Danish grey partridges amongst recent wild partridges in certain areas and significant genetic differences between farmed partridges and historical and recent partridges. The results also showed that the indigenous Danish grey partridges belonged to the western European clade (W1 haplotype). A foreign stocking effect was detected on the remote island of Bornholm, where the current population originated from introduced Danish and Bohemian grey partridges. The loss of haplotype diversity over time in certain geographical areas probably results from serious declines in wild Danish grey partridge numbers in recent decades. With the advent of sophisticated genetic markers, studies on mating systems and reproductive apportionment have become increasingly feasible. In particular, paternity analyses in haplodiploid species have gained in power as hemizygous paternal genotypes allow maternal and paternal genotypes to be directly inferred from offspring genotypes. The computer program matesoft offers both newly developed algorithms for inferring maternal and paternal genotypes, and integrated estimation and correction procedures for calculating mating frequency statistics. It is crucial to examine the genetic diversity and structure of small, isolated populations, especially those at the edge of their distribution range, because they are vulnerable to stochastic processes if genetic diversity is low and the isolation level is high, and because such populations provide insight into the consequences of population declines in a broader conservation context. The harbour seal (Phoca vitulina) population in Svalbard is the world's northernmost P. vitulina population. Nothing is known about the genetic diversity, distinctiveness, or origin of this small, marginalized mammalian population. Thus, the present study investigated its genetic status in the context of nearby P. vitulina in Iceland, south‐east Greenland, and northern Norway: this species is depleted/threatened in all of these regions. A total of 174 samples distributed between the four locations were analysed using 15 polymorphic microsatellites and variation in the displacement loop region (D‐loop). Each of the four locations was a genetically distinct population. The Svalbard population was highly genetically distinct, had reduced genetic diversity, received limited gene flow, had a rather low effective population size, and showed an indication of having experienced a bottleneck resulting from a recent population decline. The significant heterozygote excess observed in the Svalbard sample might be attributed to the low effective population size, which could initiate future population inbreeding effects. This phenomenon has not been reported earlier from other P. vitulina populations, but if the Svalbard population is experiencing inbreeding, this could reduce its resilience to climate change, disease outbreaks, or other perturbations. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 420–439.  相似文献

Abstract: We evaluated the ability of a set of published trans-species molecular sexing primers and a set of walrus-specific primers, which we developed, to accurately identify sex of 235 Pacific walruses (Odobenus rosmarus divergens). The trans-species primers were developed for mammals and targeted the X- and Y-gametologs of the zinc finger protein genes (ZFX, ZFY). We extended this method by using these primers to obtain sequence from Pacific and Atlantic walrus (O. r. rosmarus) ZFX and ZFY genes to develop new walrus-specific primers, which yield polymerase chain reaction products of distinct lengths (327 and 288 base pairs from the X- and Y-chromosome, respectively), allowing them to be used for sex determination. Both methods yielded a determination of sex in all but 1–2% of samples with an accuracy of 99.6–100%. Our walrus-specific primers offer the advantage of small fragment size and facile application to automated electrophoresis and visualization.  相似文献

THE frog embryo cell line ICR 2A is the first established haploid vertebrate cell line1. In haploid cells recessive mutations should be detectable at a frequency 106 to 109 times greater than expected in diploid cells; mutagen treatment should increase the yield further. These predictions are useful to test whether variants arising in culture are the result of gene mutation. To apply this test to frog cells, mutations for thymidine kinase were sought. Such mutants were first obtained by exposing mouse L cells to the thymidine analogue 5-bromodeoxyuridine (BUdR); a loss of thymidine kinase activity prevented the lethal incorporation of BUdR into DNA2. The new phenotype was considered to be the result of gene mutation because of its heritability and eventually because of data from Luria-Delbrück fluctuation analyses3 (a test of the spontaneity or non-inducibility of a process, not its cause). The question of origin was further complicated by a number of factors: (1) the necessity of a long, repeated, exposure to BUdR2; (2) the high mutation rate (up to 10?3) compared with bacterial mutants (10?910?6)4,5; and (3) the presence of resistant clones with intermediate enzyme levels4,5.  相似文献

1. Fish community structure and habitat distribution of the abundant species roach, perch and ruffe were studied in Lake Nordborg (Denmark) before (August 2006) and after (August 2007) aluminium treatment to reduce internal phosphorus loading. 2. Rapid changes in fish community structure, abundance and habitat distribution occurred following a decline in in‐lake phosphorus concentrations from 280 to 37 μg P L?1 and an increase in Secchi depth transparency from 1.1 to 1.9 m (August). The proportion of perch in overnight gill net catches increased, whilst roach decreased, and the average weight of all key species increased. 3. The habitat distribution of perch and roach changed from a high proportion in the upper pelagic and littoral zones in 2006, towards enhanced proportions in the deeper pelagic and profundal zone in 2007. The abundance of large‐bodied zooplankton increased and the abundance of benthic invertebrates decreased in the same period, suggesting that the habitat shift was not induced by food limitation. 4. Ruffe shifted from the littoral and upper profundal zones towards the deep profundal zone, likely reflecting an increased predation risk in the littoral zone and better oxygen conditions in the deep profundal. 5. Our results indicate that enhanced risk of predation in the upper pelagic and the littoral zones and perhaps improved oxygen concentrations in the deeper profundal zone at decreasing turbidity are responsible for the observed habitat shift. The results indicate that fish respond rapidly to changes in nutrient state, both in terms of community structure and habitat use.  相似文献

