Abstract Microorganisms frequently contaminate jet fuel and cause corrosion of fuel tank metals. In the past, jet fuel contaminants included a diverse group of bacteria and fungi. The most common contaminant was the fungus Hormoconis resinae. However, the jet fuel community has been altered by changes in the composition of the fuel and is now dominated by bacterial contaminants. The purpose of this research was to determine the composition of the microbial community found in fuel tanks containing jet propellant-8 (JP-8) and to determine the potential of this community to cause corrosion of aluminum alloy 2024 (AA2024). Isolates cultured from fuel tanks containing JP-8 were closely related to the genus Bacillus and the fungi Aureobasidium and Penicillium. Biocidal activity of the fuel system icing inhibitor diethylene glycol monomethyl ether is the most likely cause of the prevalence of endospore forming bacteria. Electrochemical impedance spectroscopy and metallographic analysis of AA2024 exposed to the fuel tank environment indicated that the isolates caused corrosion of AA2024. Despite the limited taxonomic diversity of microorganisms recovered from jet fuel, the community has the potential to corrode fuel tanks. 相似文献

Biodeterioration of archaeological sites and historic buildings is a major concern for conservators, archaeologists, and scientists
involved in preservation of the world's cultural heritage. The Maya archaeological sites in southern Mexico, some of the most
important cultural artifacts in the Western Hemisphere, are constructed of limestone. High temperature and humidity have resulted
in substantial microbial growth on stone surfaces at many of the sites. Despite the porous natureof limestone and the common
occurrence of endolithic microorganisms in many habitats, little is known about the microbial flora living inside the stone.
We found a large endolithic bacterial community in limestone from the interior of the Maya archaeological site Ek' Balam.
Analysis of 16S rDNA clones demonstrated disparate communities (endolithic: >80% Actinobacteria, Acidobacteria, and Low GC
Firmicutes; epilithic: >50% Proteobacteria). The presence of differing epilithic and endolithic bacterial communities may

A freezing slide apparatus is described for visual observation of freezing water and melting ice within plant cells. The slide consists of an ordinary microscope slide glued into a Plexiglass jacket, through which cold 90% ethyl alcohol is pumped at varying rates for temperature control. Temperature is recorded by means of an iron-constantan thermocouple wire (25-micron diameter) connected to a recording potentiometer. Tissue strips were quick frozen (at a cooling rate of 33 C per ½ minute) and then warmed very slowly (at a rate of 2 C per minute) for observation of melting points. This apparatus has been used to determine osmotic pressures of cell sap of guard and adjoining epidermal cells of Chrysanthemum morifolium and Pelargonium hortorum. An accuracy of ± 1.2 atmospheres is possible. Wide variations among osmotic pressures of both guard and epidermal cells were found at any one stomatal aperture in both species. 相似文献

Evidence is accumulating in favor of a linkage at the cellular level between various abiotic stresses. We conducted a study to evaluate the effect of water stress on the heat tolerance of zonal geraniums, Pelargonium × hortorum cv. Evening Glow. Water stress was imposed by withholding irrigation until pots reached 30% (by weight) of well‐watered controls, and by maintaining the pot weight by additions of water for another 7 days. Leaf xylem water potential (XWP, MPa), relative water content (RWC. %), and heat‐stress tolerance (HST; LT50 , defined as the temperature causing half‐maximal % injury based on electrolyte leakage) were measured in control, stressed, and recovered plants. Proteins were extracted from the leaves following the above treatments, and SDS‐PAGE and immunoblotting were performed by using standard procedures. Immunoblots were probed with antibodies to dehydrin and 70‐kDa heat shock cognate (HSC70) proteins. Data indicate that XWP and RWC, respectively, were −0.378 MPa and 92.3% for control plants and −0.804 MPa and 78.6% for stressed plants. Water‐stressed plants exhibited a significant increase in HST compared to control (LT50 of 55°C vs 51°C). Water‐stress‐induced HST was not due to heat acclimation (leaf warming in stressed plants). Data also indicate that water‐stress treatment did not increase freezing tolerance of geranium leaves. Increased HST was associated with the accumulation of several heat‐stable, dehydrin proteins (25–60 kDa), and both cytosolic and ER luminal (BiP) HSC70 proteins. Leaf XWP, RWC, and HST reversed to control levels concomitant with the disappearance/reduction of dehydrins and HSC70 proteins in water‐stress‐relieved plants. The possibility of a cellular linkage between water stress and heat‐stress tolerance is discussed. 相似文献

Limestone and marble have been used extensively in the construction of modern and historic buildings. Microbial colonization and growth on these stone structures is common. Microbial deterioration of stone has been assessed by measuring Ca2+ released from the stone, using ion selective electrodes and titration with EDTA. In this study, the calcium binding fluorochrome Rhod-5N was used to measure Ca2+ released from limestone by endolithic bacteria as an indicator of biodeterioration. In a 17 d flask experiment, Ca2+ released by endolithic bacteria was twice that of uninoculated controls. Rhod-5N is a rapid and accurate method for measuring microbial biodeterioration of stone. 相似文献

Microtubule plus end dynamics are regulated by a conserved family of proteins called plus end–tracking proteins (+TIPs). It is unclear how various +TIPs interact with each other and with plus ends to control microtubule behavior. The centrosome-associated protein TACC3, a member of the transforming acidic coiled-coil (TACC) domain family, has been implicated in regulating several aspects of microtubule dynamics. However, TACC3 has not been shown to function as a +TIP in vertebrates. Here we show that TACC3 promotes axon outgrowth and regulates microtubule dynamics by increasing microtubule plus end velocities in vivo. We also demonstrate that TACC3 acts as a +TIP in multiple embryonic cell types and that this requires the conserved C-terminal TACC domain. Using high-resolution live-imaging data on tagged +TIPs, we show that TACC3 localizes to the extreme microtubule plus end, where it lies distal to the microtubule polymerization marker EB1 and directly overlaps with the microtubule polymerase XMAP215. TACC3 also plays a role in regulating XMAP215 stability and localizing XMAP215 to microtubule plus ends. Taken together, our results implicate TACC3 as a +TIP that functions with XMAP215 to regulate microtubule plus end dynamics. 相似文献
