Incubation experiments using filtered waters from Lake Kasumigaura were conducted to examine bacterial contribution to a dissolved organic carbon (DOC) pool. Bacterial abundance, bacterial production, concentrations of DOC, total dissolved amino acids (TDAA), and total dissolved neutral sugars (TDNS) were monitored during the experiments. Bacterial production during the first few days was very high (20 to 35 μg C liter−1 day−1), accounting for 40 to 70% of primary production. The total bacterial production accounted for 34 to 55% of the DOC loss during the experiment, indicating high bacterial activities in Lake Kasumigaura. The DOC degradation was only 12 to 15%, whereas the degradation of TDAA and TDNS ranged from 30 to 50%, suggesting the preferential usage of TDAA and TDNS. The contribution of bacterially derived carbon to a DOC pool in Lake Kasumigaura was estimated using d-amino acids as bacterial biomarkers and accounted for 30 to 50% of the lake DOC. These values were much higher than those estimated for the open ocean (20 to 30%). The ratio of bacterially derived carbon to bulk carbon increased slightly with time, suggesting that the bacterially derived carbon is more resistant to microbial degradation than bulk carbon. This is the first study to estimate the bacterial contribution to a DOC pool in freshwater environments. These results indicate that bacteria play even more important roles in carbon cycles in freshwater environments than in open oceans and also suggests that recent increases in recalcitrant DOC in various lakes could be attributed to bacterially derived carbon. The potential differences in bacterial contributions to dissolved organic matter (DOM) between freshwater and marine environments are discussed.  相似文献

Measurement of nitrogen isotopic composition (15N) of plants and soil nitrogen might allow the characteristics of N transformation in an ecosystem to be detected. We tested the measurement of 15N for its ability to provide a picture of N dynamics at the ecosystem level by doing a simple comparison of 15N between soil N pools and plants, and by using an existing model. 15N of plants and soil N was measured together with foliar nitrate reductase activity (NRA) and the foliar NO3 pool at two sites with different nitrification rates in a temperature forest in Japan. 15N of plants was similar to that of soil NO3 in the high-nitrification site. Because of high foliar NRA and the large foliar NO3 pool at this site, we concluded that plant 15N indicated a great reliance of plants on soil NO3 there. However, many 15N of soil N overlapped each other at the other site, and 15N could not provide definitive evidence of the N source. The existing model was verified by measured 15N of soil inorganic N and it explained the variations of plant 15N between the two sites in the context of relative importance of nitrification, but more information about isotopic fractionations during plant N uptake is required for quantitative discussions about the plant N source. The model applied here can provide a basis to compare 15N signatures from different ecosystems and to understand N dynamics.  相似文献

Carbon and nitrogen stable isotope ratios (δ13C and δ15N) of macroinvertebrates inhabiting littoral zones of lakes can serve as useful indicators of material loading from the watershed. We collected snails (Semisulcospira spp.) and bivalves (Unio douglasiae biwae Kobelt) from 29 littoral sites in Lake Biwa near the mouths of river tributaries with various human population density (HPD) and land-use patterns. The δ13C and δ15N signatures were determined for three potential food sources: particulate organic matter in the pelagic zone (PPOM), riverine particulate organic matter from tributaries (RPOM) and epilithic organic matter in the littoral zone (EOM). The stable isotope mixing model revealed that snails relied mainly on EOM, and bivalves on PPOM and RPOM. Multiple regression analysis showed that intersite variation in δ15N for snails was best explained by HPD, while variation in δ15N of EOM and nitrate was explained to a lesser extent by HPD. Comparison with isotope signatures of their food sources and riverine nutrients revealed that snails assimilated anthropogenic nitrogen from wastewater in the watershed. Our results also showed that the δ13C value of bivalves was marginally related to the fraction of paddy fields in the watersheds. In conclusion, the isotope signatures of macroinvertebrates inhabiting the littoral zone can be useful indicators of anthropogenic impacts from the watershed.  相似文献

