The effect of ten amino acids as the sole nitrogen source for the growth of Cladosporium herbarum (Link.) Fr. and Trichothecium roseum (Bull.) Link. was studied in order to clarify the fungus-host plant relationship. Special attention was paid to some rare non-protein amino acids of legumes. The best nitrogen sources for both fungi were γ-aminobutyric acid, arginine, serine and proline. Cladosporium could use homoarginine and canavanine, but these two amino acids were not used by Trichothecium when each was given as the only nitrogen source. Both fungi utilized ornithine, homoserine and a,γ-diaminobutyric acid to a limited extent. Pipecolic acid was not growth promoting. The growth-retarding effects of rare non-protein amino acids (homoarginine, canavanine, a,γ-diaminobutyric acid and pipecolic acid) were usually reversed by higher concentrations of their normal analogues. It is possible that rare non-protein amino acids may slightly protect the host plant against fungal infections, but there are clear differences between fungi in their reaction to non-protein amino acids.  相似文献

The growth of Drosera rotundifolia was studied in aseptic cultures with 17 dipeptides as the only nitrogen source. About half of the dipeptides were well or partly utilized. Compounds containing glycine, alanine, glutamic or aspartic acid are clearly more favourable than dipeptides containing proline. Arginyl-aspartic acid (1.25 mM) promoted growth more than inorganic nitrogen (1.25 mM of NH4NO3). Glycyl-alanine gave about the same growth response as NH4NO3. The inocules died rapidly in medium containing leucyl-tyrosine and dipeptides containing methionine and valine were also toxic. There was usually a clear correlation between the growth-retarding or growth-stimulating effect of the dipeptides and the effects of their amino acid components.  相似文献

The nitrogen metabolism of a bog moss, Sphagnum nemoreum Scop., has been studied in aseptic cultures. The effect of several protein amino acids, especially those found in peat, has been investigated. NH4NO3 (1.25 mM) was the best nitrogen source but NH4+ ions were more effectively utilized than NO3 ions when given as the only nitrogen source. Some of the amino acids (2.5 mM) allowed fairly satisfactory growth (arginine and alanine) when given as the only nitrogen source, but some of them were not utilized at all (leucine, lysine, isoleucine and methionine). Given at low concentrations (0.001 and 0.25 mM) together with NH4NO3 (2.5 mM), most of the protein amino acids failed to reveal any growth-promoting or -inhibiting effect. Only lysine (0.25 mM) clearly inhibited growth under these conditions. The nitrogen metabolism of Sphagnum nemoreum seems to be rather flexible and this species is more tolerant of organic nitrogen, especially hydroxyproline, than the higher plants.  相似文献

Morphactin ester applied as a bark band to stems of Monterey pine seedlings (Pinus radiata D. Don) at first stimulated and then inhibited the rate of stem elongation over a 20-day period. The inhibition was caused by a direct action of morphactin on the shoot rather than by indirect action on the root system. 14C labelled morphactin applied to the bark accumulated in the shoot apices and the amount accumulated could be related to the inhibition of growth. More than 70% of labelled material isolated from roots and shoot apices was in the form of a metabolite similar to the carboxylic acid derivative of the morphactin ester. A preparation of 14C labelled acid derivative applied to the bark was translocated to the shoot apex and to the roots and inhibited stem elongation.  相似文献

The influence of various evolutionary factors on the populations of the colour polymorphic meadow spittlebug, Philaenus spumarius (Homoptera), was investigated on islands in the Gulf of Finland. Long-term (30-year) investigations showed that the populations can survive through very narrow bottlenecks, but after strong reduction in the size of a population, random genetic drift may remove alleles or change allele frequencies. One of the factors sustaining the presence of almost all the colour polymorphism alleles in almost all the populations is gene flow. Migration seems to be largely by air and by the sea surface (anemohydrochoric). On one rocky island, gene flow was able to carry the top dominant allele, pT, to a number of scattered plant stands. However, it was not powerful enough to alter the differences in allele frequencies between populations living on the same island in meadows of different types 3 m or 12 m from each other. It is highly probable that the factor operating is meadow-specific selection. Its action appears to be based on the ecological character of the meadow. In the different meadows, the coverages of the food plants of the spittlebug are dissimilar. Differences in plant assemblages are possibly of importance in maintaining the meadow-specific allele frequencies in the spittlebug populations.  相似文献

