Classification of freshwater fish in the subfamily Leuciscinae, Cyprinidae is hampered by complexity or lack of morphological diversity. In this study, analyses based on mtDNA sequences were undertaken to clarify phylogenetic relationships among Far Eastern, North American and European species in the Leuciscinae. Evolutionary rate in cytochrome b gene (Cyt-b) and D-loop sequences appear to be almost constant in Leuciscinae. The topology of phylogenetic trees generated by neighbor-joining (NJ) and maximum likelihood (ML) methods based on Cyt-b gene and D-loop sequences was similar. Five major clades, designated clades 1-5, and a minor clade were discriminated. Most of the Far Eastern, North American and European species were included in the major clades. Clade 1, comprised almost entirely of Far Eastern phoxinins, is monophyletic and greatly diverged from the other species of Leuciscinae. From the present phylogenetic relationships and the previous studies, we present the following hypotheses with respect to the evolutionary history of the Far Eastern phoxinins. The Far Eastern species should be classified into Far Eastern-specific genera, although ichthyologists have still insisted that the species should be included in the European genera. The Far Eastern clade 1 consists of two subclades, including genera Pseudaspius-Tribolodon and Far Eastern Phoxinus species. According to our phylogenetic analyses, Pseudaspius leptocephalus and Tribolodon species should be reclassified into the same genus. On the basis of evolutionary rate in Cyt-b gene in Cyprinidae, it is estimated that the Far Eastern lineage diverged approximately 10-14 million years ago (mya) from the common ancestor of Leuciscinae. It is deduced that speciation of the Far Eastern species occurred until approximately 4 mya, in relation to the formation of the Sea of Japan and the Japanese Islands.  相似文献

Genetic divergence estimates using p-distances and similar measures were generated for 20,731 vertebrate and invertebrate animal species. The results of this analysis demonstrate that the data series are realistic and interpretable when the p-distance and its various derivates are used. The focus is on vertebrates and fish species in particular and the newest data set. Distance data reveal increasing levels of genetic divergence of the sequences of the two genes, cytochrome b (Cyt-b) and cytochrome c oxidase subunit 1 (Co-1), in the five groups compared: populations within species; subspecies, semi-species, or/and sibling species; species within a genus; species from different genera within a family; and species from separate families within an order. Mean unweighted scores of p-distances (%) for these five groups are Cyt-b-1.38?±?0.30, 5.10?±?0.91, 10.31?±?0.93, 17.86?±?1.36, and 26.36?±?3.88, respectively; and Co-1-0.89?±?0.16, 3.78?±?1.18, 11.06?±?0.53, 16.60?±?0.69, and 20.57?±?0.40, respectively. The estimates show good correspondence with other analyses. These results testify to the applicability of p-distance for most intra-species and inter-species comparisons of genetic divergence up to the order level in animals for the two genes compared. Data reviewed provide empirical and theoretical background on the geographic speciation mode prevalence in species origin and give a framework why per-individual species identification (DNA barcoding) is usually successful.  相似文献

On the basis of nucleotide sequences of three nuclear genes and using molecular phylogenetic and evolutionary genetic approaches, the phylogeny of the main representatives of one of the largest taxa of bivalve mollusks, the family Mytilidae, was studied, and its system and taxonomy were refined. A phylogenetic system for the family Mytilidae and closely relative taxa of the order Mytilida, which currently has no consensus among specialists on the basis of traditional characters, is presented. Using nucleotide sequences of the 28S rRNA, 18S pRNA, and histone H3 genes, this consensus was established by the study of Mytilidae. Some concerns of mussel systematics were resolved; in particular, the monophyly of the family Mytilidae Rafinesque, 1815 was established with the strongest support for the subfamily Mytilinae Rafinesque, 1815. The data obtained disprove Distel’s conclusion on polyphyly of the subfamily Mytilinae Rafinesque, 1815. Isolation of the taxa in the rank of the Modiolinae G. Termier & H. Termier, 1950 and Bathymodiolinae Kenk & Wilson, 1985 subfamilies in the family Mytilidae and also the family Septiferidae Scarlato et Starobogatov, 1979 was confirmed, although the rank of the later taxon is not universally recognized and it remains to be clarified in an additional study.  相似文献

We isolated Korean soft-shelled turtle, Pelodiscus sinensis, mitochondrial DNA by long-polymerase chain reaction (long-PCR) with conserved primers and sequenced this mitochondrial genome (mitogenome) with primer walking using flanking sequences. The P. sinensis mitochondrial DNA has 17,042 bp and its structural organization is conserved compared to those of other reptiles and mammals. To unveil the phylogenetic relationship of the turtles, we used the NJ, MP, and ML analysis methods after inferring those sequences from the mitochondrial 16S rRNA gene. We also compared two P. sinensis variants from Korea and China using the mitochondrial genome. In this study, we report the basic characteristics of the P. sinensis mitochondrial genome, including structural organization and base composition of the rRNAs, tRNAs and protein-coding genes, as well as characteristics of tRNAs. These features are applicable for the study of phylogenetic relationships in turtles.  相似文献