The liverwort Marchantia polymorpha contains high proportions of arachidonic and eicosapentaenoic acids. In general, these C20 polyunsaturated fatty acids (PUFA) are synthesized from linoleic and alpha -linolenic acids, respectively, by a series of reactions catalyzed by Delta(6)-desaturase, an ELO-like enzyme involved in Delta(6) elongation and Delta(5)-desaturase. Here we report the isolation and characterization of the cDNAs, MpDES6, MpELO1 and MpDES5, coding for the respective enzymes from M. polymorpha. Co-expression of the MpDES6, MpELO1 and MpDES5 cDNAs resulted in the accumulation of arachidonic and eicosapentaenoic acids in the methylotrophic yeast Pichia pastoris. Interestingly, Delta(6) desaturation by the expression of the MpDES6 cDNA appears to occur both in glycerolipids and the acyl-CoA pool, although other lower-plant Delta(6)-desaturases are known to have a strong preference for glycerolipids.  相似文献

Trophic fractionation of carbon and nitrogen isotopes (Δδ13C, Δδ15N) was examined using previously complied databases for food chains in Lake Biwa, Lake Baikal, and Mongolian grassland. The following two features were clarified: (1) For each ecosystem, the ratios of trophic fractionation of carbon and nitrogen isotopes (Δδ15N/Δδ13C) throughout food chain could be obtained as the slope of linear regression line on the δ15N–δ13C plot. (2) Further, analysis of covariance (ANCOVA) revealed the slopes on δ15N–δ13C were not significantly different among these various ecosystems and allowed us to have the regression by setting δ15N as the response variable: δ15N = 1.61 δ13C + [ecosystem specific constant] with standard errors of [±0.41] and [±9.7] for the slope and the intercept, respectively. It was suggested that the slope of the regression (or the ratio Δδ15N/Δδ13C) could be applicable to more complicated food webs in case nitrogen and carbon isotope ratios of primary producers can be assumed constant in space and time within the ecosystems. The results from simple linear regression analyses coincided well with the ANCOVA results for these ecosystems, although there was some discrepancy between the results of the two statistical analyses. Possible factors that govern the linear relationship between δ15N and δ13C along a food chain are discussed together with a new scope for the stable isotope food chain analyses.  相似文献

Identifying sources of particulate organic matter (POM) is important for clarifying fundamental mechanisms by which lake food webs are sustained. We determined carbon and nitrogen stable isotope ratios of POM in near-shore waters of Lake Biwa, a large, meso-eutrophic lake in Japan, to estimate relative contributions of terrigenous particulate organic carbon (T-POC), plankton-derived POC (P-POC) and epilithon-derived POC (E-POC) to POC in near-shore waters. Samples were collected during different months (November, February, May and July) at 29 sites located near the mouth of tributary rivers with different discharge and catchment land use. The data revealed that POC mainly consisted of P-POC and T-POC, with relative contributions varying widely over season and among locations. E-POC generally contributed little to the near-shore POC. Path analyses revealed that the concentration of riverine POC whose isotopic signatures were similar to those of rice straws increased with a larger %paddy field area in the catchment of tributary rivers, which subsequently enhanced T-POC inputs to near-shore waters through riverine transportation. Furthermore, our results suggested that T-POC contribution was influenced, with a time lag, by wave-driven turbulence and shore topography, which appear to affect sedimentation and resuspension of T-POC.  相似文献

The stable nitrogen (N) isotope ratio (??15N) has been used to examine the anthropogenic N input (i.e., septic water, wastewater, and manure) to aquatic ecosystems, because anthropogenic N generally has a ??15N signature distinct from that found in nature. Aquatic organisms and the derived organic matter such as sediments are reported to become increasingly enriched in 15N as the human population density increases in watersheds. However, little is known about the relationship in steppe ecosystems, where the livestock population is greater than that of humans. Here, we conducted a preliminary study in the Selenga river mainstream watershed in Mongolia, which covers an area of approximately 300,000?km2. A multiple regression analysis revealed that the ??15N of the riverine sediment was significantly affected by the human population density and more significantly by livestock population density. The population density, including both humans and livestock, significantly influenced ??15N of the macrophytic Potamogeton spp. The results showed that ??15N of riverine organic matter can be an indicator of the human and livestock population density, which is likely associated with the status of N cycles in livestock-dominated watersheds.  相似文献