Screening of 55 different cyanobacterial strains revealed that an extract from Nostoc XPORK14A drastically modifies the amplitude and kinetics of chlorophyll a fluorescence induction of Synechocystis PCC 6803 cells. After 2 d exposure to the Nostoc XPORK14A extract, Synechocystis PCC 6803 cells displayed reduced net photosynthetic activity and significantly modified electron transport properties of photosystem II under both light and dark conditions. However, the maximum oxidizable amount of P700 was not strongly affected. The extract also induced strong oxidative stress in Synechocystis PCC 6803 cells in both light and darkness. We identified the secondary metabolite of Nostoc XPORK14A causing these pronounced effects on Synechocystis cells. Mass spectrometry and nuclear magnetic resonance analyses revealed that this compound, designated as M22, has a non‐peptide structure. We propose that M22 possesses a dual‐action mechanism: firstly, by photogeneration of reactive oxygen species in the presence of light, which in turn affects the photosynthetic machinery of Synechocystis PCC 6803; and secondly, by altering the in vivo redox status of cells, possibly through inhibition of protein kinases.  相似文献

Suspension-cultured cells and aseptically cultured roots ofintact plants of Atropa belladonna L. removed tropane alkaloidprecursors arginine (Arg) and ornithine (Orn) at nearly an equalrate from the feeding medium. A great part of Arg- and Orn-derived14C-label was found in ethanol-insoluble compounds, mostly inproteins already after 2 h feeding. Ethanol-soluble label inthe roots was found mainly in amino acids (e.g. glutamine, Gln)after 2 h feeding, and after 20 h also in some intermediatesof the urea cycle (e.g. argininosuccinate). In suspension cultures, subculturing of the initiation callusdecreased both the uptake of the basic amino acids tested andtheir binding on to the apoplastic space. After 20 h feedingwith Arg more label was found in organic acids in stationaryphase suspension cultures with repressed alkaloid synthesisthan in roots producing alkaloids. The growth phase and passagenumber also affected into which amino acids the label was incorporated.When the initiation callus was young (the 3rd passage), theintermediates of the urea cycle were actively labelled, butwhen the initiation callus was older (the 8th passage) and thesuspension formed roots, especially Gln was labelled. Only tracesof -N-methylornithine were detected in feeding experiments withOrn and Arg. The aim of the study was to determine how those texture attributes that elderly people find difficult to eat are related to their preferences for different modifications of carrot textures. This study was conducted with same methods in Finland and in the United Kingdom (UK). Trained sensory panels in both countries described sensory profiles of a range of carrot samples with same glossary of texture terms. The texture of carrot samples was manipulated using different preparation and cooking methods. Based on the Principal Component Analysis (PCA), boiled and puréed samples were perceived as soft, wet, pulpy and smooth, whereas raw samples were dry, brittle, rough, crispy and crunchy. Two age groups were used for the consumer tests: a young adult group aged 23 to 40 years, mean 32 (Finland) and 33 (UK) and an elderly group aged over 60 years, mean 75 (Finland) and 76 (UK). Both the elderly and young adult respondents considered samples as difficult to eat if they needed a long chewing time, were crunchy, hard, brittle, dry, rough or sharp, such as raw slices and coarsely grated samples. On the other hand, slimy, wet, smooth, soft and pulpy samples, such as boiled carrots and purée, were considered as easy-to-eat. The young adults liked more difficult textures such as rough, crispy, crunchy and hard than did the elderly respondents, but the easiest textures were not liked by either age group. 1. Extracellular enzyme activities were examined in freshwater basins representing a transition from hypertrophy to mesotrophy and in axenic cyanobacterial cultures to evaluate the ecological role of extracellular enzyme activities of cyanobacteria.
2. Aminopeptidase activity was related to the trophic status of the lake basins. The activity was highest in the most eutrophic basin and decreased in the less nutrient-rich basins. Cyanobacteria were the most important autotrophic organisms and aminopeptidase activity was positively associated with cyanobacterial biomass.
3. In an axenic Anabaena batch culture, nitrogenase activity was several orders of magnitude higher than leucine aminopeptidase activity. Nitrate did not have an effect on aminopeptidase activity or growth, but significantly reduced the rate of nitrogen fixation. A high phosphorus concentration at the beginning of the Anabaena batch-culture experiment resulted in reduced phosphatase activity.
4. In Lake Hiidenvesi, aminopeptidase activity probably originated mostly from attached bacteria and less so from cyanobacteria.  相似文献

Anatomical and submicroscopical changes in the cotyledons and radicles of Bidens cernua L. have been studied at five developmental stages. In the subcellular structure, these two plant organs are relatively similar but each developmental stage is characterized by a distinct fine structure. Protein bodies, which occupy the bulk of the cell in dormant embryos, develop as filling products of vacuoles. Ribosomes are seen abundantly at this stage, both in the nucleus and the plasma strands. Small vesicles which are the initials of globoids can be detected in the vacuoles even of rather young cells. They later associate at the periphery of protein masses secreted in the vacuoles. Many light globoids are seen in the protein bodies of mature cells. Some amyloplasts are present in the early developmental stages but not in the dormant cells. The endoplasmic reticulum becomes filled with osmiophilic storage fat, and later many spherosomes are seen between the protein bodies. Some osmiophilic material is also found in the intercellular spaces.  相似文献