Abstract. 1. We studied how colonies of the red wood ant, Formica polyctena , adjust the numbers of foragers allocated to different foraging trails. In a series of field experiments, foragers were marked and transferred from one nest to another, related nest, where they joined the foraging force. Transferred workers acted as a reserve of uncommitted, available foragers.
2. Previous work shows that each individual forager habitually uses one trail. We found that for an uncommitted forager, the influence of recruitment initially is stronger than that of directional fidelity. Transferred workers were likely to use trails leading to new food sources. When transferred to a new nest, foragers were not likely to use a trail in the same direction as their original trail in the donor nest.
3. After a week, transferred foragers tended to develop route fidelity. Even after bait was no longer present, they continued to use the trail that had formerly led to a bait source.
4. We examined how colonies adjust numbers on a trail by experimentally depleting some trails. Colonies usually did not compensate for depletion: foragers were not recruited to depleted trails.
5. In general, the dynamics of foraging in this species facilitate a consistent foraging effort rather than rapid adjustments of forager allocation.  相似文献

Submerged macrophytes as indicators of the ecological quality of lakes   总被引:1,自引:0,他引:1  
1. We analysed submerged macrophyte communities from 300 Danish lakes to determine the efficacy of different species, maximum colonisation depth (Cmax) of plants as well as coverage and plant volume inhabited (PVI) as indicators of eutrophication. 2. Most species occurred at a wide range of phosphorus and chlorophyll a (Chla) concentrations, but some species of isoetids (Lobelia, Isoëtes) and Potamogeton (Potamogeton gramineus, Potamogeton alpinus and Potamogeton filiformis) were mainly found at low nutrient concentrations and hence may be considered as indicators of nutrient poor conditions. However, species typically found in nutrient‐rich conditions, such as Elodea canadensis and Potamogeton pectinatus, were also found at total phosphorus (TP) <0.02 mg P L?1 and Chla <5 μg L?1 and therefore cannot be considered as reliable indicators of eutrophic conditions. 3. Submerged macrophyte coverage, PVI and the Cmax were negatively correlated with TP and Chla. However, variability among lakes was high and no clear thresholds were observed. At TP between 0.03 and 0.07 mg P L?1 plant coverage in shallow lakes ranged from nearly 0 to 100%, whilst at concentrations between 0.10 and 0.20 mg P L?1 only 29% of the lakes had coverage >10%. Cmax was found to be a useful indicator only in deep lakes with unvegetated areas in the deeper part, whereas the use of coverage was restricted to shallow lakes or shallow areas of deep lakes. 4. Overall, submerged macrophytes responded clearly to eutrophication, but the metrics investigated here showed no well‐defined thresholds. We developed a simple index based on species richness, presence of indicator species, coverage and Cmax, which might be used to track major changes in macrophyte communities and for lake classification.  相似文献

Colony kin structure and spatial population structure were studied in multiple populations of the ant Formica lemani , using allozymes and DNA microsatellites. Average genetic relatedness between nestmate workers varied little between populations ( r  = 0.51–0.76), indicating that the average colony kin structure was relatively simple. Worker genotypes could not be explained with a single breeding pair in all nests, however, and the distribution of relatedness estimates across nests was bimodal, suggesting that single- and multi-queen colonies co-occur. We studied spatial population structure in a successional boreal forest system, which is a mixture of different aged habitats. Newly clear-cut open habitat patches are quickly colonized by F. lemani , where it is able to persist for a limited number of generations. Newly-founded populations showed signs of a founder effect and spatial substructuring, whereas older populations were more homogenous. This suggests that new populations are founded by a limited number of colonizers arriving from more than one source. Genetic differentiation among local populations was minor, indicating strong migration between them. Facultative polygyny and habitat succession in boreal ants   总被引:2,自引:0,他引:2  
High dispersal risks of ant queens make staying in the natal patch more attractive than long range dispersal. These alternative strategies and the mode of colony founding determine the average number of queens in the population. Increasing competition and queen predation make independent colony founding increasingly difficult and the only option for new queens to reproduce in the habitat patch may be to enter an existing colony. The effect of nest-site availability to the number of queens was studied in successional spruce-dominated taiga forests in facultatively polygynous ants Myrmica ruginodis, M. sulcinodis, Leptothorax acervorum, Formica sanguinea and F. truncorum. Decreasing relatedness among worker nestmates supports an association between increasing habitat age and polygyny to some extent. M. sulcinodis and L. acervorum persist in this type of taiga only for a relatively short period. Relatedness varied only slightly among populations, but lower relatedness estimated in other studies suggested higher levels of polygyny in older populations. In M. ruginodis there was more variation in relatedness and it was possibly connected to the relative proportions of the two social forms of the species. In F. sanguinea and F. truncorum the decrease in relatedness with increasing age of the habitat was clearest. Other factors favouring limited dispersal and acceptance of new queens in the colonies are, however, hard to separate.  相似文献