In the Pacific herring, Clupea pallasii, eight samples of spawning, mature fish (collected in 1978 and 1981) and six samples of immature fish individuals (collected in 1979, 1980 and 1984) were analyzed. Mature fish were analyzed by 24 conventional morphometric characters or morphometric indices. By means of discriminant and factor analyses (principal component mode) the existence of differentiating groupings of vector scores was found that have obvious biological meaning as local Pacific herring stocks. In concordance with former morphobiological and genetic data, current multivariate observations supported the existence in the surveyed area of Sakhalin waters of at least three stocks. One of them is the Sakhalin-Hokkaido local stock of Pacific herring, which in the past provided the bulk of herring catches in the region and showed a distinct and sharper morphometric differentiation as compared with other herring assemblages.  相似文献

This paper examines the genetic variability of the Pacific mussel Mytilus trossulus and an introduced Atlantic species, M. galloprovincialis, in the northwestern Sea of Japan (Peter the Great Bay and Kievka Bay). The genotyping of individuals from eight populations was carried out using eight polymorphic enzyme loci and two nuclear DNA markers (Me-5 and ITS-1,2); the occurrence frequency of parent species and their hybrids was determined. The enzyme and nuclear markers demonstrated concordant genetic variation. The genotypes of the native species M. trossulus were predominant in the samples studied. The frequency of the introduced species M. galloprovincialis in the total material was relatively low; however, it reached 42 ± 2% in samples that were collected in Possjet Bay near the town of Zarubino in a zone of active international navigation. In this area the greatest number of hybrids was found as well. It is concluded that the invasion of M. galloprovincialis in the northwestern Sea of Japan is continuing; permanent populations of this mussel appeared in Possjet Bay that were not recorded here previously.  相似文献

A survey was made of 168 specimens of the gastropod mollusk Nucella heyseanafrom 5 samples collected in Amurskii Bay and Vostok Bay (Peter the Great Bay and the Sea of Japan). The variability of Cu, Zn, Cd, Pb, Fe, and Mn contents in gastropod soft tissues was analyzed. The statistic concentrations of the six metals differed significantly among the samples; they were sometimes taken at a distance of a mere several tens of meters apart. N. heyseanacollected near Vladivostok generally had higher concentrations of heavy metals; however, the amount of Zn in the specimens of one sample from the Vostok Bay Reserve was 400 g/g, which is two times greater than that of mollusks from the urbanized zone. The canonical and discriminant analyses and the analysis of the variance indicate that the variation in heavy metal concentration within groups of mollusks (within bays) is comparable in magnitude with the variation among the groups.  相似文献

Nucleotide diversity estimates for the genes Cyt-b (cytochrome b) and Co-1 (cytochrome oxidase 1) are analyzed. Genetic divergence of populations (1) and taxa of different rank, such as subspecies, semispecies or/and sibling species (2), species within a genus (3), species from different genera within a family (4), and species from separate families within an order (5) have been compared using a database of p-distances and similar measures. Empirical data for 20,731 vertebrate and invertebrate animal species reveal various and increasing levels of genetic divergence of the sequences of the two genes, Cyt-b and Co-1, in the five groups compared. Mean unweighted scores of p-distances (%) for five groups are: Cyt-b (1) 1.38±0.30, (2) 5.10±0.91, (3) 10.31±0.93, (4) 17.86±1.36, (5) 26.36±3.88 and Co-1 (1) 0.89±0.16, (2) 3.78±1.18, (3) 11.06±0.53, (4) 16.60±0.69, (5) 20.57±0.40. These estimates testify to the applicability of p-distance for most intraspecies and interspecies comparisons of genetic divergence up to the order level for the two genes compared. The results of the analysis of the nucleotide divergence within species and higher taxa of animals suggest that a phyletic evolution in animals is likely to prevail at the molecular level, and speciation mainly corresponds to the geographic or divergence mode (type D1). The prevalence of the D1 speciation mode does not mean that other modes are absent. At least seven possible modes of speciation are considered. The approach suggested that allows recognize the speciation modes formally with the operational genetic criteria. Such approach may help to solve a key problem of the biological species concept, i.e. the lack of ability to monitor in most cases the reproductive isolation barriers between species.  相似文献

Molecular genetic analysis of Altai osmans of the genus Oreoleuciscus from two different parts of the range was carried out. In this study, based on the mitochondrial Co-1 gene sequence, a total of 25 fish specimens belonging to four genera were examined: (1) O. humilis, 2 specimens; O. potanini, 13 specimens; (2) Pseudaspius leptocephalus, 1 specimen; (3) Tribolodon brandtii, T. hakonensis, and T. sachalinensis from the GenBank database, 8 speciens; and (4) Leuciscus waleckii, 1 specimen (used as an outgroup). The p-distances were very low both within and between the species: (1) 0.20 +/- 0.03%; (2) 0.40 +/- 0.12%; and (1-2) 0.80 +/- 0.04%. To visualize the relationships among all of the species examined, the neighbor joining (NJ), maximum parsimony (MP), Bayesian (BA), and maximum likelihood (ML) trees were constructed. The results obtained using these methods were very similar. It was demonstrated that species assignment of the individuals (barcoding) with the help Co-1 gene was effective, despite of very low divergence of the two osman taxa, which was comparable with typical intraspecific values in other animal groups. Taxonomic status of O. potanini and O. humilis requires further investigation with paying attention to low genetic distances between these species along with the lack of material from sympatric parts of the ranges.  相似文献
